[Cytophotometric investigations of the nuclear DNA content in ependymomas and plexuspapillomas (author's transl)].

Report on Feulgen-cytophotometric DNA investigations in 10 ependymomas and 2 plexuspapillomas. All ependymomas represent in their karyograms aneuploidic stem lines. In most of them one observes duplication peaks (G2 cells) as expression of proliferation behaviour. The histological benign ependymomas exhibit stem lines between the hyperdiploid and hypotetraploid values. With increasing malignancy the stem lines are elevated in higher ploidy levels and a reduction of a predominant stem line is recognizable. One of the ependymomas represents a distribution of DNA values like in a "mosaic" tumour. The measurements in the plexuspapillomas in good conformity with the histologic picture reflect the DNA distribution of a benign euploid neoplasm.

[The effect of treatment on circulating immune complexes in rheumatoid arthritis (author's transl)].

Immune complexes were demonstrated by leucocyte phagocytosis assay in 37 of 55 patients with rheumatoid arthritis. After treatment with gold salts, penicillamine, alpha-mercaptopropionylglycine, immunosuppressive agents or levamisole a substantial fall or elimination of demonstrable circulating immune complexes occurred. In-vitro studies revealed that gold salts in therapeutic concentrations inhibited the phagocytosis of immune complexes, while other drugs - penicillamine, levamisole and all anti-inflammatory agents including corticosteroids - lack this effect. The reduction in circulating immune complexes under basic therapy may be due to a beneficial effect of these agents on the disease process itself. This test may, therefore, be of value in assessing the efficacy of drug treatment.

Extracerebrospinal metastases in glioblastoma. Case report and review of the literature.

A malignant glioblastoma adherent to the dura mater was removed from the parieto-occipital lobe in a 12-year-old boy. The site of the tumor was subsequently irridiated by 4000 rads of Cobalt-60. Five months later the boy was readmitted complaining of pains in the pelvis an in both thighs. X-ray examination of the pelvis demonstrated multiple metastases. Investigation of bone marrow revealed replacement of normal haematopoiesis by a tumor cell population histologically identical to that of the brain tumor. Reviewing the literature 58 reports on glioblastomas with extracerebrospinal metastases could be found. Metastases were preferably localized in cervical or mediastinal lymph nodes, lungs, bones, liver, dura mater, and operative flap. It is suggested that extracerebrospinal metastases occur most frequently after the tumor has infiltrated the cranium and extracranial soft tissues. In the case reported here it is speculated that the tumor spread to extraneural tissues after invading the dural veins. The possible occurrence of extracerebrospinal metastases in glioblastoma emphasizes the necessity of additional chemotherapy.

Supratentorial recurrences of gliomas. Morphological studies in relation to time intervals with astrocytomas.

We report 137 recurrent supratentorial astrocytomas. The primary tumours diagnosed on the basis of a grading system with three stages were 72 astrocytomas I and 65 astrocytomas II. In the first group 14% of the recurrences were not changed, 55.5% became astrocytomas II, and 30.5% became glioblastomas. In the second group 55.4% were unchanged, and 44.6% became glioblastomas. The postoperative intervals untile reintervention or death were statistically examined. It seems that the recurrence time chielfy depends on the nature of the primary tumour. The transformation of an astrocytoma I to a glioblastoma takes longer than the transformation of an astrocytoma II into a glioblastoma. In about two thirds of all astrocytomas an increase of malignancy is to be expected. From the histological picture it is not possible in an individual case to predict the likelihood or speed of malignant change. With regard to the effect of irradiation the authors conclude that radiotherapy most probably does not produce malignancy.

Supratentorial recurrences of gliomas. Morphological studies in relation to time intervals with oligodendrogliomas.

On the basis of a three stage grading system we report 23 stage one recurrent oligodendrogliomas (O 1), and 29 stage two recurrent oligodendrogliomas (O 2). In the O 1 group after the first interval 15 became O 2 and 2 became glioblastomas. Twenty tumours of the O 2 group after the first interval were not changed, three became oligodendroglioma-astrocytomas stage 2, and six became glioblastomas. The time relation for the recurrent phase in the primary O 1 group is calculated as 42 months, and in the primary O 2 group as 22 months, but this is without significance. For the development of malignancy, especially for the change to glioblastoma, a prominent participation by transformed local astrocytes seems to be essential. Postoperative irradiation most probably does not favour malignant change. A prolongation of the expectation of life by radiotherapy is not noticed.

Spontaneous rupture of the gastrocnemius muscle.

Nineteen ruptures of the medial head of the gastrocnemius muslce are described. These were caused by sudden overstretching of the muscle though concomitant ankle dorsiflexion and knee extension. Surgical exploration in 12 patients verified rupture of the musculotendinous junction. A 2-year follow-up showed that the surgical approach with suture of the muscle gave better results than conservative treatment, especially in the younger and athletic patients.

Biotransformation of diethylstilbestrol in the rhesus monkey and the chimpanzee.

Diethylstilbestrol (DES) is considered a teratogen and a transplacental carcinogen in humans. In order to compare its biotransformation in nonhuman primates to that in humans, a metabolic study was carried out in rhesus monkeys and chimpanzees. After an oral dose of 1 mg/kg [14C]DES, approximately 59% of the ingested radioactivity was found in the urine and 28% in the feces of two female rhesus monkeys after 4 days, while in male rhesus monkeys urinary radioactivity accounted for 43% and fecal radioactivity for 35%. In chimpanzees, 63% of a 0.5 mg/kg dose was excreted with the urine in a female and 47% in a male animal. In both species, urinary radioactivity was predominantly (greater than 70%) associated with glucuronides. Besides DES, three metabolites were found in the urinary glucuronide fraction of rhesus monkeys and chimpanzees by radio gas chromatography and were identified as dienestrol and omega-hydroxy derivatives of DES and dienestrol. Fecal radioactivity in rhesus monkeys was shown to consist exclusively of DES. Glucuronidation of DES appears to occur in neonatal and fetal rhesus monkeys.

[Electron transport particles from beef-heart mitochondria as a test system for inhibitors of the respiratory chain].

Respiratory inhibitors exert a potentially biocidic action on all aerobic organisms. Therefore the systematic search for them and the elucidation of their mechanism of action is of great importance. The non-phosphorylating electron transfer particles (ETP) according to Crane et al. are a suitable test object for this aim. A standardized preparation of ETP from beef heart is described and characterized enzymatically. The ETP show a variety of advantages in comparison with other systems; their field of application is broad. Owing to the absence of bypasses of the respiration, particles from animals are more suitable than those from plants for the search for fungicides and herbicides which inhibit the respiratory chain. With relatively little expense, the two standard assay systems proposed (NADH oxidase and succinate-cytochrome c oxidoreductase) pick up with certainty all inhibitors of the main pathway of the respiratory chain; they are useful for screening studies. The system proposed is also suitable for the analysis of the exact site and the mechanism of action of respiratory inhibitors. Furthermore, ETP can be used to obtain parameters of the hydrophobicity of drugs as well as for toxicological studies.

[Mechanism of action of the inhibition of pyridine-nucleotide-dependent flavine enzymes using the systemic fungicide Dexon].

The actions of Dexon on the NADH-ferricyanide oxidoreductase and the NADPH oxidase system of electron transfer particles (ETP) from beef heart as well as on the NADPH-cytochrome c oxidoreductase from brewer's yeast (Saccharomyces carlsbergensis Hansen) were investigated. The inhibition of the NADH dehydrogenase activity of ETP and that of the yeast enzyme correspond with respect to the following characteristics: 1) increase in the inhibition, 2) enhancement of the Dexon sensitivity by one order of magnitude after preincubation in the presence of NAD(P)H, 3) irreversibility of the inhibition, 4) no detectable changes in the spectral properties and in coenzyme activity of FMN after acid extraction from Dexon-treated enzyme. The inhibition of the NADH dehydrogenase activity of ETP is diminished by both NAD+ and FMN. However, no interaction of Dexon with NAD(P)H or FMN could be detected in the absence of enzyme or apoenzyme. The concentration of half-inhibition by Dexon for the yeast enzyme corresponds with its FMN concentration. It is proposed that both apoenzyme, NAD(P)H and FMN are involved in the interaction with Dexon. Possible mechanisms of binding are both complanar complexations of the ring systems and a triazene formation between FMNH2 and Dexon. The NADPH oxidase activity of the ETP is partly inhibited; the share inhibited by Dexon may represent the pathway via the transhydrogenase reaction.
