Stereochemical importance of fucosterol epoxide in the conversion of sitosterol into cholesterol in the silkworm Bombyx mori.

Studies on the stereochemistry of 24,28-epoxy-24-ethylcholesterol, a key intermediate in sitosterol to cholesterol conversion in insects, were undertaken. In nutritional experiments, the 24R,28S and 24S,28R stereoisomers (isofucosterol epoxides) were unable to support growth and development of silkworm larvae. Incubation of 3 alpha-3H-labeled (24R,28R)- and (24S,28S)-epoxide (fucosterol epoxides) with a cell-free preparation from silkworm guts resulted in effective conversion into desmosterol and cholesterol; (24R,28R)-epoxide was the slightly better substrate. Fucosterol incubation yielded (24R,28R)-epoxide and the 24S,28S isomer in approximately equal amounts, while slightly preferential formation of the 24R,28R isomer was observed in in vivo experiments. These results indicate that both the formation of the epoxide from fucosterol and its conversion to desmosterol proceed with a low degree of stereospecificity.

Identification of carcinoembryonic antigen in the C-cell of the normal thyroid.

Carcinoembryonic antigen (CEA) activity was confirmed in the C-cell of the normal thyroid by immunohistochemical techniques. This suggests that CEA production in medullary carcinoma of the thyroid is not initiated by carcinogenesis, but reflects a function of the normal C-cell. It is not yet clear whether CEA production in the C-cell may be influenced by carcinogenesis. The C-cell is the first APUD cell that was confirmed to have CEA activity.

Cohelical arrangement of the DNA strand in the core of Marek's disease virus particles.

Electron microscopic studies on Marek's disease virus demonstrated that fibrils of 20 to 30 A were arranged in a cohelical configuration within an electronopaque band of about 100 A thickness in the core of immature particles. These fibrils were presumed to be the DNA double helix itself. This configuration may represent an arrangement of the herpesvirus DNA.

A case of primary hyperparathyroidism with low serum alkaline phosphatase levels.

A 23-year-old man with primary hyperparathyroidism which was typical except for reduced alkaline phosphatase activity is reported. Histological examination of surgical specimens revealed chief cell hyperplasia of the parathyroid glands. Systemic abnormalities of alkaline phosphatase were demonstrated, i.e., marked reduction of all isoenzymes and undetectable osseous enzyme in the serum, abnormal distribution of the enzyme in hepatocytes and diminished enzyme activities in leukocytes. In addition, diminished bone remodeling activity was revealed in a biopsy specimen of the rib. The association of hypophosphatasia is highly unlikely, because of normal urinary excretion of phosphoethanolamine, lack of osteomalacia, and no indication of an hereditary factor. The causal relationship between low remodeling activity and abnormalities in alkaline phosphatase was suggested.

Effect of thyroid-stimulating hormone and norepinephrine on cyclic AMP levels in human normal thyroids and human thyroid tumors.

The effect of TSH (100mU/ml) and norepinephrine (100 muM) on the cyclic AMP levels was studied in 10 human normal tissues, 10 thyroid adenomas and 4 thyroid carcinomas (3 papillary and 1 follicular). Normal tissues responded to TSH with a marked elevation of the cyclic AMP level. Response patterns of 10 thyroid adenomas to TSH were variable; the patterns of 6 cases resembled those of normal tissues, 3 responded mildly, and one had no response to TSH. Thyroid carcinomas had a higher basal level of cyclic AMP than those of normal tissues, although they responded only slightly to TSH. Two among 4 thyroid carcinomas had no response to TSH. Norepinephrine stimulated the accumulation of cyclic AMP in 4 thyroid adenomas and 3 thyroid carcinomas, while it had little effect on normal tissues. Responses to norepinephrine was observed only in thyroid tumors, although they had low response to TSH. It is suggested from these results that tumor cells originating from thyroid follicular cells have a modified response to hormones due to neoplastic alterations.

C26 sterol in a human urine.

A new C26 sterol, 22-trans-27-norcholesta-5,22-dien-3 beta-ol, was found in the urine of a 6-year-old girl, with a clinical diagnosis of congenital adrenal hyperplasia of the salt losing type, accompanied by symptoms of mixed sex anatomy and skin pigmentation. The structure of the sterol was determined by comparison with the synthetic compound. The sterol was also detected in ther serum. This appears to be the first case in which a C26 sterol has occurred in mammalia.
