[A complex program for microcomputer-supported immunoassay analysis].

Abstract

The increasing availability of analytical methods based on saturation analysis call for efficient assay calculation. A comprehensive computer program was developed, allowing the calculation of different kinds of assays. Due to the user friendly structure it can be used also by inexperienced laboratory staff without difficulties. Its large adaptability can be a usefull support to optimize various kinds of assays.

Advances in Single-Molecule Immunoassay: From Counting Strategies to CRISPR-Enhanced Biosensing.

Single-molecule immunoassays (SMIs) overcome the sensitivity limitations of conventional bulk measurements by enabling a paradigm shift from analog to digital signal readouts, thereby facilitating highly sensitive quantification of ultra-low-abundance biomarkers for precision diagnostics. This review provides a systematic overview of recent advances in SMI technologies and the conceptual framework underlying their evolution. First, discretization strategies for single-molecule counting are classified into hard discretization, based on physical confinement, and soft discretization, based on spatiotemporal isolation, within heterogeneous and homogeneous assay systems, respectively. The fundamental mechanisms by which these strategies mitigate diffusion limitations and enhance signal-to-noise ratios are discussed. Second, the integration of SMIs with CRISPR-based diagnostic systems (CRISPR-dx) is examined, with particular emphasis on their complementary roles in target recognition and signal amplification. Finally, recent applications of SMIs in the diagnosis of oncological, neurological, infectious, and cardiovascular diseases are summarized, along with a critical discussion of current engineering challenges and future directions toward clinical translation.

Microplate washing: process description and improvements.

Heterogeneous immunoassays require wash steps in order to separate bound from free constituents. In this paper we demonstrate that in microplate assays the washing process includes two separate physical processes: (1) a rapid and wash volume-dependent direct dilution of the droplet-shaped residual volume, and (2) a diffusion-limited and strongly time-dependent dilution of a residual layer of liquid, which necessitates the use of time-consuming soak times in the immunoassay. We have shown that optimizing the motion of the wash fluid effectively reduces the residual layer thickness that results in extended soak times. This results not only in improved washing efficiency and reduced background variance in the immunoassay, it also yields a significantly improved immunoassay sensitivity.

Development of a rapid latex enhanced turbidimetric assay for retinol binding protein in urine.

We describe a simple, rapid and sensitive homogeneous immunoassay for urinary retinol-binding protein (RBP) using latex particle-enhanced turbidimetric immunoassay. Rabbit anti-human RBP is covalently coupled to 40 nm latex particles and the assay performed on the IL Monarch 2000 centrifugal analyser, with a 20 microL sample volume and the reaction monitored at 340 nm over an 8 min period. The assay range is 0-6 mg/L with a detection limit of 25 micrograms/L. The within and between assay coefficients of variation are less than 1.5% and less than 2.5%, respectively. Comparison with radioimmunoassay for RBP showed good agreement.
