Ga-67 scan in patients with intrathoracic esophageal carcinoma planned for surgery.

The authors evaluated Ga-67 scan in 68 patients with intrathoracic esophageal carcinoma initially planned for surgery. Of these, 59 patients were staged pathologically or surgically; their Ga-67 scan results correctly predicted the presence of extraesophageal spread and lymph node metastases. Of 38 clinical Stage II patients, 15 (39%) could be Stage III by the results of Ga-67 scan. However, two of four clinical Stage I patients (50%) and 35 of 38 clinical Stage II patients (92%) were eventually pathologic or surgical Stage III. This high conversion rate and the high incidence of Stage III in esophageal carcinoma patients may not justify routine use of Ga-67 scan only for staging. Ga-67 scan, however, was useful for planning radiotherapy, because missing extraesophageal extension or gross metastases from the radiation fields were detected in six patients of 51 so treated. Ga-67 scan has its value in some patients with intrathoracic esophageal carcinoma.

The 38,000-Mr poly(A)-binding protein of non-polysomal messenger ribonucleoproteins of cryptobiotic gastrulae of Artemia salina.

The 38,000-Mr poly(A)-binding protein has been purified to near homogeneity from non-polysomal messenger ribonucleoprotein of Artemia salina [Slegers, H., De Herdt, E., and Kondo, M. (1981) Eur. J. Biochem. 117, 111-120]. The protein consists of approximately 357 amino acids and is characterized by a high glycine content of 22.5% and the presence of dimethylarginine. From polynucleotide-protein binding experiments a stoichiometry of 9-11 adenylate and 10-12 uridylate residues per protein molecule is calculated. The polypeptide is devoid of poly(A) polymerase and RNase activities. The poly(A)-binding protein and the helix-destabilizing protein HD40 [Marvil, D. K., Nowak, L., and Szer, W. (1980) J. Biol. Chem. 255, 6466-6472] have the same mobility in polyacrylamide/dodecylsulphate gel electrophoresis and exhibit a comparable amino acid composition and protein-polynucleotide stoichiometry. Based on the length of poly(A) sequences of mRNA and from protein-poly(A) binding experiments, a repetitive binding of the 38,000-Mr protein on the poly(A) sequence is demonstrated. The 38,000-Mr protein of cytoplasmic and membrane-bound non-polysomal messenger ribonucleoproteins is also compared.

Thiobarbituric acid reactive substances and vitamin E in serum and cerebrospinal fluid from patients with cerebral apoplexy.

Thiobarbituric acid (TBA) reactive substances in serum and cerebrospinal fluid from patients with cerebral apoplexy were determined and their relationship to this disease was studied. TBA reactants in serum were elevated in patients with cerebral apoplexy, but there was no difference between patients with brain tumor or other neurological disease and healthy persons. TBA reactants in cerebrospinal fluid in patients with cerebral aneurysm, cerebral arterial and venous anomalies or intracerebral hematoma, and its levels in patients with cerebral infarction or brain tumor were also higher than in healthy persons. Serum alpha-tocopherol levels were elevated in patients with cerebral infarction, but were relatively low in patients with cerebral aneurysm, cerebral arterial and venous anomalies or intracerebral hematoma. The reverse was the case with cerebrospinal fluid levels of alpha-tocopherol. That is, the sequence of decreasing level of alpha-tocopherol in cerebrospinal fluid was intracerebral hematoma, cerebral aneurysm, cerebral arterial and venous anomalies, and cerebral infarction. The levels of TBA reactive substances in patients with cerebral apoplexy showed a positive correlation between serum and cerebrospinal fluid, and the levels of alpha-tocopherol showed a similar correlation.

Effects of thiamin on vitamin B6 synthesis in yeasts.

The effects of exogenous thiamin on the growth yield and vitamin B6 content of 18 strains of yeasts and a few strains of bacteria were examined. The addition of thiamin hardly affected the growth yield of the yeasts tested, except for two strains Saccharomyces uvarum strain 4228 and Saccharomyces uvarum IFO 0751. In contrast, the vitamin B6 content of all the yeasts tested, except Pichia membranaefaciens IFO 0189, decreased markedly in the presence of thiamin. In S. uvarum IFO 1265, the synthesis of vitamin B6 was maximally inhibited by the addition of thiamin (1.5 nmol ml-1) to the growth medium without affecting cell growth, whereas the amounts of cellular vitamin B6 increased in the presence of the thiamin antagonist, pyrithiamin or oxythiamin, at concentrations that did not affect growth. When [4'-14C]pyridoxine. HCl (0.5 micrograms ml-1) was added to the growth medium at least 54% of the added isotope was incorporated into the cells during 24 h incubation. In the presence of thiamin (15 nmol ml-1), at least 32% of the added isotope was incorporated. The metabolism of [4'-14C]pyridoxine. HCl to inactive forms having no vitamin B6 activity was not stimulated by the addition of thiamin. Thus, vitamin B6 synthesis in many yeasts was affected by thiamin, whereas, in bacteria, growth yield and vitamin B6 content were not affected by thiamin.

Properties of Hg- and Cd-spores of Bacillus megaterium QM B1551.

Hg- and Cd-spores of Bacillus megaterium QM B1551 were produced in Schaeffer's medium containing mercuric chloride and cadmium chloride respectively. Metals were added to the medium at 9 hr of incubation (Stage V) to give a final concentration of 50 microM. It was found by electron microscopic and biochemical studies that the coats of both Hg- and Cd-spores were thinner than those of control spores. Of the total Hg and Cd in the spores, 77% of the Hg and 63% of the Cd were detected in the spore coats. Hg- and Cd-spores were less resistant to heat and more sensitive to germinants than control spores. Other properties of Hg- and Cd-spores were similar to those of control spores. These results suggest that the spore coat has some relationship to the heat resistance and germinability of spores.

Studies on contact hypersensitivity in the guinea pig.

A method to determine the quantitative induction and challenge of the allergenicity of externally applied toiletories and cosmetics, including their components, is described. The experiment used oil-soluble cinnamic aldehyde and water-soluble formalin as allergens, and guinea pigs as the experimental animals. A high sensitization method resulted, carried out as follows. A 24-h closed patch is attached to the skin every other day over a period of 2 weeks (a total of 4 applications). Freund's complete adjuvant is administered intradermally just before the 3rd application of the patch. The challenge step is performed by directly applying the test material. This method was compared with other allergenicity evaluation methods. As a result, this method was found to be in no way inferior in sensitization performance to the other methods. The method was used on perfume mixtures and tested for its evaluation effectiveness. It proved to be satisfactory.

Postoperative thallium-201 myocardial images. Evidence of regression of right ventricular hypertrophy in man.

Thallium-201 myocardial scintigraphic studies were performed on 24 patients with chronic right ventricular overload before and after surgical correction of haemodynamic overload. The ages of the patients ranged from 20 to 65 years (mean 39 years) at operation. The degree of right ventricular visualisation remained essentially unchanged in an early postoperative study (four to 60 days), though a decrease in right ventricular cavity size was noted in patients with right ventricular volume overload. On later follow-up (18 to 36 months), thallium uptake in the right ventricle was definitely less than before operation in all 13 patients studied at this time. Because thallium-201 radioactivity reflects myocardial blood flow and mass, our study indicates that right ventricular hypertrophy is largely reversible. Thus, thallium-201 myocardial scintigraphy can be used to assess non-invasively regression of right ventricular hypertrophy in patients with right ventricular overload.

Effects of vitamin E on D-galactosamine-induced or carbon tetrachloride-induced hepatotoxicity.

Experimental liver disorders were induced by the use of carbon tetrachloride or D-galactosamine hydrochloride in rats maintained on a vitamin E deficient diet and in rats fed a diet supplemented with vitamin E, and the protective effect of vitamin E on the liver was determined. After exposure to carbon tetrachloride or D-galactosamine hydrochloride the serum levels of transaminases, lysosomal enzyme beta-glucuronidase, and acid phosphatase were elevated, and thiobarbituric acid reactive substances in serum and liver homogenate were also increased. The changes were conspicuous in the vitamin E deficient rats, but were only slight in rats fed a diet supplemented with vitamin E. The results of this study suggest that vitamin E has a protective effect on liver disorders by inhibiting lysosomal enzyme liberation and lipid peroxidation.

Effects of barbiturates on the decerebrate rigidity in rats.

Effects of barbiturates (phenobarbital, mephobarbital, pentobarbital, hexobarbital and amobarbital) on the tonic and phasic responses of forelimbs were studied in the rats in which rigidity was induced by means of anemic decerebration. The inhibitory pattern of the tonic and phasic responses to phenobarbital was different from those to the other barbiturates, i.e. phenobarbital gradually inhibited both tonic and phasic responses, whereas other barbiturates suppressed significantly only the tonic response immediately after the administration of a relatively low dose (10 mg/kg) but hardly inhibited the phasic response unless at higher doses (20 and 40 mg/kg). Possible involvement of GABA-system in the inhibitory action of phenobarbital and pentobarbital on the rigidity was investigated. Since inhibition of the phasic response to pentobarbital was potentiated by the pretreatment of animals with carboxymethoxylamine hemihydrochloride (AOAA) and suppressed by the pretreatment with semicarbazide, it was suggested that GABAergic mechanism may be involved in the particular inhibition of phasic response to pentobarbital. However, the inhibitory action of phenobarbital appeared to be independent of GABA-system.

Radiation therapy of early glottic carcinoma. A Japanese experience.

From 1969 and through 1979, 105 patients with T1 N0M0 and T2 N0M0 glottic carcinoma were treated with irradiation alone. The minimum follow-up period was 2 years, and only one patient was lost to follow-up. There were 62 T1a lesions, 14 T1b, 19 T2 with normal mobility, and 10 T2 with impaired mobility; relapses occurred in 10, 4, 1 and 3 patients, respectively. All but one patient who relapsed had local recurrence with or without nodal or distant spread. One patient had only nodal metastasis. Six of the 18 relapses were successfully cured by surgery. None of the patients who had nodal or distant spread survived. The relapse rates had no clear relation to total dose or TDF value. T1 lesions treated through larger fields (greater than or equal to 25 cm2) had a lower relapse rate than lesions treated through smaller fields, but the difference was not statistically significant.
