Determination of plasma mepindolol levels by high-performance liquid chromatography and electrochemical detection.

A method for determining the plasma concentration of the beta-receptor blocking agent mepindolol by high-performance liquid column chromatography (paired-ion chromatography) and electrochemical detection is described. Pindolol is used as an internal standard and the detection limit after extraction of 1 ml of plasma is less than 1 ng of mepindolol. Reproducible results can be obtained with relative standard deviations from replicate analyses of 5 ng/ml plasma samples within +/- 4.7%. The method is also suitable for quantification of plasma pindolol levels with mepindolol as an internal standard.

Quantitative culture of ureaplasma urealyticum in patients with chronic prostatitis or prostatosis.

We studied 187 men with symptoms and signs of chronic prostatitis or prostatosis and 108 healthy controls. Ureaplasma urealyticum was isolated from 103 (55.1 per cent) of the patients and from 24 (22.2 per cent) of the controls. When quantitative culture of Ureaplasma urealyticum is considered a typical histogram of prostatitis was evident in 36 patients (19.3 per cent) using the Stamey localization technique. In 20 of the 36 patients the etiology was uncertain for various reasons. Evidence was provided that Ureaplasma urealyticum could be considered as the etiologic agent of the disease in at least 16 (8.6 per cent) of the 187 patients with chronic prostatitis studied.

The effect of intravenously administered human chorionic gonadotrophin on plasma testosterone in patients with sexual impotence and oligozoospermia.

The intravenous administration of human chorionic gonadotrophin (hCG) leads to a significant increase of plasma testosterone in normal men after two h (from 5.85 to 8.35 ng/ml). The rise of plasma testostosterone occurs also in men with idiopathic oligozoospermia (5.81 to 6.84 ng/ml) but it is absent in men with sexual impotence (5.30 resp. 5.37 ng/ml). These data refer to men aged between 20 and 30 years. It is concluded, that the response to hCG may be of diagnostic help in sexual impotence and in oligozoospermia, especially in respect to a therapeutic approach.

Pharmacokinetics and pharmacodynamics of mepindolol sulphate.

Five clinically healthy, male volunteers were each given an intravenous dose of 1.19 mg of 2-methyl-14C-mepindolol sulphate, followed one week later by an oral dose of 20 mg. In addition, the effect of mepindolol sulphate was studied in 3 male test subjects after repeated oral administration for 8 days with a daily dose of 2 x 2.5 mg using the heart rate behaviour model during submaximum ergometer exercise. Synchronously with pharmacodynamics, pharmacokinetic data were also obtained after multiple dosing.

[Endocrine changes in gestagen treatment in a patient with scleroderma diffusa].

In a male patient, treated with gestagens and corticosteroids for diffuse scleroderma, endocrine alterations over two years are followed. A decrease in gonadotrophin plasma levels and an increase of prolactin is observed. Levels of testosterone, DHEA, and 17-alpha-hydroxyprogesterone are lowered, while those of estradiol and androstenedione remain unchanged. The alterations may be explained by a suppression of pituitary function and following decrease of testicular (and adrenal) steroid secretion.

[Morbidity studies, following continuous electronic and biochemical monitoring of 1,000 childbirths (author's transl)].

A group of 1,000 electronically and biochemically monitored children of high-risk deliveries was examined for morbidity up to the age of one year.--Transfer to the Paediatric Hospital of Jena University had been necessary for 147 newborns for low weight and/or impaired adaptation. Sixteen of 18 neurologically conspicuous children, which had temporarily displayed electro-encephalographic or echo-encephalographic pathological or marginal findings, did no longer exhibit any neurological deficiency or signs of impaired intelligence, when re-examined after ten months. Only two children had severe disorders in terms of infantile cerebral paresis, one of them with a genetically determined disease (Morbus Langdon Down).--Incidence of infantile cerebral paresis dropped from more than three to something between one and two per cent owing the antepartum, intrapartum, and postpartum intensive monitoring as well as to intensive neonatal care. It is assumed that this will act to decrease the number of children with mental retardation.

[Seasonal variations of testicular and pituitary function in men].

In 21 young, healthy men typical parameters of pituitary and testicular function were recorded over a period of one year in intervals of two months. The mean sperm count in semen had a minimum in december, a maximum in june. Serum-FSH showed low levels in june, october and december, a peak in august. Levels of FSH following stimulation by LH-RH varied in the same manner, but the response to LH-RH was lower in april. A peak of serum-LH appeared in april, other values being rather constant. The response to LH-RH showed parallel variations. Testosterone levels exhibited a significant peak in june. The mean increase of testosterone following hCG depended on basal levels, but showed a distinct peak in december.

[Effect of multivalent cations on the enzymatic determination of glucose with glucose oxidase].

Iron and copper ions, in a concentration greater than 10(-4) M, inhibit the indicator reaction of a glucose oxidase-peroxidase reagent for the enzymatic determination of glucose, when weakly complexing buffers or buffer-free reaction media are used. The addition of EDTA and other complexing agents or, time-dependently, the buffer ions themselves reverse the inhibition to a great extent. The discussed mechanism of inhibition is based on the assumption that the metal ions share in the re-oxidation of the co-enzyme of glucose oxidase.

Behavioral and biochemical effects of chronic consumption of ethanol by hamsters.

Male Syrian Golden-Hamsters consumed about 90% of their fluid as an ethanol-water solution when given a choice between water and ethanol-water solutions. This resulted in a daily alcohol intake of about 11 g/kg. After 53 days of alcohol ingestion there was no evidence of tolerance to the soporific effects of parenterally administered ethanol and removal of the ethanol solutions failed to produce any signs of alcohol withdrawal. However, after cessation of ethanol ingestion, the acquisition of escape responding from footshock was impaired in the alcohol group as compared to a control group. Biochemical and ultrastructural analyses demonstrated that chronic ethanol ingestion hepatic glycogen and plasma triglycerides without altering either the hepatic or plasma concentrations of phospholipids, free cholesterol or cholesterol esters. These results demonstrate that the hamster has an unusual metabolic response to ethanol. These findings are contrasted to the biochemical and behavioral effects of the drug in other species.

[Bacteriaemia during parturition (author's transl)].

Bacteriological tests were made of 606 blood cultures which had been obtained from 64 women in labour. Positive findings were recorded from 8.1 per cent, whereas another positive percentage established before delivery amounted to 4.7 only. The percentage of bacteriaemia findings was not substantively increased following electronic monitoring of childbirth. Beta-haemolysing streptococci were found to grow in 7.8 per cent of vaginal smear cultures taken from women in labour and in six per cent of cultures taken from newborns. Amniotic fluid was also bacteriologically tested during labour.

[The maternal and perinatal mortality and morbitity in attendance to Caesarean section in the period from 1956--1976 at the UFK-Jena (author's transl)].

In the period from 1956--76 1 468 caesarean sections were accomplished at the UFK-Jena. 44 043 children were delivery during the 21-year period. The number of caesarean sections rose from 1,08% to 1956 to 10,8% in 1976. The fetal indication in 1956 was zero, on the other side it amounts to 34,3% in 1976. First place of the maternal indication in the 21-year period took in the cephalo pelvine disproportion. In the same time the maternal mortality in attendance of caesarean section was 12,3%. Considering the period between 1969--76, the maternal mortality fell to 2%. Maternal morbidity diminished from 1965 and obtained 23% in the period between 1974/1976. The feverish childbed took in the first place of the maternal morbidity (30%). The perinatal mortality fall from 28,57% in the period between 1956/1958 to 3,71% in the period between 1974/1976. The perinatal morbidity we could'nt state certainly. In 8,3% we found perinatal morbidity by acidosis. 5,5% of children, delivered by caesarean section needed medical care. 16,4% of all children were either small for gestational age or immature. The problem of caesarean section as an important method for termination of labor is discussed.

[Perinatal mortality and morbidity for childbirths from breech presentation at Gynaecological Hospital of Jena University, between 1966 and 1976/(77) (author's transl)].

Studies were conducted at the Gynaecological Hospital of Jena University, between 1966 and 1976, into perinatal mortality and morbidity, in the context of 950 childbirths from breech presentation. Unambiguous decline in uncleared perinatal mortality was characterised by a numerical drop from 11.7. per cent in 1966/67 to 6.3 per cent in 1976/77.-Cleared perinatal mortality went down, similarly, for the same period of time, from 6.1 per cent to 2.5 per cent.-Uncleared perinatal mortality in the context of caesarean section was 0.65 per cent, while that related to vaginal delivery amounted to ten per cent. The cleared figures (after correction) were 0.54 and 5.4 per cent, the difference having been statistically secured with high significance. A decisive role was played, in that context, by the introduction of selective section as of 1975. Intranatal mortality has ever since dropped to zero, as compared to 1.9 per cent between 1966 and 1974.-Acidosis morbidity due to breech presentation was three times as high as that associated with cephalic delivery. The Apgar score was zero in 6.4 per cent of all children born by vaginal delivery. Therapeutic consequences are discussed.
