Experimental transplacental infection of pregnant dogs with canine herpesvirus.

Fetal infection was established in 28 of 33 pups obtained from 7 pregnant dogs (47th to 53rd day of gestation) inoculated IV with canine herpesvirus (CHV). The pathogenesis of transplacental CHV infection was investigated by using pathologic examination, immunofluorescence microscopy, and viral assay. Infection with the virus was recognized in 13 of 18 neonatal pups obtained from 4 bitches by spontaneous delivery at full term. Eleven pups died within a week of birth and all had characteristic lesions of CHV infection. Thirteen live-born and 2 stillborn pups derived from 3 bitches by cesarean section died within 9 days of birth. Characteristic lesions of CHV infection were observed in various organs, such as liver, spleen, kidney, and lung. Fourteen of the fetal placentas had variable degree of necrotizing lesions. The lesions were characterized by degenerative changes and focal necrosis in the placental labyrinth. Lesions were present in the walls of allantoic blood vessels. Intranuclear inclusions were evident in degenerating trophoblast cells and in the cells of maternal and fetal blood vessels. Infective virus and viral antigen were detected in the various organs of pups and in the trophoblastic cells of junctional zone. Virus also was found in the walls of maternal and fetal blood vessels of the placental tissues. The results strongly indicated that transplacental transmission of CHV may occur in pregnant dogs at a late stage of gestation.

Chemically induced rat B cell leukemia KNL-14, and its major histocompatibility complex products.

Five rat leukemia cell lines induced with chemical agents and/or leukemia viruses were investigated in terms of their surface antigens. One of them, KNL-14, which was induced with 1-butyl-1-nitrosourea in WKA rats had RT1-B region-associated antigens on the cell surface. In addition, Thy-1.1 antigens and surface and cytoplasmic mu chains were detected by serological and immunochemical means. From the results it was concluded that this leukemia originated from immature B cells. In the present work, RT1 antigens on the KNL-14 cells were further investigated. The class I and class II antigens were examined and KNL-14 cells were found to have phenotypical antigens and immunogenicity similar to those of B cells from the WKA lymph nodes. As for class II antigens, which are responsible for various immunologic functions, two specificities were detected, Ba-1.2 and Ba-2.7. These KNL-14 cells should be useful for studying the process of rat B cell differentiation and for immunochemical studies of the rat MHC gene products.

Effect of phospholipase C on lipid peroxidation of rat liver mitochondria.

Effect of phospholipase C on lipid peroxidation in rat liver mitochondria was studied. The lipid peroxidation of mitochondria in the presence of ascorbic acid and Fe2+ (FeSO4) was inhibited by approximately 45% after pretreatment of phospholipase C for 30 min. However, when phosphatidylcholine instead of mitochondria was used for the peroxidation, phospholipase C showed no inhibition on lipid peroxidation. Therefore, it may be considered that phospholipids in mitochondria are modified in diglyceride structure after digestion with phospholipase C, and the diglyceride is poor substrate for lipid peroxidation in mitochondria.

Effect of ascorbic acid and ferrous iron on p-aminohippurate accumulation by rat kidney cortical slices.

The effect of ascorbic acid and Fe2+ on p-aminohippurate accumulation by rat kidney slices was examined. Ascorbic acid and Fe2+ promoted lipid peroxidation of rat renal cortical slices was found to be dose related. Moreover, ascorbic acid (1.0 mM) and Fe2+ (0.4 mM) increased tissue water and decreased the accumulation of p-aminohippurate. The addition of N,N'-diphenyl-p-phenylenediamine (antioxidant) at the concentration of 1 x 10(-6) M recovered the accumulation of p-aminohippurate by about 43%. The apparent Km of p-aminohippurate uptake was increased by ascorbic acid and Fe2+ with no change in the apparent V. These data suggest that ascorbic acid and Fe2+ can cause a significant alteration in p-aminohippurate and water transport of renal cortical slices and that these effects may be, at least, partly due to the lipid peroxidative action of ascorbic acid and Fe2+.

Effect of various diuretics on lipid peroxidation in rat renal cortical mitochondria and in the supernatant.

Effect of various diuretics of lipid peroxidation in rat kidney cortical mitochondria and in the supernatant (11,000 x g) was studied. Mersalyl (mercurial diuretic) markedly stimulated the lipid peroxidation in mitochondria and in the supernatant at a concentration of 1 mM, and metolazone (sulfonamide derivative, 1 mM) stimulated only that in the supernatant. Mersalyl-induced lipid peroxidation was markedly inhibited by several classical radical scavengers such as sodium diethyldithiocarbamate, 2,6-di-tert-butylphenol and N,N'-diphenyl-p-phenylenediamine, but metolazone-induced lipid peroxidation was not inhibited by these scavengers. The addition of superoxide dismutase to the reaction mixture inhibited the mersalyl-induced lipid peroxidation, but did not inhibit the peroxidation by metolazone or ascorbate. These results suggest that lipid peroxidation by mersalyl is mediated by free radicals or superoxide anion, which were probably produced by mersalyl, and the mercurial diuretics-induced nephrotoxicity may be due to the lipid peroxidation in the kidney.

A case of Maffucci's syndrome associated with primary hyperparathyroidism.

Maffucci's syndrome (association of multiple enchondromatosis and multiple hemangiomata) is rarely associated with other ectodermal tumors. The present case is the second in literature in which the Maffucci's syndrome is associated with the parathyroid adenoma resulting in primary hyperparathyroidism. Although the cystic lesions of the bone and the ectopic calcifications are common to both diseases, pathogenesis apparently not related. The association of parathyroid adenoma with Maffucci's syndrome suggests the potential involvement of mesenchymal organs in this ectodermal neoplastic disorder.

Transformed phenotypes in long-term cultures persistently infected with bovine leukemia virus.

Two cell lines, 3178 and FLK, were established respectively from calf form bovine lymphosarcoma and from fetal lamb kidney cells inoculated in vitro with bovine leukemia virus. These two cell lines persistently infected with bovine leukemia virus were maintained for more than 150 passages over three years. They exhibited the characteristics of transformed cell lines in vitro: 1) anchorage independence, 2) increased saturation density and decreased population doubling time, 3) increased uptake of 2-deoxy-D-glucose and 4) tumorigenicity in athymic nude mice.
