Cholestyramine, a bile-binding agent, markedly inhibited the development of gastric lesions induced by water-immersion stress in rats and of gastric and intestinal lesions induced by indomethacin in rats and dogs with an intact pylorus. However, cholestyramine had no effect on gastric lesions induced by stress in pylorus-ligated rats and on aspirin-induced gastric lesions in rats and dogs with or without pylorus ligation. Hydrotalcite, a potent antacid, significantly inhibited the gastric lesions induced by stress and aspirin in rats and dogs with or without pylorus ligation. Hydrotalcite also inhibited the gastric lesions induced by a single administration of indomethacin in rats with an intact pylorus. In contrast, the agent had no effect on gastric and intestinal lesions induced by repeated administration of indomethacin to rats and dogs with an intact pylorus. These results suggest that while bile probably plays an important role in the pathogenesis of stress-induced gastric lesions and of indomethacin-induced gastric and intestinal lesions, bile does not appear to play a role in the pathogenesis of aspirin-induced gastric lesions.
In Ames test, the quinoline derivatives, 6-nitroquinoline (6-NQ), 8-nitroquinoline (8-NQ), 6-methylquinoline (6-NQ), 8-methylquinoline (8-MQ) and 8-hydroxyquinoline (8-HQ) are mutagenic, whereas 5,7-dibromo-quinoline (5,7-DQ) is not. These compounds were administered to groups of male and female F344 rats in their diet for 104 weeks. 8-NQ induced forestomach tumors in male and female rats: it induced squamous cell papillomas in 28 (93.3%) of 30 males and in 36 (97.3%) of 37 females, and squamous cell carcinomas in 20 (66.7%) of 30 males and 24 (64.9%) of 37 females. Thus there was no sex difference in its carcinogenicity. 6-NQ induced only focal hyperplasia of the forestomach at an incidence of 30.0% in male rats and 52.8% in female rats. These results indicate a discrepancy between the mutagenicities of quinoline derivatives and their carcinogenicities to rats: only 8-NQ was carcinogenic to F344 rats.
Factors enhancing renal carcinogenesis induced by formic acid 2-[4-(5-nitro-2-furyl)-2-thiazolyl] hydrazide (FNT) were studied. Five groups of rats were used. Groups 1 and 2 were subjected to right nephrectomy and 2 days late they were injected with 20 mg/kg body weight of dimethylnitrosamine (DMN). From one week after nephrectomy, group 1 was fed on 0.2% FNT diet continuously to the end of the experiment, and group 2 was given normal diet. In week 2, they were given 400 mg/kg body weight of chloroform (CHCl3) orally. Groups 3 and 4 were given ChCl3 first and 2 days later injected with DMN. Group 3 was then fed FNT diet and group 4 was fed basal diet continuously. In week 2, groups 3 and 4 were subjected to unilateral nephrectomy. Group 5 was given FNT diet without other treatment. The incidences of tubular hyperplasia in groups 1, 3 and 5 were 53.3%, 52.9%, and 0%, respectively, at week 16 and 100%, 94.9% and 94.1%, respectively, at week 32. Moreover, the incidences of renal cell tumors in these groups were 85.0%, 88.9% and 17.6%, respectively, at week 32. Thus, the inductions of tubular hyperplasia and renal cell tumor were markedly enhanced by combined treatment with CHCl3, DMN, unilateral nephrectomy and FNT in the initiating and promoting stages.

The effect of heparin Na on the actions of various bronchoconstrictors was examined. Guinea pig tracheal strips were suspended in bioassay glass chambers and superfused with Krebs-Henseleit solution, pH 7.4, at 37 degrees C saturated with oxygen and carbon dioxide gas (95:5, v/v). Contraction of the tissue was detected by an isotonic transducer and displayed on a polyrecorder. Heparin Na attenuated histamine-, prostaglandin F2 alpha-, acetylcholine-, serotonin-induced contractile responses dose-dependently, and shifted the dose-response curves of histamine, prostaglandin F2 alpha, acetylcholine, serotonin and bradykinin downward.
