Effect of photoperiod on pineal gland volume and pinealocyte size in the Chinese hamster, Cricetulus griseus.

Male adult (200-day-old) Chinese hamsters (Cricetulus griseus) raised from weaning under either LD 16:8 or LD 8:16 were used. The pineal gland of the Chinese hamster consists of superficial (major) and deep (minor) components and a continuous, or interrupted, narrow parenchymal stalk interposed between them. The volume of the superficial pineal including the parenchymal stalk is greater under LD 16:8 than under LD 8:16. Under both photoperiods, pinealocytes in the superficial pineal have larger nuclei and more abundant cytoplasm than those in the deep pineal. Nuclei in the superficial pineal appear pale and usually have irregular profiles, whereas those in the deep pineal appear dark and have round profiles. In the superficial pineal, pinealocyte nuclei are larger, paler, and more irregular; and, in addition, nuclear density is lower under LD 16:8 than under LD 8:16. Similar, but less prominent, photoperiod-induced changes occur in the volume of the deep pineal, the size of pinealocytes, and pinealocyte nuclear morphology in the deep pineal. The results indicate that the development and differentiation of pinealocytes in both pineal portions may be advanced under long photoperiods and delayed under short photoperiods, although pinealocytes in the deep pineal may remain not fully differentiated even in adults. Since testicular weights and body weights are similar under both photoperiods, the photoperiod may exert marked influences on the development of the pineal gland without affecting reproductive activity and growth rates of animals.

Prevention of experimental acute pancreatitis by intraduodenal trypsin inhibitor in rat.

To confirm that trypsin activity is a most important initiating factor in closed duodenal loop pancreatitis in rats, we observed the course of acute pancreatitis when trypsinogen activation was inhibited by intraduodenal infusion of a potent synthetic trypsin inhibitor (TI, nafamostat mesilate) but the other conditions were left unchanged. Intraduodenal and intrapancreatic trypsinogen activation was inhibited for 16 hr after the intraduodenal infusion of the inhibitor, although elevation of serum amylase and immunoreactive trypsin and pancreatic trypsinogen remained similar both in the TI and control groups. The mortality decreased from 44% (control) to 4% (TI) at 48 hr after establishing the model. Active trypsin in duodenal reflux is an initiating factor for further development of acute pancreatitis in the closed loop model, and inhibition of the initial activation of trypsinogen has a favorable effect on acute pancreatitis even if other deleterious factors remain unchanged.

Mapping of the Hox-3.1 and Myc-1.2 genes on chromosome 15 of the mouse by restriction fragment length variations.

Restriction endonuclease fragment length variations (RFLV) were detected by use of the cDNA probe Hox-3.1 for the homeo box-3.1 gene and also the c-myc oncogene probe for exon 2. RFLV of Hox-3.1 were found in HindIII restriction patterns, and RFLV of the Myc-1.2 gene in EcoRV patterns. From the RFLV, the Hox-3.1 and Myc-1.2 genes were mapped on chromosome 15. Three-point cross test data showed that the frequency of recombination is 26.4% between Myc-1.2 and Gpt-1, 30.2% between Gpt-1 and Gdc-1, and 9.4% between Gdc-1 and Hox-3.1. The following order of these genes is proposed, Myc-1.2--Gpt-1--Gdc-1--Hox-3.1. All laboratory strains carry the Hox-3.1a and Myc-1.2a alleles. Among strains of wild origin, domesticus strains carry only the Hox-3.1a and Myc-1.2a alleles, as do the laboratory strains. One strain of brevirostris carries the Hox-3.1a and Myc-1.2b alleles. Other wild subspecies from Europe and Asia, M. m. musculus, M. m. castaneus, M. m. molossinus, Chinese mice of wild origin, and M. m. yamashinai carry the Hox-3.1b and Myc-1.2b alleles.

Inhibitory action of peony root extract on the mutagenicity of benzo[a]pyrene.

The inhibitory effects of peony root extract on the mutagenicity of benzo[a]pyrene (B [a]p) have been investigated in the Salmonella typhimurium reversion test. Four kinds of experiments were performed: direct chemical reaction (1) between peony root extract and B [a]p, and (2) between peony root extract and active metabolite(s) of B [a]p, (3) inhibition of metabolic processes of B[a]p with S9 mix, and (4) inhibition of activation on mutagenicity. Peony root extract interfered with the action of enzymes in the S9 mix, and inactivated the activity of B[a]p metabolites. The bio-antimutagenic effect was assayed by reversion in Salmonella typhimurium TA98 and TA100.

GABAB receptors expressed in Xenopus oocytes by guinea pig cerebral mRNA are functionally coupled with Ca2(+)-dependent Cl- channels and with K+ channels, through GTP-binding proteins.

Transmembrane currents induced by (-)-baclofen (BAC), a specific agonist of the gamma-aminobutyric acid-B (GABAB) receptor, in Xenopus oocytes injected with guinea pig cerebral mRNA were electrophysiologically and pharmacologically characterized under a voltage-clamp condition. The oocytes injected with mRNA acquired responsiveness to BAC and showed two types of currents at a holding potential of -50 mV. One was the slow and smooth inward current which had a short latency and associated with a decrease in membrane conductance, and its amplitude was decreased by hyperpolarization and increased by depolarization. The other was the large fast oscillatory inward current with a long-latency, which was decreased in amplitude by depolarization and reversed at -26 mV. Both currents were not blocked by bicuculline but were depressed by 2-hydroxysaclofen (2-OH-SAC), though the smooth current was less sensitive to 2-OH-SAC; about 40% blockade at the 2-OH-SAC concentration capable of abolishing the oscillatory current. The smooth current was depressed by Ba2+. The oscillatory current was time-dependently attenuated and almost abolished by intracellularly injected pertussis toxin (PTX), while the smooth current was not depressed by this toxin even when the oscillatory current was nearly abolished. The intracellular injection of GTP-gamma-S into oocytes attenuated both oscillatory and smooth currents. These results suggest the possibility that GABAB receptors expressed in Xenopus oocytes by cerebral mRNA are functionally coupled with two signal transduction systems, one is the opening of Ca2(+)-dependent Cl- channels mediated by PTX-sensitive GTP-binding protein(s) and the other is the closure of K+ channels through PTX-insensitive GTP-binding protein(s).

Dopamine receptors expressed in the Xenopus oocytes injected with bovine striatal mRNA.

The response to dopamine (DA) of Xenopus oocytes injected with bovine striatal mRNA was electrophysiologically and pharmacologically investigated under a voltage-clamped condition. In oocytes injected with bovine striatal mRNA and then treated with collagenase (denuded oocytes), DA applied by superfusion dose-dependently induced oscillatory inward currents with a long latency and an ED50 value of 12.0 microM. mRNA non-injected denuded oocytes did not respond to DA up to concentrations of 1 mM. The reversal potential of DA-induced currents was about -25 mV, indicating that DA increased a Cl(-)-conductance. DA-induced currents were easily desensitized by repeated applications of DA. Haloperidol and SCH-23390 abolished DA-induced currents, while propranolol and mianserin showed no effect. Furthermore, SK&F-38393, a specific agonist of the DA D1 receptor, induced similar oscillatory currents as DA did. These results suggest the possibility that functional D1 and D2 DA receptors were co-expressed in Xenopus oocytes by injection of bovine striatal mRNA.

Alcohol consumption and blood pressure: a comparison of native Japanese to American men.

We compared the cross-sectional association of alcohol consumption with blood pressure in 810 Japanese men (JM) living in Tokyo and 946 white men (WM) living in New York. Mean systolic (JM and WM, p less than 0.001) and diastolic blood pressure (JM, p less than 0.002; WM, p less than 0.001) were associated with alcohol consumption in both groups. Compared to abstainers, the heaviest drinkers had the highest systolic (JM, p = 0.001; WM, p less than 0.01) and diastolic (JM, p less than 0.002; WM, p less than 0.05) blood pressures. The relation of blood pressure to alcohol intake was J-shaped in the Americans, but linear in the Japanese. Exploratory analyses revealed that the J-shape may have been due to under-reporting of heavy alcohol ingestion by American abstainers. When abstainers were excluded, the relationships were similar in both the American and Japanese. The positive association between blood pressure and alcohol consumption persisted after adjustment for age, cigarette smoking, use of antihypertensive medications, body mass index, heart rate, abdominal skinfold thickness, hematocrit, fasting blood glucose, serum uric acid levels and urinary sodium/potassium ratio. Alcohol use was also related to prevalence of hypertension. These findings confirm the presence of an independent association between alcohol intake and blood pressure in both JM and WM and suggest that, despite differences in the metabolism of alcohol, the relation of alcohol consumption to blood pressure is similar in both nationalities.

Serum and transforming growth factor beta regulate glial fibrillary acidic protein in serum-free-derived mouse embryo cells.

Serum-free mouse embryo (SFME) cells, derived in medium in which serum is replaced with growth factors and other supplements, display distinctive properties: (i) SFME cells do not lose proliferative potential or show gross chromosomal aberration upon extended culture, (ii) these cells depend on epidermal growth factor for survival; and (iii) SFME cell proliferation is reversibly inhibited by serum. Treatment of SFME cells with serum or transforming growth factor beta led to the appearance of glial fibrillary acidic protein, a specific marker for astrocytes. The appearance of glial fibrillary acidic protein in cultures was reversed upon removal of transforming growth factor beta or serum. Cells with properties similar to SFME cells were also isolated from adult mouse brain. These results suggest a role for transforming growth factor beta in astrocyte differentiation in developing organisms and in response to injury and identify the cell type that has the unusual properties of SFME cells.

Actin with tumor-related mutation is antimorphic in Drosophila muscle: two distinct modes of myofibrillar disruption by antimorphic actins.

A mutant beta-actin with an amino acid substitution from Gly-245 to Asp has been shown to be related to tumorigenic transformation of a human fibroblast cell line (Leavitt, J. et al. (1987) Mol. Cell. Biol. 7, 2467-2476). To examine the effects of this mutation, we artificially introduced the same amino acid change into the Act88F actin gene of Drosophila melanogaster. The gene (Act88FGD245) was inserted in the Drosophila genome to make transgenic adult flies which synthesize the mutant actin in the indirect flight muscles. The mutant actin was found to be antimorphic with regard to flight and also to cause myofibrillar disruption in transformants even in the presence of two normal alleles. It was initially incorporated into myofibrils and later induced their degeneration from center to periphery. This mode of myofibrillar disruption is distinct from that of previously reported Act88F mutations, where defects are found only in the peripheral region of myofibrils. This indicates that actin functions are altered differently in the two classes of antimorphic mutations.

A newly designed deodorant pad for urinary incontinence.

The authors designed a new pad for urinary incontinence. It is composed of a cloth of acrylonitrile copper sulfide and another cloth of iron-phthalocyanine. In vitro experiments showed that acrylonitrile copper sulfide cloth inhibited the growth of most bacteria causing urinary tract infection, and that iron-phthalocyanine cloth effectively eliminated bad-smelling gases. The use of this pad relieved diaper-dermatitis and diminished offensive smells from feces and urine. This pad was well received by nurses and helpers taking care of incontinent elderly people.

Stimulatory effects of bombesin on plasma trypsin release and exocrine pancreatic secretion in dogs.

We examined the effect of bombesin on plasma trypsin release and exocrine pancreatic secretion in dogs. Bombesin significantly increased plasma immunoreactive trypsin (IRT). Atropine significantly inhibited the response of plasma IRT to bombesin. Pancreatic trypsin secretion was also increased by bombesin, as well as bicarbonate and protein outputs. Atropine failed to inhibit pancreatic trypsin secretion. In conclusion, bombesin has a stimulatory effect on plasma trypsin release mediated by a cholinergic mechanism and different from pancreatic secretion.

An early phase II study of CPT-11: a new derivative of camptothecin, for the treatment of leukemia and lymphoma.

An early phase II study of a new camptothecin analog and an inhibitor of topoisomerase I, CPT-11, was conducted in 62 patients with refractory leukemia and lymphoma by four different treatment schedules in a multiinstitutional cooperative study. CPT-11 therapy resulted in four complete remissions (CRs) and three partial remissions (PRs) in 29 assessable non-Hodgkin's lymphoma (NHL) patients, one PR in three Hodgkin's disease (HD), one CR and one PR in 11 acute lymphoblastic leukemia (ALL), and one PR in 15 acute myelogenous leukemia (AML) patients. Single infusion of 200 mg/m2 every 3 to 4 weeks produced no response in both leukemia and lymphoma patients. Sixty-minute infusions of 40 mg/m2/d for 5 days every 3 to 4 weeks or for 3 days weekly produced four CRs (17%) and four PRs (17%) in 24 patients with malignant lymphoma. Sixty-minute infusions of 20 mg/m2 twice a day for 7 days every 3 to 4 weeks resulted in one CR and two PRs in 12 patients with acute leukemia. No response was seen in an acute leukemia patient by another treatment schedule. CPT-11 was effective in two (15%) of 13 primarily refractory leukemia and lymphoma cases, in two of four relapsed cases, and in seven (17%) of 41 relapsed and refractory cases. Major side effects were leukopenia (91%) and gastrointestinal (GI) (76%). CPT-11 was shown to be effective against refractory leukemia and lymphoma, and thus deserves further clinical study; the novel antitumor activity mode of this drug predicts no cross-resistance to presently available antitumor drugs.

Characteristics of the H(+)-translocating adenosine triphosphatase of Vibrio parahaemolyticus.

We have characterized H(+)-translocating adenosine triphosphatase (ATPase) in membrane vesicles of Vibrio parahaemolyticus. The ATPase required high concentrations (about 0.5 M) of Na2SO4 (or other salts) for its maximum activity. Magnesium ion stimulated the ATPase activity, but Ca2+ did not. The activity of ATPase was inhibited by tetrachlorosalicylanilide, an H+ conductor, but not by another H+ conductor, carbonylcyanide-m-chlorophenylhydrazone. The activity was strongly inhibited by dicyclohexylcarbodiimide or Zn2+, and partially inhibited by azide, but not at all by vanadate.

Enhancement of the mutagenicity of Trp-P-1, Trp-P-2 and benzo[a]pyrene by bupleuri radix extract.

Boiling-water extract of Korean-Saiko (Bupleuri Radix, from South Korea, Bupleurum falcatum L.) enhanced the mutagenic activity of Trp-P-1, Trp-P-2 and benzo[a]pyrene with S9mix. The boiling-water extract was fractionated with ether and then n-BuOH. Both the ether and the n-BuOH fractions also enhanced mutagenicity of Trp-P-1, respectively. The n-BuOH fraction was separated into seven fractions by silica gel chromatography and the chloroform eluate had the strongest enhancing effect on the mutagenic activity of Trp-P-1 with S9mix. The chloroform eluate fraction was further separated into five spots by thin-layer chromatography. Two of the spots had the strongest enhancing effect on the mutagenic activity of Trp-P-1. Since saikosaponin is a well known component in Bupleuri Radix, the effects of its existence were tested and saikosaponins a and c were found. The enhancement activity of saikosaponin a was very weak. The effective components are now being studied.

Coronary artery disease associated with aortitis syndrome in older patients.

Coronary artery disease associated with aortitis syndrome was studied in relation to the coronary artery lesions and age. Four (21%) of 19 patients with aortitis syndrome had coronary artery disease and the mean age of the 4 patients was 53 +/- 14 years. The coronary artery lesions included ostial stenosis in 2 patients and main branch stenosis in 2, but both types of lesion were isolated. The latter patients were older and had coronary lesions like arteriosclerosis on angiogram. Myocardial infarction and congestive heart failure were not present, but a severe coarctation of the aorta was observed in the latter. It was suggested that coronary artery disease might not be rare past middle age, and might include coronary ostial stenosis caused by aortitis and coronary main branch stenosis probably based on secondary arteriosclerosis whose onset was past middle age.

Glycine-insensitive NMDA-sensitive receptor expressed in Xenopus oocytes by guinea pig cerebellar mRNA.

The electrophysiological and pharmacological properties of N-methyl-D-aspartate (NMDA)-sensitive receptors expressed in Xenopus oocytes by injection of total poly(A)+RNAs (mRNAs) from the cerebellum and cerebrum of guinea pigs were compared. The inward current induced by NMDA under voltage-clamp in cerebellar mRNA-injected oocytes was depressed in a voltage-dependent fashion by Mg2+ to show a negative slope conductance and selectively antagonized by D-2-amino-5-phosphonovalerate (D-APV) and phencyclidine (PCP). Glycine (0.01-10 microM) did not potentiate NMDA-induced currents in cerebellar mRNA-injected oocytes, while it potentiated NMDA-induced currents in cerebral mRNA-injected oocytes in a dose-dependent fashion. 6-Cyano-7-nitroquinoxaline-2,3-dione and 7-chlorokynure-nate suppressed the NMDA response but significantly less potently in cerebellar mRNA-injected oocytes than in cerebral mRNA-injected oocytes. These results suggest that the NMDA-sensitive receptor expressed in Xenopus oocytes by guinea pig cerebellar mRNA resembles the cerebral NMDA receptor in its high sensitivities to Mg2+, PCP, and D-APV, but it is distinct from the cerebral NMDA receptor in responsiveness to glycine.

[Quantitative measurement of liver fatty acid binding protein in human gastrointestinal tract].

Quantitative measurement of the liver fatty acid binding protein (L-FABP) in human gastrointestinal tract was carried out by the single radial immunodiffusion method, and its tissue distribution was also determined. L-FABP existed only in the epithelial absorptive cells of duodenum, jejunum, ileum and colon, but not in esophagus and stomach. Jejunum showed the highest content of L-FABP at the level of 1.5% of the cytosolic protein. These findings support the concept that FABP has a role in the fatty acid absorption and transport. Intestinal L-FABP content revealed no difference between patients given ordinary diet and intravenous hyperalimentation, although earlier report by experimental animal showed that oral high fat loading increased the L-FABP concentration. L-FABP concentration had no correlation with the duration of IVH, but was well correlated with serum triglyceride and total-cholesterol levels. The colon of normal controls showed similar level of L-FABP concentration to that of distal intestine. L-FABP content in the colorectal cancer tissues and that in the specimen of the ulcerative colitis was also assayed, the former showed similar level to, and the latter showed significantly lower level than that in normal controls.
