[Experimental investigation on intrapulmonary and tracheobronchial distribution of ampicillin after administration of a beta-lactam antibiotic, bacampicillin].

Bacampicillin (BAPC) is a esterified derivative of ampicillin (ABPC) developed recently by the Swedish company. In present investigation we measured the concentration of ABPC in plasma, lung and tracheobronchial tissues from normal, pleuritis and pneumonitis male albino rabbits after an oral administration of BAPC. In these 3 groups plasma level of ABPC reached the maximum at 30-50 minutes after oral administration, then showed a gradual decrease. The decaying curves of pleuritis and pneumonitis groups were steeper than that of control group. Plasma level of ABPC in pneumonitis group at the maximum value was about 1/2 comparing with that in control and pleuritis groups. ABPC levels of tracheobronchial tissues in pleuritis and pneumonitis groups were higher than that in control group at 60 minutes after oral administration of BAPC. On the other hand, tissue level of ABPC in control group was higher than that in pleuritis and pneumonitis groups at 120 minutes after oral administration of BAPC. ABPC level of lung tissues (L1 and L5) in control group was higher than that in pleuritis and pneumonitis groups at 60 minutes after oral administration of BAPC, on the other hand ABPC level of L3 in control group was lower than that in pleuritis and pneumonitis groups. ABPC levels of lung tissues (L1 - L5) at 120 minutes after oral administration of BAPC were higher in control group comparing with that in pleuritis and pneumonitis groups.

Contiguous malignant astrocytoma and Wilms'-like tumor in the brain.

A case of primary brain tumor composed of two contiguous neoplasms in presented. At operation, a nodular tumor was embedded in the infiltrating tumor within the brain parenchyma. With light and electron microscopy, the nodular tumor was similar to Wilms' tumor (nephroblastoma). The infiltrating tumor was malignant astrocytoma. Autopsy revealed, besides the recurrence of malignant astrocytoma in the brain and its subarachnoid dissemination, extracranial metastases to the abdominal cavity, liver, lung, and bone marrow. Recurrent and metastatic tumors were glioblastoma multiforme, which was more malignant than the surgical specimen. The possibility of metastatic Wilms' tumor from the kidney was completely ruled out by extensive autopsy survey. The authors present an extremely rare tumor including detailed observations on the electron microscopic appearance of the tumor; the histogenesis of these tumors is briefly discussed.

Mepirizole-induced duodenal ulcers in rats and their pathogenesis.

Duodenal ulcers were produced in rats following either an oral or parenteral administration of 200 mg/kg of mepirizole, a nonsteroidal antiinflammatory agent. Deep ulcers, including perforated ones, were induced in the proximal duodenum with an incidence of over 90%. Mortality due to perforation was less than 5%. The agent also induced several erosions in the antrum. Feeding of animals after the ingestion of mepirizole markedly suppressed the development of both duodenal ulcers and gastric erosions. Antacids, anticholinergic agents, a histamine H2-receptor antagonist and 16-DMPGE2 dose-dependently inhibited mepirizole-induced duodenal ulcers. Gastric erosions were also significantly inhibited by antacids and anticholinergic agents but not by a histamine H2-receptor antagonist and 16-DMPGE2. Intraduodenally administered mepirizole dose-dependently inhibited the gastric secretion in pylorus-ligated rats. This ulcer model should be useful for screening antiulcer agents and for the study of pathogenesis of duodenal ulcers and gastric erosions.

Effect of ipratropium bromide on the action of bronchoactive agents.

The effect of (8r)-3 alpha-hydroxy-8-isopropyl-1 alpha H, 5 alpha H-tropanium-bromide-(+/-)-tropate (ipratropium bromide, Sch 1000), a newly developed anticholinergic agent, on the action of various bronchoactive agents was examined using isolated guinea pig strips. Ipratropium bromide has not only an anticholinergic effect but also has an attenuating one on the contractile responses induced by serotonin, prostaglandin F2 alpha, histamine and bradykinin, and has a potentiating one on the relaxation responses induced by prostaglandin E2, isoprenaline and adrenaline. Ipratropium bromide could be a useful therapeutic agent for patients with chronic obstructive lung diseases.
