[A comparison of chromosome fragile sites in elderly and young people].

In this comparative study, the authors investigated the chromosome aberrations and expression of fragile sites in the lymphocytes which were collected from 45 elderly people and 29 young people and were cultured with FUDR for 24 hours. The results showed that the aberration rate was 24.75% for elderly people, but 4.22% for young people (P < 0.01): that the expression of fragile sites was 45.94% for elderly peoples, but 23.02% for young people (P < 0.01): and that the relation of aberrations with fragile sites was 79.98% for elderly people, and 92% for young people. These findings suggest that chromosome aberration is in close association with fragile site.

[Relationships between intrahepatic metastasis of hepatocellular carcinoma and in situ microenvironment, and the abundance of nm23-H1 H-ras mRNA].

OBJECTIVE: To reveal intrahepatic metastasis of hepatocellular carcinoma (HCC) in relation to its micro-environment and nm23-H1 H-ras mRNA expression. METHODS: In HCC specimens from 25 patients, the level of nm23-H1 and H-ras mRNA was examined by quantitative reverse transcription-polymerase chain reaction(RT-PCR). Tumor microvessel density(MDV), an essential factor of microenvironment and proliferating cell nucleus antigen (PCNA), an index of tumor cell proliferation, were also studied by immunohistochemical methods using antibodies against endothelial protein factor VIII-related antigen(F8RA) and antibody PC-10. RESULTS: The MDV and PCNA index in the group with intrahepatic metastasis were remarkably higher than those in the group without it (P < 0.01), while nm23-H1 mRNA expression level was negatively correlated with PCNA index (P < 0.05). The level of H-ras mRNA expression did not correlate with intrahepatic metastasis. MDV was positively correlated with PCNA index (P < 0.01). CONCLUSION: Intrahepatic metastasis of HCC depends very much on its in situ microenvironment. The decline in nm23-H1 mRNA may implicate a preferential proliferation of nm23-negetive HCC cells with highly metastastic potential which adapt well to the microenvironment.

[Quantitation of nerve fibers in habenula perforata in chinchilla].

OBJECTIVE: To examine the number of nerve fibers in the habenula perforata of the cochlea in normal and carboplatin-treated chinchillas. METHODS: Three months following administration of carboplatin (100 mg/kg x 2), six carboplatin-treated animals, together with 6 controls, were sacrificed, prepared for semi-thin sectioning of the cochlea. The sections were stained with toluidine blue and basic fuchsin, examined under a light microscope and photographed. RESULTS: In normal chinchillas, the numbers of the nerve fibers in each habenula opening varied across the cochlea, with 93.7 +/- 12.2(mean +/- s) fibers at the hook, 181.5 +/- 30.4 at the upper of basal turn, 129.7 +/- 27.8 at the second turn and 75.8 +/- 13.2 at the apical turn. In cochlear devoid of type I ganglion cells and inner hair cells as a result of carboplatin treatment, only one-seventh of the fiber population remained. CONCLUSION: Approximately six-seventh of the nerve fibers in the habenula are type I afferent axons that innervate the inner hair cells. The remaining fibers are type II afferents and surviving efferents.

[A new method for evaluating malaria surveillance effectiveness--establishment and application of an index system].

AIM: To establish a comprehensive index system which is suitable for evaluating the effectiveness of malaria surveillance schemes. METHODS: Delphi method was used to screen the existed effectiveness evaluation indices by experts to determine key indices and weighted coefficients. RESULTS: Three indices, i.e. autochthonous incidence, ratio of cases detected (ROCD) and ratio of introduced/imported cases were selected. Their weighted coefficients were 0.36, 0.31 and 0.33, respectively. An index system named the malaria surveillance effectiveness index (MSEI) was built up and formulated as [formula: see text] which was applied to study cost-effectiveness of the two malaria surveillance schemes (A and B). According to the formula, the MSEIs in the study areas using the schemes A and B were 48.56 and 45.93, respectively. However, the cost for increasing a score of MSEI by implementing the scheme A was four times as high as that by implementing the scheme B. CONCLUSION: The MSEI is practicable for evaluating the overall effectiveness of malaria surveillance schemes, especially in the cost-effectiveness analysis and health-economic evaluation.

[An optical model of axial reflection and its clinical application].

A developed optical model of axial reflection on retinal blood vessels has been presented in this paper. The loss of the transparency of retinal arterial vessel wall is a frequent expression of retinal vascular pathology. With the change of parameters, the model can simulate the progress of the axial reflection exactly during the onset and progression of hypertension. This non-invasive and quantitative method is useful for study in the relationship between hypertension and retinal vessel axial reflection. It can help doctors approach a diagnosis of hypertension.

[A robust method for automated retinal vascular images mosaic based on phase correlation method and mathematical morphology].

Precise mosaic techniques are essential for quantitative evaluation of retinal images to make early detection of fundus anomalies feasible. Opening of a gray-scale image by a gray-scale structuring element(SE) can generate a background image. Image mosaic was achieved by subtraction this background image from the original image and then applying a phase correlation method to find translation difference. Because of the accuracy characteristics of the phase correlation method and the speed of the FFT hardware, this new algorithm can work very fast and accurately, compared to conventional techniques. The method was also characterized by an outstanding robustness against correlated noise and disturbances, such as those encountered with nonuniform illumination. The results confirm the robustness of the chosen approach.

[A quantitative measurement of retinal nerve fiber layer thickness by optical coherence tomography in normal Chinese people].

PURPOSE: To measure retinal nerve fiber layer(RNFL) thickness in normal Chinese people for the use of early diagnosis in glaucomatous patients. METHODS: Optical Coherence Tomography(OCT) was used in 111 eyes of 77 Subjects to create cylindrical cross sections of the RNFL around the optic nerve head with scan diameter of 3.46 mm. The differences in RNFL thickness were analyzed with age, sex, either eye. RESULTS: We found a decrease in RNFL thickness with aging especially in the superior quadrant(P < 0.05), but there weren't significant difference between right and left eyes, neither the sex(P > 0.05). The Mean RNFL thickness of different ages(mean +/- s) was as following: the superior quadrant 128.00 +/- 10.31-149.00 +/- 13.17, the inferior quadrant 132.95 +/- 9.54-142.33 +/- 10.60, the overall mean RNFL thickness 109.00 +/- 4.72-114.28 +/- 7.98 microns. CONCLUSION: The preliminary study showed that neither sex nor either eye were associated with significant differences in RNFL thickness. It is necessary to define the normative value by age group on a larger scale.

Functional significance of cysteine residues in the delta opioid receptor studied by site-directed mutagenesis.

Previous work suggested that sulfhydryl groups and disulfide bridges have important functions in opioid binding to the delta opioid receptor. The question regarding which cysteines are essential for ligand binding was approached by replacement of cysteine residues in the cloned delta opioid receptor using site-directed mutagenesis. The wild-type and mutant receptors were expressed stably in Chinese hamster ovary cells. The two extracellular cysteine residues and the six located in transmembrane domains were mutated to serine or alanine, one at a time. Replacement of either of the extracellular cysteines produced a receptor devoid of delta agonist and antagonist binding activity. Immunofluorescence cytochemistry, performed with anti delta opioid receptor antibodies in washed cell monolayers in one of these mutants (Cys-Ser121), and immunoblots, performed on cell extracts, indicate that the receptor was expressed and seems to be associated with the cell membrane. The existence of an essential extracellular disulfide bridge, previously postulated by analogy to other G protein coupled receptors, is strongly supported by our results. Replacement of any one of the six transmembrane cysteines was virtually without effect on the ability of the receptor to bind delta agonists and antagonists. Since there is strong evidence that the transmembrane domains are involved in ligand binding, these results suggest that the cysteine residues, even those near or at the binding site, are not essential for receptor binding. Furthermore, these results support the idea that the striking effects of sulfhydryl reagents on ligand binding of opioid receptors are likely to be due to steric hindrance by the large moieties transferred to the sulfhydryl groups of cysteine residues by these reagents.

Bovine proenteropeptidase is activated by trypsin, and the specificity of enteropeptidase depends on the heavy chain.

Enteropeptidase, also known as enterokinase, initiates the activation of pancreatic hydrolases by cleaving and activating trypsinogen. Enteropeptidase is synthesized as a single-chain protein, whereas purified enteropeptidase contains a approximately 47-kDa serine protease domain (light chain) and a disulfide-linked approximately 120-kDa heavy chain. The heavy chain contains an amino-terminal membrane-spanning segment and several repeated structural motifs of unknown function. To study the role of heavy chain motifs in substrate recognition, secreted variants of recombinant bovine proenteropeptidase were constructed by replacing the transmembrane domain with a signal peptide. Secreted variants containing both the heavy chain (minus the transmembrane domain) and the catalytic light chain (pro-HL-BEK (where BEK is bovine enteropeptidase)) or only the catalytic domain (pro-L-BEK) were expressed in baby hamster kidney cells and purified. Single-chain pro-HL-BEK and pro-L-BEK were zymogens with extremely low catalytic activity, and both were activated readily by trypsin cleavage. Trypsinogen was activated efficiently by purified enteropeptidase from bovine intestine (Km = 5.6 microM and kcat = 4.0 s-1) and by HL-BEK (Km = 5.6 microM and kcat = 2.2 s-1), but not by L-BEK (Km = 133 microM and kcat = 0.1 s-1); HL-BEK cleaved trypsinogen at pH 5.6 with 520-fold greater catalytic efficiency than did L-BEK. Qualitatively similar results were obtained at pH 8.4. In contrast to this striking difference in trypsinogen recognition, the small synthetic substrate Gly-Asp-Asp-Asp-Asp-Lys-beta-naphthylamide was cleaved with similar kinetic parameters by both HL-BEK (Km = 0.27 mM and kcat = 0.07 s-1) and L-BEK (Km = 0.60 mM and kcat = 0.06 s-1). The presence of the heavy chain also influenced the rate of reaction with protease inhibitors. Bovine pancreatic trypsin inhibitor preferred HL-BEK (initial Ki = 99 nM and final Ki* = 1.8 nM) over L-BEK (Ki = 698 nM and Ki* = 6.2 nM). Soybean trypsin inhibitor exhibited a reciprocal pattern, inhibiting L-BEK (Ki* = 1.6 nM), but not HL-BEK. These kinetic data indicate that the enteropeptidase heavy chain has little influence on the recognition of small peptides, but strongly influences macromolecular substrate recognition and inhibitor specificity.

Ca2+ influx amplifies protein kinase C potentiation of recombinant NMDA receptors.

Protein kinase C (PKC) potentiates NMDA receptors in hippocampal, trigeminal, and spinal neurons. Although PKC phosphorylates the NMDA receptor subunit NR1 at four residues within the C terminal splice cassette C1, the molecular mechanisms underlying PKC potentiation of NMDA responses are not yet known. The present study examined the role of Ca2+ in PKC potentiation of recombinant NMDA receptors expressed in Xenopus oocytes. We found that Ca2+ influx through PKC-potentiated NMDA receptors can further increase the NMDA response ("Ca2+ amplification"). Ca2+ amplification required a rise in intracellular Ca2+ concentration at or near the intracellular end of the channel and was independent of Ca2+-activated Cl- current. Ca2+ amplification depended on extracellular Ca2+ concentration during NMDA application and not during PKC activation. Ca2+ amplification was reduced by the membrane-permeant Ca2+-chelating agent BAPTA-AM. Mutant receptors with greatly reduced Ca2+ permeability did not exhibit Ca2+ amplification. Receptors containing the NR1 N-terminal splice cassette showed more Ca2+ amplification, possibly because of their larger basal current and therefore greater Ca2+ influx. Contrary to expectation, splicing out the two C-terminal splice cassettes of NR1 enhanced PKC potentiation in a manner independent of extracellular Ca2+. This observation indicates that PKC potentiation does not require phosphorylation of the C1 cassette of the NR1 subunit. PKC potentiation of NMDA receptors in vivo is likely to be affected by Ca2+ amplification of the potentiated signal; the degree of amplification will depend in part on alternative splicing of the NR1 subunit, which is regulated developmentally and in a cell-specific manner.
