Characterization of Peroxisomes from the Alga Bumilleriopsis filiformis.

The Xanthophycean alga Bumilleriopsis filiformis possesses peroxisomes which on electron micrographs show a mostly spherical or ovoid shape with a diameter in the range of 0.3 micrometer. Their granular matrix is usually of moderate electron density and in a very few cases contains amorphous inclusions. No associations with other organelles could be observed.During separation in a sucrose gradient, the peroxisomes from Bumilleriopsis equilibrate at a density of 1.22 grams per cubic centimeter. Glycolate oxidase and glyoxylate-glutamate aminotransferase were found in the isolated organelles along with catalase and uricase. However, no further leaf peroxisomal enzymes were detected. This is the first time that an alga of the group of Xanthophyceae has been demonstrated to possess a glycolate oxidase.The organelles from Bumilleriopsis differ from leaf peroxisomes also by the absence of enzymes of the beta-oxidation pathway. All enzymes for the degradation of fatty acids which were tested are located solely in the mitochondria.

[Manometry studies in the evaluation of anal continence].

In a complex measuring program performing anorectal perfusion manometry a great number of parameters were determined in incontinent patients, patients after continence improving operations and in healthy persons. In this way, we were able to select 20 parameters in which healthy persons differ from incontinent patients, the difference occurring with a security of 99.0 to 99.9%. Our investigations were carried out with test persons at rest, after distension of the rectal ampulla, during abdominal straining and coughing. Rectoanal resting profiles and stress profiles were recorded and the measuring values obtained compared with an anamnestic score.

Diclofenac sodium: blood concentration of the slow-release form and influence on the metabolism of kallikrein.

The blood concentration of the slow-release formulation of diclofenac sodium (Voltaren) has been determined in 10 patients at various intervals over a period of 22 days. In parallel, the influence of diclofenac on the kallikrein-kinin system was investigated since important interactions between the prostaglandins and kallikrein are known. The peak concentration of the slow-release formulation was significantly decreased compared with data of the standard formulation from other studies and the peak was observed later. The areas under the concentration curves were, however, similar. No signs of drug cumulation were observed during the treatment. The kallikrein concentration decreased after one week in plasma and after two weeks in urine. The plasma concentration of kallikrein reaches control level again after two weeks. Its urine excretion was increased to control levels after the third week. These changes in the kallikrein-kinin system did not reach the level of significance. It is concluded that diclofenac sodium has only a short-term influence on the kallikrein-kinin system.

[Circulating immune complexes and NK activity in systemic lupus erythematosus].

NK-activity of normal mononuclear cells is inhibited in vitro by addition of immune complexes or aggregated IgG. We therefore studied 20 sera of patients with systemic lupus erythematosus in order to investigate whether there is any correlation between the presence of circulating immune complexes and suppression of NK-activity. A correlation was not found.

[Therapy of activated gonarthrosis with etofenamate cream (Rheumon cream)].

In a multicenter single-blind study on 60 patients with unilateral gonarthrosis the efficacy and tolerance of etofenamate cream were compared to a commonly used ointment containing salicylic acid and other ingredients. With respect to therapeutic success etofenamate was superior to the control medication. Mobility and circumference of the knee joint as well as pain parameters improved more rapidly in the etofenamate group, whereby the difference between both groups frequently was statistically significant. With the control medication therapy failed in three cases, with etofenamate cream clinical symptoms improved in all patients. With the exception of a transient local reddening of the skin in one patient of the etofenamate group no adverse drug reactions were observed.

Quantification of human serum lipoprotein Lp(a): zone immunoelectrophoresis assay, a new sensitive method as compared to electroimmuno assay.

This study aimed at improving the immunological procedures for the quantification of human serum lipoprotein Lp(a). Lipoprotein Lp(a) was estimated by two immunoelectrophoretic methods: electroimmunoassay and zone immunoelectrophoresis assay. Whereas the electroimmunoassay gave curvilinear calibration lines (in peak height versus concentration plots) the zone immunoelectrophoresis assay revealed linearity in the same concentration range. The results obtained are in good agreement with each other (r = 0.975). The inter-assay coefficient of variation for the zone immunoelectrophoresis assay was 12% as determined by the use of a lyophilised reference serum. Zone immunoelectrophoresis assay is recommended for the routine quantification of lipoprotein Lp(a). Although the levels for total cholesterol (p less than 0.0025), HDL-cholesterol (p less than 0.001) and triglycerides (p less than 0.025) differed significantly between male (n = 24) and female (n = 22) individuals, no difference could be detected comparing the frequency distributions of Lp(a)-levels found for the male and the female group.

Determination of the urethral stress profile with the aid of perfused catheters.

To make manometric studies of urethral function and obtain exact recordings of the so-called stress profile requires a pressure rise rate which exceeds that of the pressure complex due to coughing. The syringe perfusion pump commonly used in urodynamics limits the pressure rise rate. The rate ist also reduced by catheters with high compliance. A suitable recording accuracy necessitates values greater than 150 kPa/s. This can be achieved with the aid of an improved pneumatic-hydraulic infusion system and low-compliance catheters. We describe the improvement of a recording system and demonstrate the importance of a suitable pressure rise rate.

Separation of apolipoproteins of human very low density lipoproteins by chromatofocusing.

Chromatofocusing represents a new chromatographic procedure for the separation of proteins according to their isoelectric points. We describe the application of this method for the fractionation of the urea-soluble apolipoproteins of very low density lipoproteins. They were separated into five peaks, four of which were homogeneous as judged by polyacrylamide gel electrophoresis in the presence of 7 mol/l urea.

[Diagnosis, classification and therapy of urge incontinence].

The motor urge incontinence is characterized through a hyperactivity of the detrusor muscle. Whereas in the secundar form of motor urge incontinence the evoked moment must be abolished in the primar form no method of choice exist. Many different drugs are used in the pharmacological therapy. Biofeedback will be more important in the future. The sensor urge incontinence is caused through urethral relaxation. For the objective assessment must be taken out intraurethral pressure measurements. In this measurements it gives some problems due to the influence of physiological functions.

[Genital Chlamydia infections in female patients at risk of having sexually transmitted diseases].

Investigations for Chlamydia infections of the genital tract were performed in 270 Viennese registered, as well as in 51 non-registered prostitutes. The results were compared with findings obtained in 56 female patients attending an out-patient STD clinic and 48 pregnant women seeking antenatal care in a department of obstetrics. Diagnosis of Chlamydia infection was the organisms on cultured McCoy cells after treatment with cycloheximide. The highest infection rate (31%) was found in non-licensed prostitutes. In registered prostitutes, 20.4% of the cervical smears proved to be positive for Chlamydia. The control groups showed lower infection rates of 9% and 4%, respectively. These results indicate that women who frequently change their sexual partners are considerably more at risk of contracting Chlamydia infections. Hence, this group of persons should be more regularly investigated and--if necessary--given appropriate treatment in order to prevent further spread of the disease.

[Simultaneous urodynamic and electromyographic micturition studies].

The clinical significance of micturition investigations is to be seen not only in the quantification of urethral obstructions e.g. after operations for stress incontinence. Such measurements in fact give information on very different forms of voiding dysfunctions with and without morphological substrates. Simultaneous EMG measurements permit further differentiation, especially in the case of purely functional neurogenic or mitigated neurogenic voiding dysfunctions.

[Intrapartum tcpO2 measurement (author's transl)].

The value of intrapartum tcpO2 measurement is discussed, with reference being made to clinical examples. Also mentioned are certain setbacks of the method which, at present, still are limiting factors to general clinical intrapartum use.
