Radioimmunological quantitation of urinary trypsin inhibitor.

Using monospecific antibody to human urinary trypsin inhibitor, we developed a highly specific and sensitive radioimmunoassay (RIA) for measuring human urinary trypsin inhibitor. No cross-reactivity of the antibody with protein standard serum, which contained albumin, alpha 1-antitrypsin, haptoglobin, alpha 2-macroglobulin, transferrin, IgG and IgA, was observed. The sensitivity of the system was 10 ng of trypsin inhibitor per assay tube, and 5-10 microliters of urine was sufficient to determine the concentration of trypsin inhibitor in urine. The amounts excreted in the urine of 10 healthy men and 10 healthy women were 4.83 +/- 2.46 (mean +/- SD) and 3.86 +/- 1.35 mg/day, respectively. The correlation between estimates by RIA and those by enzymic assay was r = 0.96 (p less than 0.005). The method proposed here can be used to determine the concentration of urinary trypsin inhibitor in a small amount of biological fluids and cells.

Effects of tunicamycin and various monosaccharides on phytohaemagglutinin-induced autorosette formation.

The structural characteristics of autologous red blood cell (ARBC) receptors on human peripheral blood lymphocytes (PBL) induced by phytohaemagglutinin (PHA) were examined. Tunicamycin, which is known to be a blocker of the protein glycosylation of N-glycosidic type glycoprotein, significantly inhibited the production of ARBC receptors. When trypsinized PBL were cultured in the presence of tunicamycin, the inhibitory effect was enhanced. When several monosaccharides were added to the mixture of ARBC and PBL pre-treated with PHA for 24 h, D-mannose caused the most inhibition, and galactose and D-fucose also caused significant inhibition. These data suggest that N-glycosidic type glycoproteins play an important role in binding sites to ARBC on PBL surface membranes and that more than one type of glycose may be involved.

Evaluation of lipid peroxidation of human erythrocyte hemolysates.

Lipid peroxidation of human erythrocyte hemolysates was evaluated by determining thiobarbituric acid reactive substance by fluorospectrophotometer. Because of the non-linear relationship between malondialdehyde formation and hemoglobin concentration, a constant hemoglobin concentration is required to obtain reproducible results. Disruption of the hemolysate by sonication and the presence of formed elements are necessary to induce the reaction, the optimal pH of which was found to be limited to the region of 7.4. Either ferric or ferrous ions are essential for the reaction to proceed, but if both are present, their catalytic activity is abolished. The present method will be useful in the investigation of the mechanism of oxidative damage to erythrocytes under various pathological conditions.

Hexosaminidase isoenzyme profiles in leukemic cells.

The isoenzyme profiles of hexosaminidase in leukemic cells from 39 patients were examined with DEAE-Sephadex chromatography. There was a clear difference in the isoenzyme composition between normal lymphocytes and granulocytes. In acute non-T/non-B lymphocytic leukemia (ALL) a characteristic alteration was found in the intermediate forms of hexosaminidase (Hex I), which was significantly higher than those of normal lymphocytes (p less than 0.001). Subtypes of Hex I (Hex i1-i4) and the heterogeneity of their abnormal expression in ALL was demonstrated. There was no type-specific alteration of the isoenzyme profiles in T-cell leukemia. Leukemic cells of acute myelocytic leukemia (AML) had significantly higher Hex P component than granulocytes (p less than 0.01) and ALL cells (p less than 0.01). The increase of Hex P was evident in childhood AML. There was a significant difference in Hex P level between childhood AML and adult myelo(mono)genous leukemia (p less than 0.001). Chronic myelo(mono)genous leukemia showed similar isoenzyme compositions to normal granulocytes. The isoenzyme profiles in acute undifferentiated leukemia differed from those in other types of leukemia. Isoenzyme analysis might be useful for probing the nature and the intrinsic biochemical abnormality of leukemic cells.

A case of 21q-syndrome with half normal SOD-1 activity.

A male Japanese infant was found to have a chromosomal aberration of del(21)(qter leads to q22.1-2) and decreased superoxide dismutase (SOD) activity in erythrocytes and polymorphonuclear and mononuclear leukocytes. The cuprozinc enzyme (SOD-1) level was 40-50% of normal, while the cyanide-insensitive manganese enzyme (SOD-2) activity was within the normal range. Determination of SOD activity in blood cells is a valuable method of classification of the syndrome.

Free amino acid concentrations in blood cells of two brothers with gyrate atrophy of the choroid and retina with hyperornithinaemia.

In this study we selected blood cells as accessible tissues and measured the free amino acid concentrations in erythrocytes, lymphocytes, and granulocytes of two patients with gyrate atrophy with hyperornithinaemia. We found that ornithine concentrations were elevated in blood cells, to different degrees depending on the type of cell, and that aspartic acid concentrations were low in erythrocytes. We also investigated the fluctuation of free amino acid concentrations in blood cells up to 24 h after the oral administration of ornithine. The ornithine concentration in the blood cells of the patients reached a peak 2 h after the oral administration, and remained high for at least 4 h, although after 2 h the concentration curves differed in the three types of cells.

Androgen receptor in electroresected and cold punch-resected specimens. Usefulness of Kaplan cold punch resectoscope.

The measurement of androgen receptor and 5 alpha-dihydrotestosterone (DHT) levels in prostatic carcinoma may be of value in predicting responsiveness to anti-androgenic therapy. A sufficient amount of prostatic carcinoma tissue must be removed for measuring androgen receptor and DHT levels. We have studied the usefulness of Kaplan cold punch-resection compared with electroresection for obtaining tissue in 29 cases of enucleated benign prostatic hyperplasia. DHT level in electroresected specimens was similar to controls. However, following low, moderate, and high-power electroresection an average of total R1881 binding sites (Bmax) of electroresected specimens was reduced to 82.2, 71.9, and 52.1 per cent of corresponding controls, respectively. In contrast, the per cent decrease of Bmax of Kaplan cold punch-resected specimens was only 10 per cent of the control. From our data, Kaplan cold punch-resection of the prostate appears to be a useful tool for obtaining tissue suitable for measuring androgen receptor levels.

Complete deficiency of adenine phosphoribosyl transferase: report of a new family.

We report a case of 2,8-dihydroxyadenine urinary lithiasis with complete deficiency of adenine phosphoribosyl transferase. Adenine phosphoribosyl transferase activities in the erythrocytes, lymphocytes and granulocytes of the patient's family also were determined. The propositus and her younger brother were homozygotes for adenine phosphoribosyl transferase deficiency and her parents were heterozygotes. This is the third family with this disease to be reported.

The incidence of cystinuria in Japan.

Screening tests for cystinuria were done in approximately 110,000 children and students. The rates of discovery were chemical cystinuria 920 to 1, cystine crystalluria 16,000 to 1 and homozygous cystinuria 18,000 to 1 (8,000 to 1 if a correction was made by the rates of performances of secondary screening and urinary amino acid assay). No stone formation was observed in a 7-year followup of 6 patients considered to be homozygotes. A correlation was observed between urinary cystine and urinary uric acid levels in 39 cases of chemical cystinuria.

Erythrocyte manganese concentration in healthy Japanese children, adults, and the elderly, and in cord blood.

This study showed differences in erythrocyte manganese concentrations in different age groups and in cord blood. All determinations were performed with a flameless atomic absorption spectrophotometer. The mean erythrocyte manganese concentration at one month of age was 435.1 +/- 118.7 ng/g Hb, three to four times higher than in adults. Thereafter, it decreased rapidly and was constant from 4 months to 11 yr of age. However, at 12 to 19 yr of age the concentration in males decreased (108.1 +/- 20.2 ng/g Hb) and was significantly lower than in females (141.7 +/- 25.5 ng/g Hb) (p less than 0.01). This sex difference became even greater in adults aged 20 to 40 yr, when it was 91.5 +/- 22.4 ng/g Hb in males and 141.2 +/- 19.8 ng/g Hb in females (p less than 0.001). The erythrocyte manganese concentration in adults was 31.6 +/- 9.2 ng/ml, somewhat higher than the value previously reported, which may reflect dietary habits in Japan or a racial difference.

Effect of porcine calcitonin therapy on vitamin D metabolism and clinical response in a patient with osteogenesis imperfecta.

A 1 11/12-year-old girl with osteogenesis imperfecta was treated with porcine calcitonin. Eight bone fractures occurred in the previous 20 months before therapy, but none occurred during eight months of therapy. There was also a significant improvement in linear growth and radiographic bone density. This is the first study of the effect of calcitonin on vitamin D metabolism in a human. The high plasma levels of 1,25 dihydroxy-vitamin D (1,25-(OH)2-D) and 24,25 dihydroxy-vitamin D (24,25-(OH)2-D) before calcitonin therapy decreased after therapy. Plasma 25 hydroxy-vitamin D (25-OH-D) concentration, which normal in level before calcitonin therapy, was normal or slightly decreased during administration. It is concluded that calcitonin probably influences vitamin D metabolism in a patient with osteogenesis imperfecta.

Elevation of lipid peroxide in children treated with a combination of chemotherapeutic agents including doxorubicin.

The fluorometric method of Yagi was used to determine serum lipid peroxide levels in children with various types of malignancies, treated with or without doxorubicin. It was found that patients on regimens of intermittent combination chemotherapies including doxorubicin had higher levels of serum lipid peroxide and were all apparently free from cardiotoxicity. Doxorubicin was the only determinant in affecting the serum lipid peroxide value. Coenzyme Q10 given concurrently failed to reduce the elevated levels to normal.

Levels of vitamin D-metabolites in a case of acro-osteolysis syndrome.

On the initial examinations of a 17-year-old boy with acro-osteolysis syndrome, the levels of serum 25-hydroxycholecalciferol (25OHD) were low, whereas serum levels of 1,25-dihydroxycholecalciferol (1,25(OH)2D) were normal. Administration of 1 alpha-hydroxycholecalciferol (1OHD) failed to improve the osteoporosis, although the serum 25OHD level rose to normal. It is unlikely that impaired bone remodeling due to abnormal vitamin D-metabolism was a primary defect in our patient.
