Determination of aceclofenac in bulk and pharmaceutical formulations.

Three sensitive and reproducible methods for quantitative determination of aceclofenac (AC) in pure form and in pharmaceutical formulation are presented. The first method is based on the reaction between the drug via its secondary aromatic amino group and p-dimethylaminocinnamaldehyde (PDAC) in acidified methanol to give a stable coloured complex after heating at 75 degrees C for 20 min. Absorption measurements were carried out at 665.5 nm. Beer's law is obeyed over concentration range 20-100 microg ml(-1) with mean recovery 100.33 +/- 0.84. The other two methods are high performance liquid chromatography (HPLC) and densitometric methods by which the drug was determined in the presence of its degradation products over concentration range of 20-70 microg ml(-1) and 1-10 microg per spot and mean recoveries are 99.59 +/- 0.90 and 99.45 +/- 1.09, respectively.

Chloro(2,9-dimethyl-1,10-phenanthroline-N,N')(isoquinoline-1-carboxylato-O,N)copper(II).

The asymmetric unit of the title compound, [CuCl(C(10)H(6)NO(2))(C(14)H(12)N(2))], contains two monomeric copper molecules, A and B. Each Cu atom is coordinated to one 2,9-dimethyl-1,10-phenanthroline (neocuproine) ligand via both N atoms, to one isoquinoline-1-carboxylate anion (IQC(-)) via the N and one O atom, and to one Cl(-) anion. The environment of the Cu atom is approximately square pyramidal, with the apical position occupied by an N atom of neocuproine. In molecule A, the Cu atom is 0.301 (1) A above the basal plane; this distance is 0.316 (1) A in molecule B. The crystal packing is characterized by several hydrogen bonds.

Genetic analysis as a valuable key to diagnosis and treatment of periodic Fever.

We describe 2 Dutch patients with recurrent fever attacks undiagnosed for more than 40 years. The diagnosis of periodic fever was made when molecular analysis revealed novel mutations in the tumor necrosis factor (TNF) receptor gene (TNFRSF1A), establishing the diagnosis of TNF receptor-associated periodic syndrome. This syndrome is an autosomal dominant disorder characterized by recurring episodes of fever, arthralgia, and skin lesions that is caused by mutations in the 55-kd TNFRSF1A gene. This finding has facilitated treatment for TNF receptor-associated periodic syndrome because blocking of TNF signaling seems to alleviate the symptoms. Use of a short course of recombinant p75TNFR:Fc fusion protein (etanercept) induced prolonged remission in one patient.
