Expression levels of functional folate receptors alpha and beta are related to the number of N-glycosylated sites.

In a previous study with inhibitors of N-glycosylation, it was proposed that core glycosylation of the folate receptor (FR) is required for the proper folding of the protein [Luhrs (1991) Blood 77, 1171-1180]. The human FR isoforms type alpha and type beta have three and two candidate sites for N-glycosylation respectively, only one of which is conserved. The significance of N-glycosylation at each of these loci in the expression and function of FR was examined by eliminating the sites both individually and in combination by introducing Asn-->Gln substitutions. Translation experiments in vitro showed that the mutations did not alter the synthetic rates of the polypeptides. The recombinant proteins were expressed in human 293 fibroblasts. Treatment with N-glycanase and analysis by Western blotting of the wild-type and mutant proteins revealed that all of the candidate sites in both FR-alpha and FR-beta are glycosylated. When all of the N-glycosylation sites were abolished, 2% and 8% of FR-alpha and FR-beta respectively were expressed on the cell surface compared with the corresponding wild-type proteins; the residual FR polypeptides in the cell lysates were unable to bind [3H]folic acid. In both the proteins, the inclusion of each additional N-glycosylation site partly contributed to restoration of cell surface [3H]folic acid binding and receptor-mediated folate transport. Further, in FR-beta the introduction of an additional unnatural site of N-glycosylation resulted in the enhancement of the expression of the cell surface receptor compared with the wild-type protein. The results indicate that the total mass of N-glycosylation, not a specific locus of the modification, is critical for the efficient folding and optimal expression of functional FR-alpha and FR-beta.

Identification of amino acid residues that determine the differential ligand specificities of folate receptors alpha and beta.

The homologous folate receptor (FR) types alpha and beta from both human and murine sources have opposite stereospecificities for reduced folate coenzymes and different affinities for a variety of (anti)folate compounds. The present study identifies the critical amino acid sequence divergence underlying functional differences between FR-alpha and FR-beta. Chimeric constructs of the cDNAs encoding human FR-alpha and FR-beta were expressed in human 293 fibroblasts. The resulting membrane associated proteins were characterized in terms of their ability to bind [3H]folic acid and their relative affinities for the (6S) and (6R) diastereoisomers of N5-methyltetrahydrofolate. Substitution of the amino-terminal portion (residues 1-92) in the mature FR-alpha polypeptide with the corresponding segment of FR-beta resulted in folate binding characteristics similar to FR-beta. Next, a series of chimeric constructs were generated, involving substitution of progressively shorter segments within residues 1-92 in FR-alpha with the corresponding peptides of FR-beta. In this fashion, it was determined that the alanine residue at position 49 in FR-alpha was critical for its functional divergence from FR-beta, since substitution at this position with Leu (the corresponding residue in FR-beta) resulted in the folate binding characteristics of FR-beta. Reciprocal substitution in FR-beta with peptide 1-92 of FR-alpha resulted in poor expression of a [3H]folic acid binding protein. By analysis of chimeric constructs, the poor [3H]folic acid binding of the FR-alpha(1-92)/beta(93-237) chimera could be attributed to interference of a short segment from FR-alpha in the vicinity of Ala 49 (peptide 39-59) with proper folding of the chimera. Conversion of the ligand binding properties of FR-beta to those of FR-alpha required the reciprocal mutation of Leu 49 to Ala, but in addition, substitution of one or more residues downstream of amino acid 92 of FR-beta with the corresponding residues in FR-alpha was essential. The homologous murine FR types alpha and beta, which are functionally analogous to the human receptor isoforms, also contain a similar Ala vs Leu substitution. These results indicate that steric/hydrophobic effects of the side chains of Leu vs Ala at position 49 will critically modulate the affinities and stereospecificities of FR isoforms for folate compounds. Furthermore, additional amino acid sequence divergence at one or more positions downstream of residue 92 in FR-alpha is also an essential determinant of the unique functional characteristics of this receptor isoform.

Endometrial hyperplasia and apoptosis following neonatal diethylstilbestrol exposure and subsequent estrogen stimulation in both host and transplanted hamster uteri.

Prenatal exposure to the synthetic estrogen diethylstilbestrol (DES) causes morphogenetic alterations and neoplasia in the human reproductive tract. In the hamster, neonatal DES exposure alters early uterine morphogenesis and induces endometrial adenocarcinomas in adults. We now demonstrate that the preneoplastic stages of this phenomenon in the hamster reflect an abnormal uterotropic response to estrogen that is characterized by hyperplastic lesions in the endometrial epithelium and includes an immune and/or inflammatory component. Interestingly, biochemical and in situ analysis revealed that the hyperplastic epithelium is also an active site of cell death by apoptosis. To further probe the mechanism of this phenomenon, uteri from 7-day-old control or DES-exposed donors were transplanted into the cheek pouches of control or neonatally DES-exposed adult hosts, and both host groups were treated to provide high circulating levels of estradiol. Among the four ectopic scenarios, histopathological lesions (epithelial hyperplasia, dysplasia, and apoptosis), segregated almost exclusively to the two that consisted of neonatally DES-exposed uteri. The virtual absence of lesions in control uteri transplanted to DES hosts eliminated host systemic factors as causative agents. Therefore, we conclude that DES or its metabolites alter the cellular physiology and/or composition of the developing uterus (initiating event) in such a way that it thereafter responds abnormally to estrogenic stimulation (promoting event). These observations serve to further define a unique experimental system for probing: (a) various aspects of the clinical "DES Syndrome"; (b) how estrogen regulates normal uterine growth and morphogenesis; and (c) how this process can degenerate to the unregulated neoplastic state.

Adenosine A3 receptor stimulation inhibits migration of human eosinophils.

Activation of adenosine A3 receptors (A3-R) produced a dose-dependent reduction in the chemotaxis of human eosinophils to platelet-activating factor (PAF), RANTES, and leukotriene B4 (LTB4) to a maximum of 58, 48, and 52%, respectively (P < 0.02). This effect was completely reversed by selective A3-R antagonists. In contrast, activation of A1 or A2a-R did not affect PAF-induced eosinophil chemotaxis. PAF up-regulated the expression of CDllb/CD18, down-regulated L-selectin, and also increased F-actin assembly in eosinophils. The expression of these activation markers was not influenced by A3-R, A2a, or A1-R stimulation. Activation of A3-R may play an important role in inflammation by inhibiting eosinophil migration.

Isolation of Mycoplasma hominis from a brain abscess.

Mycoplasma hominis is a commensal in the genital tract of women and has been associated with urogenital and extragenital infections. However, central nervous system infections with this organism in adults are very rare. Here we describe the recovery of M. hominis from a brain abscess associated with a postpartum infection. Seroconversion to the isolated strain was detected by both a metabolic inhibition test and an immunoblotting assay. This case demonstrates the pathogenic potential of M. hominis and the need for rapid recognition of the organism so that appropriate chemotherapeutic intervention can occur.

[Effect of Astragalus Angelica mixture on serum lipids and glomerulosclerosis in rats with nephrotic syndrome].

OBJECTIVE: To confirm the renal protective action of Astragalus Angelica mixture (AAM). METHODS: Effect of AAM was observed in puromycin amino-nucleoside induced nephrotic syndrome model of rat, and pravachol, an effective lipid lowering medicine, was used as positive control. RESULTS: AAM could not only reduce the serum levels of total cholesterol and triglyceride as pravachol did, but also could lower levels of low density lipoprotein and apolipoprotein B100 (P < 0.05). Light microscopic and immuno-histochemical examination revealed that in the same time of lowering serum lipids, this mixture could also cause reduction of glomerular sclerosing index (P < 0.01) and less accumulation of extracellular matrix (P < 0.01). CONCLUSIONS: AAM has the effects of regulating lipid metabolism, attenuating renal damage and maintaining renal function in rats with nephrotic syndrome.

[Lipid-lowering effect of pravastatin on glomerulosclerosis protection and treatment].

The lipid-lowering effect of pravastatin on glomerulosclerosis was observed in nephrotic syndrome model, which was induced in rats by using repeated puromycin injections. The histologic changes of the model were similar to those of human focal segmental glomerulosclerosis. Rats were divided into three groups, namely, normal control, nephropathy and nephropathy with pravastatin. Pravastatin was administered through gastric tube 6mg.kg-1.d-1 for 12 weeks. The lipoprotein profile and histologic development were observed. The results showed that pravastatin-treated rats had significantly lower cholesterol, triglycerides, very low density lipoprotein and high density lipoprotein than the nephropathy group at 7 weeks (P < 0.05). Moreover, the renal damage was also less marked in pravastatin-treated rats. Light microscopic and immunohistochemical examination revealed that lipid-lowering therapy was associated with significant lower level of glomerular sclerosing index and less accumulation of extracellular matrix including collagen III, IV, laminine and fibronectin as compared with the nephropathy group. However urinary protein and serum albumin levels were similar in both groups. It is suggested that lipid-lowering thyrapy may retard the progression of glomerulosclerosis and thus preserve renal function. The results imply that hyperlipoidemia plays an important role in the pathophysiological progression of renal disease.

[Histological and ultrastructural studies of experimental herpetic keratitis in the rabbit].

OBJECTIVE: In order to investigate the histological and ultrastructural changes at different post infection days in herpes simplex keratitis (HSK). METHODS: The corneas of New Zealand white rabbit eyes experimentally infected with primary HSK were studied by light microscopy and electron microscopy. The histological and ultrastructural changes in the rabbit corneas at 3, 7, 14, 21, 60 post infection days were investigated by light microscopy with HE, Mallory dyes and electron microscopy. RESULTS: Infiltration of corneas with inflammatory cells was found in rabbit eyes infected with HSK, polymorphonuclear leukocytes being predominant. Contact of inflammatory cells with fibroblast cells and corneal edema were seen in the stroma on acute post infection days 3 and 7. In severe cases inflammatory cells incarcerated in the endothelium. Only did two cases of the specimens present with herpes simplex virus type I granules in the acute stage under electron microscopy. CONCLUSION: The direct HSK-1 destruction together with immunopathogenetic injury caused by the body immune system may serve as the pathologic base of HSK. Decompensation of corneal endothelial function due to infiltration with inflammatory cells may be responsible for corneal edema.

A study of Helicobacterium pylori and prevention and treatment of chronic atrophic gastritis.

The rate of detection of helicobacterium pylori (HP) in chronic atrophic gastritis (CAG) is 67%, HP being a slightly aerobic Gram-negative bacterium. Some drugs of traditional Chinese medicine (TCM) for clearing away heat and for removing blood stasis have significant effect on inhibition of HP. The drugs in both the recipe for nourishing yin, clearing away heat and invigorating blood circulation and the recipe for clearing away heat and invigorating blood circulation have inhibitory action on HP in vitro. All patients with CAG have symptoms of damp-heat and blood stasis. Radix et Rhizoma Rhei and Rhizoma Coptidis Decoction for Purging Stomach-Fire ([symbol: see text]) as the basic recipe and "Huo Wei" ([symbol: see text]) No. 2 recipe as the supplementary have good inhibitory effect on HP in inhibition tests. The results show several forms of the medicine all resulted in significant good therapeutic effect.

[Effect of Helicobacter pylori on gastric epithelial apoptosis].

To investigate the mechanisms by which Helicobacter pylori (Hp) induces gastric mucosal damage, gastric epithelial apoptotic index (AI) of normal gastric mucosa and Hp positive gastritis before and after anti-Hp treatment was studied with the method of terminal transferase mediated dUTP nick end labeling. The results showed that AI in Hp positive gastritis was higher than that in normal mucosa (P < 0.001). After the eradication of Hp, AI fell significantly to the normal level (P < 0.001). AI in persisting Hp positive gastritis had no statistically significant decrease after the anti-Hp treatment. There was no correlation between AI and the severity of gastritis. These results indicated that Hp could induce gastric epithelial apoptosis, which may be an important mechanism involved in gastric mucosal damage.

[An adaptive method for image enhancement of retinal vascular images based on morphology and human visual properties].

A method of contrast enhancement for retinal vascular images is provided. Opening of a gray-scale image by a gray-scale structuring element(SE) can generate a background image. Enhancement is achieved by subtracting this background image from the original image and then applying a adaptive gray-scale modification technique which is based on the local contrast of the image and the lobserver's Just-Noticeable-Difference(JND). As the method includes certain human visual properties and treates smooth and detail areas of a retinal image differently, the common problems of producing ringing artifacts around sharp transition regions and excessive enhancement of noise for many traditional enhancement methods are avoided. Another advantage of this method is the retention of vessel dimension which directly benefit from using morphological opening filter. The suggested method performed very well both analytically and visually.

[Influence of leaf-picking on effective constituents of the Astragalus mongholicus roots].

After the application of the orthogonal design, experiments show that the proportion and time of leaf-picking have a great influence not only on the total content of 17 kinds of amino acids but also on that of Astragaloside. If the proportion is large and the time is delated, both contents of 17 kinds of amino acids and Astragaloside will be reduced accordingly compared to the control group. There is less influence on the contents if the time of leaf-picking is set on June 15 each year and the picking proportion is 1 out 3.
