Renal hemangioma.

Four cases of renal hemangioma are presented. Renal hemangioma is difficult to detect because this benign vascular tumor never demonstrates any abnormalities on renal arteriography. Computed tomography in two resected cases of cavernous hemangioma revealed a low-density mass without any enhance effect, while the others diagnosed by the selective renal arteriography demonstrated no abnormality on computed tomography. We postulate that both angiographic and computed tomographic appearances of the renal hemangioma could depend on its vascular components. Related reports are also reviewed.

Health status of Indochinese refugees in Japan: statistical analyses on anemia, eosinophilia and serum alkaline phosphatase.

Detailed statistical analyses were attempted on anemia, eosinophilia and elevation in serum alkaline phosphatase in the Indochinese refugees in Japan. A high statistical correlation was found between hemoglobin concentration and hematocrit for most of the refugees except the Cambodian females. Refugees with parasitic infections, particularly hookworm infection had significantly lower values for hemoglobin concentration and hematocrit. Significant eosinophilia was observed in the refugees infected with hookworms, Strongyloides stercoralis, Rhabditis sp., Endolimax nana and/or Hymenolepis nana. However, these parasites were found in the same subpopulations more frequently than expected by chance. Elevation in serum alkaline phosphatase was observed in young Cambodian immigrants. Analyses showed that the variations of this enzyme could be well predictable by those of enzymes such as serum glutamic oxaloacetic transaminase, which are good markers for liver functions, in Cambodian immigrants, 13-24 of age, whereas no such correlation was observed in non-Cambodians of the same age group.

[Spergualin a novel antitumor antibiotic produced by Bacillus laterosporus].

Spergualin, named after the spermidine and guanidine moieties in its structure, was isolated from culture broth Bacillus laterosporus of based on its greater growth-inhibitory activity against chick embryo fibroblasts (CEF) transformed by Rous sarcoma virus compared with that against normal CEF. The structure of spergualin was determined as (-)-(15S)-1-amino-19-guanidino-11, 15-dihydroxy-4, 9, 12-triazanonadecane-10, 13-dione. Spergualin is water-soluble, basic and a white powder. It shows cytotoxicity in vitro against mouse leukemias L1210 and L5178Y. It significantly prolongs the life span of mice transplanted with either of the above leukemic cells. It induces a strong and specific immunological effect on L1210 which has been maintained in our laboratory; L1210-transplanted CDF1 mice, once cured by spergualin, reject a second challenge of the leukemic cells. Spergualin hibits in growth in vitro of both Gram (+) and (-) bacteria at concentrations ranging from 6.25 to 100 micrograms/ml.

[A case of perirenal encysted hematoma secondary to renal cell adenocarcinoma].

Herein we report a case of perirenal encysted hematoma secondary to renal cell adenocarcinoma. A 57-year-old woman was hospitalized because of a palpable mass in the left flank. She had had an episode of left flank pain. X-ray studies demonstrated a tumor in the left kidney and a large cystic mass beneath the left kidney. The cyst was excised by nephrectomy. Histological examination of the large cyst revealed perirenal encysted hematoma secondary to renal cell adenocarcinoma. Six months postoperatively, she is now free from left lumbar pain, and alive with lung and liver metastasis.

Cell behaviour in a polygonal cell sheet.

Cell monolayers on culture dishes were divided into two groups: tensile monolayers and non-tensile ones. In the development of an epithelium, a non-tensile cell monolayer turns into a tightly bound tensile one. Detection of these states was carried out by using the boundary shortening procedure, a computer-based geometrical method to show how much the polygonal cell boundary contracts. Non-tensile monolayers were divided further into two groups according to their motility: a fluctuating monolayer in which cells move laterally, and a stable monolayer in which cells are immobilized. Quantitative determination of cell motility was performed by analysing time-lapse cellular patterns. These computer-based geometrical analyses enabled us to divide monolayers into three groups: tensile stable monolayers, non-tensile stable monolayers and fluctuating monolayers, and this study therefore gives an insight into the way in which changing conformations of cells may be assayed.

The differentiation in vitro of the neural crest cells of the mouse embryo.

A culture method for neural crest cells of mouse embryo is described. Trunk neural tubes were dissected from 9-day mouse embryos and explanted in culture dishes. The developmental potential of mouse neural crest in vitro was shown to be essentially similar to that of avian neural crest. In the mouse, however, melanocytes always appeared in association with the epithelial sheet close to the explant. Neural crest cells surrounding the epithelial sheet, which probably migrated from the neural tubes in the early culture phase, never differentiated into melanocytes. The bimodal behaviour of mouse crest cells seems to be due to the heterogenous potency of the crest cells and the interaction of these cells with the surrounding microenvironment. This culture system is well suited for various experiments including the analysis of gene control on the differentiation of neural crest cells.

[A case of giant prostate cancer].

A 67-year-old patient with stage C giant prostate cancer was treated with a combination of endocrine administration and chemotherapy. After administration of diethylstilbestrol diphosphate (250 mg/D, 16 days), bilateral orchiectomy and subsequent CDDP administration (30 mg/D X 5 days, e. 3 weeks, 4 courses), the primary tumor was reduced by about 90%. Clinical response was evaluated as partial response. Serum acid phosphatase activity, prostatic acid phosphatase, prostate antigen and nuclear DNA histogram served as useful tumor markers and changed in parallel to clinical course.

Electron microscopic study of cells in vitreous of guinea pig.

The fine structure of the cells in the posterior segment of the vitreous body of three Hartley strain guinea pigs was investigated by electron microscopy. A total of fifty-four cells were observed. Fifty-three of the 54 cells had specific granules which manifested homogeneous or heterogeneous appearance and were considered as hyalocytes. A difference in cell activity was demonstrated among these cells. Only one cell of the 53 suggested phagocytosis. Another cell was discharging lipopigment. This suggests that hyalocyte granules are gradually degraded, becoming of more complicated structure, and are discharged into extracellular space. One of the 54 cells had cytoplasmic filaments and was not considered as a hyalocyte.

Effect of bestatin on syngeneic tumors in mice.

The antitumor effect of bestatin against syngeneic tumors such as colon adenocarcinoma 26 (colon 26) and myeloid leukemia C1498 ( C1498 ) was investigated. Bestatin effectively inhibited the growth of both tumors in vivo. The inhibition observed for colon 26 was the largest when bestatin was administered on days 1 to 5 after transplantation of the tumor. The inhibition of C1498 was the largest when bestatin was administered on days 7 to 11. In contrast to these results, bestatin was not at all inhibitory to either tumor line in vitro. Thus, the effect of bestatin administration on the functions of the spleen cells of mice bearing tumors was examined. Spleen cells taken from mice bearing colon 26 and C1498 did not neutralize the corresponding tumor. However, when bestatin was administered to these mice, spleen cells from the administered mice did neutralize the corresponding tumor. Bestatin enhanced antibody-dependent cell-mediated cytotoxicity and natural killer cell activity of the spleen cells of colon 26-bearing mice. Further, bestatin did not inhibit the growth of colon 26 in BALB/c nu/nu mice. It was concluded that bestatin exhibits its antitumor effect against colon 26 and C1498 indirectly by immunomodulation via spleen cells and possibly via T cells.

Anti-idiotypic antibodies in a patient with monoclonal rheumatoid factor after pneumococcal bacteremia.

A 51-yr-old Japanese female patient with monoclonal IgM gammopathy with rheumatoid factor activity was admitted because of pneumococcal bacteremia. About 2 wk after admission, her rheumatoid factor activity became undetectable by RAHA test and radioimmunoassay, subsequent to the initial marked elevation. The suppressive capacity of the patient's IgG fraction on the rheumatoid activity of her monoclonal IgM on January 11 was determined. The IgG fraction obtained on February 22 blocked the binding of the rheumatoid factor to rabbit IgG. The suppressive activity in the IgG fraction of February 22 was shown to be localized within the F(ab')2 fragment. Furthermore, the specificity of the suppressive serum factor was shown by the inability to block the binding of SRBC coupled with diazotized phosphorylcholine to anti-pneumococcal antibody. Thus, the marked reduction of rheumatoid factor activity was considered to result from anti-idiotypic antibody transiently appearing in her serum after pneumococcal bacteremia.

Improved determination of transketolase activity in erythrocytes.

We describe a new and better method for determining transketolase (EC 2.2.1.1) activity in human erythrocytes. Heating the hemolysate (55 degrees C, 5 min) inactivates transaldolase (EC 2.2.1.2), the enzyme that catalyzes the formation of fructose 6-phosphate and erythrose 4-phosphate from sedoheptulose 7-phosphate and glyceraldehyde 3-phosphate. The net effect is that the quantity of sedoheptulose 7-phosphate formed more precisely represents the transketolase activity, which is unaffected under these conditions. Use of ribosephosphate isomerase (EC 5.3.1.6) and ribulose-phosphate 3-epimerase (EC 5.1.3.1) to establish an equilibrium among the pentoses allowed us to confirm the stoichiometry of the transketolase reaction. We also discuss the effect of thiamin pyrophosphate, which is used to reflect thiamin deficiency.
