Novel method for the preparation of controlled-release theophylline granules coated with a polyelectrolyte complex of sodium polyphosphate-chitosan.

A novel method for the preparation of theophylline granules coated with a polyelectrolyte complex of sodium tripolyphosphate and chitosan was developed. The theophylline granules containing sodium tripolyphosphate were stirred in an HCl solution of chitosan. During the mixing, the dissolved sodium tripolyphosphate in the granule moved to the surface and reacted with the chitosan, resulting in the formation of the polyelectrolyte complex film. The factors affecting the drug content, the particle size, and the coating-film thickness of the resultant coated granules were determined. The theophylline content in the coated granule decreased with increasing content ratio of sodium tripolyphosphate to theophylline in the original granule and with increasing chitosan concentration in the coating solution. The coated granule size increased with increasing chitosan concentration in the coating solution and with decreasing agitation speed. The coating-film thickness increased with an increase in the chitosan concentration, the pH of the coating solution, and the sodium tripolyphosphate to theophylline content ratio in the original granule. The drug-release pattern of the coated granules followed zero-order kinetics and the release rates were significantly reduced compared with that of the original granule.

Chemical Phenomena↗

Visual sensitivity and Wiener kernels.

Visual sensitivity is defined in terms of the static and dynamic parts of a photo-evoked response. The first-order kernel induced by a white-noise modulated light is associated directly with both the incremental and contrast sensitivities classically defined, making a comprehensive measure of neuron sensitivity in the visual system.

Cerebral blood flow velocity in preterm neonates.

The postnatal changes of cerebral blood flow velocity in 35 preterm neonates with or without complications were measured with a bidirectional Doppler flow meter. In the cases without complications, the pulsatility index for the anterior cerebral artery (ACA-PI) was high shortly after birth, and gradually decreased after 6 hrs as that in normal term neonates. However ACA-PI showed large variations in the cases with respiratory distress. On the other hand, ACA-PI in the cases with subependymal hemorrhage (SEH) was low during the first 6 hrs after birth, and thereafter showed no significant changes. ACA-PI in the cases with intraventricular hemorrhage (IVH) was high during the first 6 hrs after birth and then decreased when IVH was found on ultrasound scanning. ACA-PI increased gradually with progression of post-hemorrhagic ventricular dilatation. The difference of high PI in IVH and low PI in SEH during the first few hrs after birth suggests that the pathogenesis of SEH may be different from the mechanism of intraventricular rupture from SEH. Low PI values in the infants with SEH may be related to intrapartum events, while the changing patterns of high to low PI in the infants with IVH suggest postnatal hemodynamic changes.

Heat resistance, spore germination, and enterotoxigenicity of Clostridium perfringens.

Heat resistance at 95 C, heat activation at 75 C, and germination response were determined for spores of 10 serotype strains of Clostridium perfringens type A, including five heat-resistant and five heat-sensitive strains. The D95-values ranged from 17.6 to 63.0 and from 1.3 to 2.8 for the heat-resistant and the heat-sensitive strains, respectively. The heat-activation values, the ratios between the heated and unheated viable counts of spore suspensions, ranged from 0.0035 to 0.65 and from 6.5 to 60.0 for the heat-sensitive and the heat-resistant strains, respectively. Spores of these strains were divided into two distinct germination types on the basis of their germination response; spores of the heat-resistant strains germinated in KC1 medium after heat activation (K-type), and spores of the heat-sensitive strains germinated in a mixture of L-alanine, inosine, and CaCl2 in the presence of CO2 without heat activation (A-type). The strains were tested for enterotoxigenicity by a reversed passive latex-agglutination (RPLA) test. All the heat-resistant strains were RPLA-positive, whereas the heat-sensitive strains were all RPLA-negative. A total of 37 strains of the organism isolated from food-poisoning outbreaks were tested for spore germination and enterotoxin formation. All of the 20 heat-resistant strains showed K-type spore germination and, except for three strains, were RPLA-positive, whereas all of the 17 heat-sensitive strains showed A-type spore germination and, except for only one strain, were RPLA-negative.
