[Environmental pollution, human exposure and its health effect of sodium pentachlorophenate in schistosomiasis prevalent area].

Sodium pentachlorophenate (Na-PCP) has been used in China for years as an molluscacide to kill oncomelania, which is an intermediate host of Schistosome. To evaluate the effects of its long-term successive usage on environment, human exposure and health, studies were carried out in Sichuan, Jiangxi, Jiangsu and Fujian provinces, with a time gap of more than one month between sample collection and last spray of Na-PCP. Results indicated that PCP contents in surface water, soil, sediment, animals and plants were significantly higher in studied areas than in control areas. The daily intake and the content in urine of PCP were also sigificantify higher in studied areas. But, there was no difference on physical and biochemical examinations except that a 22%-28% decrease of blood cholinesterase activity was found in studied areas. The health effect of impurities in Na-PCP, dioxins and furans, was assessed and discussed.

[A laplace transform method for analysis of fluorescence dynamics].

A Laplace transform formula is developed for analysis of a multi-exponential fluorescence dynamic process. A dynamic spectrum function is introduced, calculation of which directly yields parameters of the corresponding dynamic process. The formula is applied to analyze fluorescence dynamics of beta subunit from R-phycoerythrin. It is demonstrated that electronic excited-states of the biomolecular system exhibit two exponential decay processes, the faster of which relates to excitation equilibrium among chromophores, and the slower to radiative relaxation of the chromophores.

[Study on the interaction of metal ion in active center of superoxide dismutase with cobalt chloride by visible spectroscopy].

The direct interaction of metal ion in active center of copper-zinc superoxide dismutase (Cu2Zn2SOD) with inorganic cobalt chloride (CoCl2) in buffer has been first studied by visible spectroscopy. The effects of increasing amounts of CoCl2, changing pH,different acid salt and equilibrium time on the interaction have been discussed. It was found that the Co(II) of CoCl2 makes an exchanging interaction with Zn(II) in Cu2Zn2SOD and thus the partial Cu2Co2SOD is formed.

Neonatal diethylstilbestrol treatment alters the estrogen-regulated expression of both cell proliferation and apoptosis-related proto-oncogenes (c-jun, c-fos, c-myc, bax, bcl-2, and bcl-x) in the hamster uterus.

In the Syrian hamster, neonatal diethylstilbestrol (DES) treatment and then postpubertal estrogen stimulation induces hyperplasia plus apoptosis (preneoplastic responses) and ultimately neoplasia in the endometrial epithelial cell compartment. As part of a project to investigate the molecular and cellular mechanisms responsible for this phenomenon, expression of several proto-oncogenes (c-jun, c-fos, c-myc, bax, bcl-2 and bcl-x) was compared in estrogen-stimulated uteri from control versus neonatally DES-treated hamsters. According to Northern blot analysis of total uterine RNA, levels of the 3.2-kb c-jun and 2.4-kb c-myc transcripts were not altered by neonatal DES treatment. However, the 1.0 kb bax and 2.7 kb bcl-x transcript levels were significantly increased in the neonatally DES-exposed uteri. According to immunohistochemical analysis of paraformaldehyde-fixed and paraffin-embedded tissue sections, levels of c-Jun, c-Fos, c-Myc, Bax, and Bcl-x proteins were enhanced dramatically in both the luminal and glandular epithelial cells of neonatally DES-exposed uteri. In contrast, the immunostaining signal for Bcl-2 protein was decreased consistently in the epithelial cells of neonatally DES-exposed uteri. In conclusion, neonatal DES treatment induced persistent and epithelial cell-specific imbalances in the estrogen-regulated uterine expression of c-jun, c-fos, c-myc, bax, bcl-2, and bcl-x proto-oncogenes. These imbalances likely play a role in the molecular mechanism by which neonatal DES treatment induces altered estrogen responsiveness including hyperplasia, apoptosis, and ultimately neoplasia in the epithelial compartment of the hamster uterus.

[A cohort study of HIV infection among intravenous drug users in Ruili and other two counties in Yunnan Province, China, 1992-1995].

Since 1992, cohort study of prevaience of HIV infection and incidence rate among drug users has been conducted annually at the study spots along southwestern border areas of Ruili, Longchuan and Luxi counties, Yunnan province of China. From 1992 to 1995, 859, 437, 343 and 540 drug users with a total number of 2179 cases were studied respectively. The proportion of IDUs among drug users was about 30% in 1992, and went up to about 50% in 1995. HIV serloprevalence rates among IDUs in Ruili was comparatively high. From 1992 to 1995, the rates were 81.8%, 85.7%, 61.8% and 51.9% respectively. Longchuan county was at medium, 44.6%, 40.0%, 42.2% and 43.5% respectively. Luxi kept at a lower level, which were 5.1%, 0%, 0% and 3.1%. From 1992 to 1995, the increase of incidence rates were 30.1% from 21.7% to 42.1% in Ruili county, 17.2% (12.1% to 25.9%) in Long Chuan, and 0.6% (0% to 2.3%) in Luxi. The sero-prevalence rate among the spouses of these HIV infected persons was 3.1% in 1990, but increased to 9.8%, 7.4%, 0% and 10.3% respectively from 1992 to 1995 and was 12.3% in 1997. CD4 counting was conducted in 107 cases. The result indicated at the earlier of the year being infected. The higher proportion of CD4 count which was < 500/mm3 on NASBA was applied to analyse the viral load, 3 out of 29 cases were < 1,000 copies/ml, while Log mean of viral load in 26 cases was 4.37.

[Histological change of keratinocyte in full-thickness skin autograft and its effect on hyperpigmentation of the graft].

To understand the reason of hyperpigmentation following full-thickness skin autograft, 54 guinea pigs were selected as skin autograft models. The changes of density of epidermal keratinocyte (KC) and the amount and distribution of melanin in the epidermal KC were observed by histological, histochemical and autoradiographic techniques. The results showed: (1) The histological changes of KC were in fact a wound-repair process; (2) The increased melanin in KC coordinated with "multi-overlapping screen" effect, a result of multiple layers of "supranuclear cap", was the direct cause of hyperpigmentation of the graft and (3) The hydrolytic disturbance of melanin in KC and the longer life span of the KC were probably the important reasons for the increase of melanin in the epidermis.

[Histological study on the using of autogenous costal perichondrium graft to repair the cartilage of condylar process of mandible].

In order to observe the histological changes of the autogenous perichondrium graft from rib in the repair of injured articular cartilage of the condylar process of mandible, 50 rabbits were used, in which 15 were served as control. The articular cartilage with its subchondral bone were resected and an autogenous graft of costal perichondrium was sutured onto the raw surface of the condylar process, and in the controls, only the articular portion of the condylar process was resected without the application of autogenous costal perichondrium graft. The morphological changes of the newly formed cartilage during the process of its development were investigated by hiostological and autoradiog aphic techniques. The result revealed that 10 days after operation, the graft had increased in thickness and was richly populated form the proliferation of mesenchyme-like cells. Twenty to thirty days later, the chondrocytes were matured and the newly formed cartilage had covered the bony surface of mandibular condyle. At 60 days, the newly formed cartilagenous joint surface became glossy, and the morphology and arrangement of cells tended to be regular simulating the morphology of normal articular cartilage. From the experiment, it could be concluded that (1) The autogenous perichondrium graft placed on the condylar surface of mandible could form new articular cartilage which was similar in tissue morphology to the normal condylar cartilage. (2) The process of development of newly formed cartilage was similar to that of the normal cartilage. (3) The motion and loading on the joint could promote the formation of new cartilage and undergo biological reformation, gradually resulting in normal joint morphology. On this basis, the clinical application of autogenous perichondrium graft to repair injured cartilage of the condylar process of the mandible was feasible.

Detection of HIV type 1 env subtypes A, B, C, and E in Asia using dried blood spots: a new surveillance tool for molecular epidemiology.

Global surveillance of HIV-1 subtypes for genetic characterization is hampered by the biohazard of processing and the difficulties of shipping whole blood or cells from many developing country regions. We developed a technique for the direct automated sequencing of viral DNA from dried blood spot (DBS) specimens collected on absorbent paper, which can be mailed unrefrigerated in sturdy paper envelopes with low biohazard risk. DBS were collected nonrandomly from HIV-1-infected, mostly asymptomatic, patients in five Asian countries in 1991, and shipped via airmail or hand carried without refrigeration to Bangkok, and then transshipped to North America for processing. After more than 2 years of storage, including 6 months at ambient temperatures, proviral DNA in the DBS was amplified by nested PCR, and a 389-nucleotide segment of the C2-V3 env gene region was sequenced, from which 287 base pairs were aligned and subtyped by phylogenetic analysis with neighbor-joining and other methods. From southern India, there were 25 infections with subtype C and 2 with subtype A. From Myanmar (Burma), we identified the first subtype E infection, as well as six subtype BB, a distinct cluster within subtype B that was first discovered in Thailand and that has now appeared in China, Malaysia, and Japan. From southwest China, one BB was identified, while a "classical" B typical of North American and European strains was found in Indonesia. From Thailand, five DBS of ambiguous serotype were identified as three B, one BB, and one E. A blinded control serotype E specimen was correctly identified, but a serotype BB control was not tested. Most HIV-1 in southern India appears to be env subtype C, with rare A, as others have reported in western and northern India. The subtypes BB and E in Myanmar, and the BB in China, suggest epidemiological linkage with these subtypes in neighboring Thailand. DBS are a practical, economical technique for conducting large-scale molecular epidemiological surveillance to track the global distribution and spread of HIV-1 variants.

Bleomycin-induced pulmonary fibrosis in transgenic mice that either lack or overexpress the murine plasminogen activator inhibitor-1 gene.

Impaired fibrinolytic activity within the lung is a common manifestation of acute and chronic inflammatory lung diseases. Because the fibrinolytic system is active during repair processes that restore injured tissues to normal, reduced fibrinolytic activity may contribute to the subsequent development of pulmonary fibrosis. To examine the relationship between the fibrinolytic system and pulmonary fibrosis, lung inflammation was induced by bleomycin in transgenic mice that either overexpressed or were completely deficient in murine plasminogen activator inhibitor-1 (PAI-1). 2 wk after 0.075 U of bleomycin, the lungs of transgenic mice overexpressing PAI-1 contained significantly more hydroxyproline (118 +/- 8 micrograms) than littermate controls (70.5 +/- 8 micrograms, P < 0.005). 3 wk after administration of a higher dose of bleomycin (0.15 U), the lung hydroxyproline content of mice completely deficient in PAI-1 (49 +/- 8 micrograms) was not significantly different (P = 0.63) than that of control animals receiving saline (37 +/- 1 micrograms), while hydroxyproline content was significantly increased in heterozygote (77 +/- 12 micrograms, P = 0.06) and wild-type (124 +/- 19 micrograms, P < 0.001) littermates. These data demonstrate a direct correlation between the genetically determined level of PAI-1 expression and the extent of collagen accumulation that follows inflammatory lung injury. These results strongly support the hypothesis that alterations in fibrinolytic activity influence the extent of pulmonary fibrosis that occurs after inflammatory injury.

Protein C inhibitor (PCI).

PCI is a non-specific serpin that inhibits several proteases of the coagulation and fibrinolytic systems as well as plasma- and tissue kallikreins and the sperm protease acrosin. The precise physiological role of PCI has not been defined yet. Heparin stimulates most PCI/protease reactions, but interferes with the tissue kallikrein/PCI-interaction. Thereby heparin not only regulates PCI-activity but also its specificity in systems containing two or more of its target proteases. This effect is not restricted to heparin, but is also seen with other glycosaminoglycans (GAGs) and large, negatively charged molecules. PCI also binds to GAGs present on the surface of epithelial kidney cells, and GAGs isolated from these cells have a similar effect on PCI activity as heparin. Studies analyzing the role of PCI as an acrosin inhibitor revealed that endogenous PCI is immunocytochemically localized to disrupted acrosomal membranes of morphologically abnormal sperms, while intact sperms are negative for PCI-antigen. In a mouse in vitro fertilization model human PCI inhibited sperm/egg binding and decreased the fertilization rate. Northern blotting of human and mouse mRNA using human and mouse PCI-cDNA probes revealed that in the mouse PCI is exclusively synthesized in the genital tract (testis, seminal vesicle, ovary), while in humans PCI is additionally synthesized in many other organs (e.g., liver, pancreas, heart). Therefore PCI might regulate enzymes involved in fertilization (e.g. acrosin) in both species. Other proteases (e.g., tissue kallikrein) are possibly regulated in a species specific manner by different inhibitors.

Epitope mapping of the variable repetitive region with the MB antigen of Ureaplasma urealyticum.

One of the major surface structures of Ureaplasma urealyticum recognized by antibodies of patients during infection is the MB antigen. Previously, we showed by Western blot (immunoblot) analysis that any one of the anti-MB monoclonal antibodies (MAbs) 3B1.5, 5B1.1, and 10C6.6 could block the binding of patient antibodies to MB. Subsequent DNA sequencing revealed that a unique six-amino-acid direct tandem repeat region composed the carboxy two-thirds of this antigen. In the present study, using antibody-reactive peptide scanning of this repeat region, we demonstrated that the amino acids defining the epitopes for MAbs 3B1.5 5B1.1 and 10C6.6 are EQP, GK, and KEQPA, respectively. Peptide scanning analysis of an infected patient's serum antibody response showed that the dominant epitope was defined by the sequence PAGK. Mapping of these continuous epitopes revealed overlap between all MAb and patient polyclonal antibody binding sites, thus explaining the ability of a single MAb to apparently block all polyclonal antibody binding sites. We also show that a single amino acid difference in the sequence of the repeats of serovars 3 and 14 accounts for the lack of reactivity with serovar 14 of two of the serovar 3-specific MAbs. Finally, the data demonstrate the need to obtain the sequences of the mba genes of all serovars before an effective serovar-specific antibody detection method can be developed.

[The radiobiologic characteristics of DNA polymerase beta in hepatomas].

DNA polymerase beta activity, its content and gene transcription levels in SMMC-LTNM hepatoma were investigated, using 3H-TTP incorporation, immunocytochemistry and cytoplasmic dot hybridization, respectively. The relations between the biological properties of the enzyme and DNA repair synthesis induced by gamma-ray irradiation were also studied. It was found that DNA polymerase beta activity, its content and the amount of its mRNA were much higher in hepatoma than those in normal hepatocytes (P < 0.01). Following whole-body irradiation of the nude mouse bearing SMMC-LTNM with 2 Gy of gamma ray, the polymerase beta activity in hepatoma increased temporarily and the gene transcription of the enzyme seemed to be more active. DNA polymerase beta participated in DNA repair synthesis and this effect was different between hepatoma and hepatocyte because of the biological differences of DNA polymerase beta. The results presented here indicated that DNA polymerase beta could affect radiation damage and radiotherapy of cancer.

Expression of transforming growth factor-alpha and epidermal growth factor receptor in middle ear cholesteatoma.

To study the possible autocrine growth stimulation of cholesteatoma epithelium, the expression of transforming growth factor alpha (TGF-alpha) and epidermal growth factor receptor (EGFR) in middle ear cholesteatoma was investigated by immunohistochemical staining techniques. Twenty cholesteatoma samples obtained at operation and six normal skin specimens collected from the external ear canal were used in the study. Immunostaining for TGF-alpha showed a diffuse cytoplasmic staining pattern. It was weakly expressed in the basal layer of the normal epidermal epithelium but was more strongly expressed in all cholesteatoma specimens. EGFR showed a dot-like/diffuse cytoplasmic and cell membrane staining pattern. EGFR-positive cells were seen in the basal layer of normal epidermis. In the cholesteatoma specimens, expression of EGFR was not only confined to the basal layer but persisted into the upper layers of the epithelium. Our findings indicate that an autocrine growth stimulation by TGF-alpha and EGFR may contribute to the unrestrained growth of cholesteatoma epithelium in the middle ear cavity.

[A comparative study of benign and malignant pulmonary masses with coloured Doppler sonography].

In order to differentiate benign from malignant pulmonary masses by means of observing blood flow signal, with coloured Doppler sonography, the authors examined 67 in-patients who had been found having masses in the lungs by X-ray or CT. For comparison 43 benign cases showing blood signals in the lung was also examined. The results showed that the blood flow signals of all lung cancers were of relatively low blood flow resistance or arteriovenous shunting, while those of benign lesions were of high blood flow resistance. The pulsatility index in lung cancer was 1.02 +/- 0.30, the restance index was 0.52 +/- 0.13, the peak systolic velocity was 0.19 +/- 0.11 m/s, the end diastolic velocity was 0.06 +/- 0.02 m/s. The pulsatility index in benign pulmonary lesions was 1.53 +/- 0.30, the resitance index was 0.86 +/- 0.05, the peak systolic velocity was 0.28 +/- 0.05 m/s and the end diastolic velocity was 0.04 +/- 0.01 m/s. There were significant differences between the benign and malignant pulmonary lesions in the four indices (P < 0.01) by t test and t' test. Lower pulsatility index, lower resistance index, lower peak systolic velocity and higher end diastolic velocity were characteristics of malignancy.

[Detection of Epstein-Barr virus DNA in renal tissue from patients with interstitial nephritis].

In order to demonstrate whether Epstein-Barr virus (EBV) infection might play a role in the pathogenesis of interstitial nephritis as suggested by many scholars, EBV DNA was detected in twelve specimens of frozen renal biopsy tissue from patients with interstitial nephritis by using nest polymerase chain reaction (nest PCR). For comparison, frozen renal biopsy tissue from ten patients with minimal change disease was used as control. Southern blot hybridization was used to check the specificity of PCR product. The results showed that eight of twelve frozen renal biopsy specimens from interstitial nephritis patients were EBV DNA positive (66.7%), as compared with negative in all the ten frozen renal specimens from minimal change disease patients. The differences was statistically significant (P < 0.01). The study strongly suggests that EBV infection may play an important role in the pathogenesis of interstitial nephritis. The location of EBV in renal tissue and the mechanism inducing interstitial nephritis by EBV are to be clarified.
