Vascular permeability activity in Escherichia coli heat-stable enterotoxin.

Purified heat-stable enterotoxin (ST) prepared from supernatants of Escherichia coli strain 53402 A-1, isolated from a patient with diarrhea, caused an increase in the permeability of the small blood vessels of the skin of adult rabbits after intracutaneous injection. Increased permeability was manifested by localized accumulation of intravenously injected blue dye at the injection sites. Permeability factor (PF) activity reached a peak 1 h after injection, and recovery of normal permeability was nearly complete in 3 h. Residual PF activity had disappeared by 24 h. Dose-response curves demonstrated a straight-line relationship between the logarithm of the dose and mean blueing diameter over a range of 3 to 12 mm. The PF assay was less sensitive that the suckling mouse assay, and adult rabbits varied in their sensitivity to the PF effect. Five to 100 mouse units of ST were required to consistently evoke strong and unequivocal blue lesions of 7 mm or more in diameter. PF activity was reduced 50 to 70% by heating at 70 degrees C for 30 min, 60 to 80% by boiling for 30 min, and 95 to 98% by autoclaving for 15 min, but it was unaffected by treatment with cholera antitoxin. These findings are consistent with the notion that PF activity is a property of the ST molecule itself. This PF assay cannot be used for the detection of ST in crude culture supernatant fluid because of low sensitivity and the presence of nonspecific PF effects of culture media and other E. coli products.

Lysyl oxidase activity in the mouse uterine cervix is physiologically regulated by estrogen.

Lysyl oxidase activity in the mouse cervix was examined during the estrous cycle, pregnancy, and the postpartum period. It was highest in estrus and lowest in diestrus. On day 1 of pregnancy, the activity was comparable to that seen in estrus. It then declined continuously until day 17 of pregnancy, when it was approximately equal to that seen during diestrus. The activity showed a slight increase toward term, then a precipitous increase occurred on the first postpartum day. The activity fell by the seventh postpartum day to a value comparable to that seen in diestrus. Enzyme activity fell to one fourth of its former value 3 days after ovariectomy. The injection of 1 microgram 17 beta-estradiol or more in 0.9% NaCl 10 days after ovariectomy raised the activity, in 18 h, to the level seen in estrus. One milligram of progesterone had no effect on the enzyme activity, but a combination of 1 microgram 17 beta-estradiol and 1 mg progesterone raised the activity to that seen with 1 microgram 17 beta-estradiol alone. These results suggest that lysyl oxidase activity in the mouse cervix is under the control of estrogen.

Blood pressure variations to be considered in the treatment of hypertension.

Diurnal variations of the basal blood pressure in patients with essential hypertension were investigated by the indirect recording methods. To estimate the reduction in blood pressure at midnight, disturbance of sleep in the patients should be considered. Relationship between neurohumoral factors and the diurnal blood pressure variations were also evaluated. 1) Variations of the casual blood pressure at the clinic visits may be stabilized beyond 15 minutes of the sitting rest. 2) During the out-patient treatment of essential hypertension with placebo for 6 weeks, 16% of the patients showed reduction in mean arterial pressure of 13 mmHg or more. 3) Significant influence of seasons on blood pressure was not apparent for 6 weeks of the placebo treatment of hypertension. But in patients under the long-term antihypertensive treatment, blood pressure in summer was maintained significantly lower than in winter. 4) Self-monitoring of blood pressure by patients at home is valuable for estimating diurnal changes of blood pressure with the antihypertensive treatment.

Dilatation of the aortic valve portion in aortitis syndrome. Angiographic evaluation of 70 patients.

The dilatation of the aortic valve portion was evaluated by angiography in 70 patients with aortitis syndrome (Takayasu's arteritis). The diameter at aortic valve commissure was 20 +/- 2.8 min/M2 (NS, vs control) in 46 patients without aortic insufficiency (AI), 23 +/- 2.0 (p less than 0.02, vs without AI) in 11 patients with AI (I or II in grade), 27 +/- 2.0 (p less than 0.01, vs I or II in grade) in 13 patients with AI (III or IV in grade). The diameter was 20 +/- 2.0 in 13 normal control cases and 20 +/- 4.0 in 6 patients with rheumatic AI. The diameters at the levels of aortic valve annulus, sinus of Valsalva and ascending aorta increased also significantly in the patients with AI secondary to aortitis syndrome. The diameter at aortic valve commissure in 24 patients with AI was correlated (r = -0.48, p less than 0.05) to the diastolic blood pressure. Any diameter at the aortic valve portion was not different significantly by the complication of systolic hypertension (greater than or equal to 180 mmHg) in both groups of cases with and without AI. These results indicated that the dilatation of the aortic valve portion was observed in the patient with AI secondary to aortitis syndrome and might be the primary and common cause of AI in all grades, and that the dilatation might not be accelerated by the associated hypertension.

The effect of L-asparaginase on cholesterol biosynthesis.

The liver from the Wistar rat was incubated either in the solution of 1 micro Ci acetate-1-14C or 0.1 micro Ci mevalonic acid-2-14C, and incorporations of radioactivity to phospholipid and cholesterol were estimated respectively. The incorporation of labeled acetate to cholesterol in the L-asparaginase-treated rat was significantly lower than that in the controls. However, there were no differences of the incorporation into the mevalonic acid between the study group and the controls. These results suggest that the inhibitory mechanism may exist between the steps of acetate and mevalonic acid.

Prophylaxis of bacterial infection by sulfamethoxazole-trimethoprim (SMX-TMP) during chemotherapy in patients with childhood acute leukemia.

A combination of sulfamethoxazole and trimethoprim was given orally to 13 children with acute leukemia on 16 occasions of hospitalization during remission induction chemotherapy for the prophylaxis of bacterial infection. Frequency of episodes of persistent fever in this group of patients was markedly low, namely 0.38 per one hospitalization, whereas that in control group which was given no drug was 0.98. Furthermore, frequency of episodes of definite bacterial infection in the patients given SMX-TMP was 0.25 per one hospitalization. This was significantly low as compared with control patients whose frequency was 0.84. Although, there occurred slight rash and liver dysfunction as the side effects, they were reversible. These results suggest that the prophylactic use of SMX-TMP in children with acute leukemia during chemotherapy is effective and valuable for the protection from bacterial infection.

Impaired leukocyte mobility and production of monocyte-derived granulotactic factor in pediatric malignant disease during chemotherapy.

Random mobility and chemotactic responses of granulocytes and monocytes, and production of monocyte-derived chemotactic factor for granulocytes (CFG) in the 17 patients with childhood malignant disease were estimated using an agarose plate method. Mononuclear cells (MNC) of the majority of the patients contained a significantly decreased number of monocytes with normal migratory capacity, whereas random mobility of granulocytes of all the patients was within normal range. Chemotactic responses of granulocytes and monocytes to zymosan activated serum (ZAS) were decreased in 11 to 17 occasions. All these impairments seemed to be induced by chemotherapeutic agents, because there were no differences in the degree of the impairment between group in remission with maintenance chemotherapy and that during induction chemotherapy.

Estrogen induces different responses in dermal and lung fibroblasts: special reference to collagen.

Fibroblast-like cells derived from dorsal skin and lung of AKR strain mouse embryos were cultured with estrogen for 4 days. The labeled hydroxyproline content was measured as newly synthesized collagenous protein in the cell layer and media. Although collagen synthesis by both cell lines was increased in both fractions under the influence of beta-estradiol-3-benzoate, at physiological concentrations of 10(-3)-10(-1) microgram/ml, the rate of increase differed. Fibroblasts derived from skin showed increased collagen synthesis of approximately 76%, while those from lung showed an increase of approximately 25%. Total protein synthesis by both cell lines also increased. In lung fibroblast cultures the synthesis of total protein was increased more than the synthesis of collagens; on the other hand in skin fibroblast cultures the synthesis of collagen was increased more than the synthesis of total protein. DNA synthesis by both cell lines was not affected by estrogen at the concentrations used. These findings suggest that fibroblasts develop in a different manner in each organ.

Genetic labeling of an Ent plasmid that encodes heat-stable enterotoxin of enterotoxigenic Escherichia coli isolated from patients.

Ent plasmids of enterotoxigenic Escherichia coli that encode heat-stable enterotoxin (ST) and both ST and heat-labile enterotoxin (LT) were labeled with a kanamycin- and an ampicillin-resistance transposon, respectively. These transposons were kept stably in the respective Ent plasmids. By monitoring antibiotic resistance we could easily select strains carrying Ent plasmids. Strains carrying the labeled plasmids produced enterotoxins that were immunologically indistinguishable from those produced by the parent strains. Moreover the strain carrying a labeled ST-Ent plasmid produced 16 times more ST than the parent strain.

Evidence for receptor-dependent response to dihydrotestosterone in cultured human fibroblasts.

The effect of dihydrotestosterone on the production of proteins and collagen and the synthesis of DNA was studied in cultured fibroblasts obtained from a normal woman and from a patient with complete testicular feminization. The concentration of dihydrotestosterone binding sites in fibroblasts from vulvar skin of a normal woman and from perineal skin of a patient with complete testicular feminization was 35.9 +/- 1.7 fmoles/mg protein and 4.61 +/- 0.37 fmoles/mg protein, respectively. The apparent dissociation constant was 0.24 +/- 0.015 nM for fibroblasts from a normal woman and 0.21 +/- 0.031 nM for fibroblasts from a patient with complete testicular feminization. Fibroblasts grown to confluence were exposed to DHT in the concentrations of 10, 100 and 1000 ng/ml of the culture medium and cultures were maintained for up to six days thereafter. These hormone treatments brought about a significant elevation in the production of proteins and collagen by fibroblasts from a normal woman but failed to produce any perceivable changes in fibroblasts from a patient with complete testicular feminization. The synthesis of DNA in fibroblasts from either subject was unaffected by hormone treatment. These results suggest that augmented production of proteins and collagen in cultured human fibroblasts which follows stimulation by dihydrotestosterone occurs via a receptor system.

Dissociation of the 5 alpha-reductase activity from the specific dihydrotestosterone binding activity in cultured human fibroblasts.

Four fibroblast lines (F1-4) were established from abdominal (F1 and F2) and vulvar (F3 and F4) skin samples obtained from women who underwent delivery. The concentration of dihydrotestosterone (DHT) binding sites was measured along with the maximum velocity for the formation of DHT from testosterone in fibroblasts from each cell line. The concentration of DHT binding sites (fmoles/mg protein) was 12.8 +/- 1.1 for F1, 12.3 +/- 1.0 for F2, 14.7 +/- 1.3 for F3 and 13.4 +/- 1.1 for F4 (mean +/- S.D. of two determinations). The respective values for the maximum velocity for the formation of DHT from testosterone (pmoles DHT formed/mg protein/h) were 8.40 +/- 0.64, 21.5 +/- 1.69, 11.8 +/- 0.85, and 31.0 +/- 2.26 (mean +/- S.D. of two determinations). These results, which demonstrate a wide diversity among values for the maximum velocity for the formation of DHT from testosterone while values for the concentration of DHT binding sites in these fibroblasts are uniform, suggest a lack of correlation between these two activities of fibroblasts.
