Cerebellar granule cells in culture: monosynaptic connections with Purkinje cells and ionic currents.

Electrophysiological properties of cerebellar granule cells and synapses between granule and Purkinje cells were studied in dissociated cultures. Electrophysiological properties of neurons and synapses in the mammalian central nervous system are best studied in dissociated cell cultures because of good target cell visibility, control over the contents of the extracellular solution, and the feasibility of whole-cell patch electrode recording, which has been a powerful technique in analyzing biophysical properties of ionic channels in small cells. We have applied this whole-cell recording technique to cultured cerebellar granule cells whose electrophysiological properties have been almost unknown because of their small cell size. In this study, simultaneous intracellular recordings from presynaptic granule and postsynaptic Purkinje cells demonstrated that granule cells made functional monosynaptic connections with Purkinje cells in dissociated cell cultures. Further, the existence of Na, Ca, and K channels in granule cells is demonstrated by external ion substitutions.

Urban concentration and rural growth: a two-sector analysis.

"This paper examines the mechanism and extent of urban concentration using a two-sector growth model. The cause of urbanization is sought in the existence of urban scale economies. Assuming that temporary production decisions are concerned with optimal labor inputs and that profit incomes are reinvested according to relative profitability, the model shows persistent tendency for urban concentration and the transition of the economy from unlimited to limited labor supply. A simple example suggests that urban concentration is more likely than rural expansion with urban contraction."

Contact sensitivity to unsaponifiable substances in sesame oil.

A case of contact sensitivity to the unsaponifiable substances in sesame oil is reported. An ointment composed of 60% sesame oil was used for the treatment of a burn on the forearm. 10 days later, contact dermatitis developed. The allergens were shown to be sasamin and sesamolin, which are unsaponifiable substances in sesame oil.

Clinical features of aluminum-associated bone disease in long-term hemodialysis patients.

We encountered 11 patients with aluminum-associated bone disease (AABD), and treated them with deferoxamine (DFO). In 3 patients, a second bone biopsy was done during DFO treatment. Clinical features of AABD were compared with surgically proven secondary hyperparathyroidism (2 degrees HPT) with osteitis fibrosa on X-ray. Patients with AABD had disabling bone pain. This disease showed radiological signs ranging from normal, localized bone atrophy, to multiple fractures. It was characterized by increased soft tissue activity and localized abnormal uptake of 99mTc-MDP, detected by skeletal scintigrams. Patients with AABD had low levels of parathyroid hormone and alkaline phosphatase, but high aluminum (Al) levels compared to those with 2 degrees HPT. Serum Al increased after DFO administration, and the patients improved both clinically and histologically. 1-alpha-Hydroxyvitamin D3 (1-alpha-OH D3) was not effective for AABD. We concluded that the administration of antacids containing Al should be minimized in dialysis patients.

[New additional operation in the prevention of gastric stasis after selective gastric vagotomy with antrectomy].

The new additional operation for the prevention of the gastric stasis after the selective gastric vagotomy with antrectomy (SV + A) was performed. Our additional operative procedure was followed: After selective gastric vagotomy and antrectomy, gastroduodenostomy was anastomosed at acute angle with the longitudinalis of the stomach. Then, both lesser and greater omentum were incised outside of the gastric vessels. After these procedure, posterior wall sided to lesser curvature was fixed with the edge of the hepatogastric ligament and posterior wall sided to greater curvature was fixed with the retroperitoneum inferior to the pancreas by several sutures. The outcome of these treatments of the additional operation on SV + A enabled to shorten the duration of drainage of gastric juice, as well as smooth intake. By fluoroscopic examination one month after operation, gastric stasis was observed on SV + A due to the contrast medium stored in the ptotic corpus, whereas, in the case of SV + A with our additional operation, smooth gastric emptying was observed without any stasis of the contrast medium, because the corpus was placed upper from the anastomosis portion. In conclusion, our additional operation to SV + A was able to perform easy and safely, and was observed the effective prevention of gastric stasis.

[Intra-arterial administration of epirubicin in the treatment of non-resectable hepatocellular carcinoma. Epirubicin Study Group for Hepatocellular Carcinoma].

A group study was conducted to investigate the effect of intra-hepatic arterial administration of epirubicin in the treatment of non-resectable hepatocellular carcinoma(HCC). Sixty-four patients were entered into the study. There were 51 males and 13 females, with an age range from 32 to 79 years, the average being 59.1 years. Fifty-four patients had associated cirrhosis of the liver. Epirubicin at a dose of 60-90 mg/m2 was infused as a bolus into the hepatic artery 1 to 4 times (average 1.8) at an interval of 3 weeks to 3 months. Tumor size was properly evaluated in 53 patients. There were 1 CR, 7 PR, 7 MR, 27 NC, and 11 PD. Thus, the response rate (CR + PR) was 15.1%. Seventeen patients are still alive 305 to 720 days (mean 505 days) after the initial treatment. The most common side effects of this drug were bone marrow suppression, gastrointestinal complaints, and alopecia. Cardiac toxicity was negligible at the doses used in this study. A retrospective comparison of the present results with those for patients treated by intra-arterial administration of doxorubicin demonstrated that epirubicin is more effective than doxorubicin in terms of survival rate. The current study indicates that epirubicin may be a promising alternative in the treatment of HCC. The appropriate dosage of the drug and the interval for infusion are to be elucidated. Also, the combination of epirubicin with other cytostatic drugs is open for study.

Wetting characteristics and blood clotting on surfaces of copoly(gamma-Benzyl-L-glutamate, gamma-hydroxyethyl-L-glutamine).

The film surface of poly(gamma-benzyl-L-glutamate) (PBLG) was modified with 2-aminoethanol to enhance its hydrophilicity. Controlling the reaction conditions of PBLG and 2-aminoethanol, various types of copoly(gamma-benzyl-L-glutamate, gamma-hydroxyethyl-L-glutamine) film surfaces were obtained. Surface free energy (gamma sv), the dispersive component of gamma sv (gamma dsv), the nondispersive component of gamma sv (gamma psv), and the interfacial free energy of polymer surface with water (gamma sw), which were obtained by using the contact angle measurement and calculation method proposed by Andrade et al., were changed remarkably by the aminolysis. The gamma sv value increased after 2 h of aminolysis from 48.2 (PBLG) to 65.3 dyn/cm and gradually increased to around 70 dyn/cm after 12 h reaction. (gamma dsv) and (gamma psv) changed from 31.0 and 17.2 dyn/cm (PBLG) to 26.5 and 44.3 dyn/cm, respectively. These parameters of the material surfaces, modified over 12 h reaction, were found to be similar to those of the surfaces of canine aorta, vein, and human fibrin membrane. Blood clotting times on these polymer surfaces were comparatively longer than on siliconized glass surfaces.

Removing dowels in difficult teeth.

An improved method for removing dowels from endodontically treated teeth has been presented. It includes a cast supplementary device that supports the tooth while the dowel is being removed. Although the procedure requires more than one appointment, it provides a safe method for removing the dowel from the tooth.
