Biochemical characterization of eel virus european.

The polypeptide and RNA compositions of purified virions of eel virus European were compared to those of three strains (VR-299, SP and AB) of infectious pancreatic necrosis virus (IPNV) isolated from trout. All three IPNV strains could be distinguished by the relative mobilities of either the virion polypeptides or the double-stranded RNA genome segments. The eel virus had a similar polypeptide profile to strain AB IPNV but differences between the two viruses in the migration of genome segments indicated each was unique.

Immunohistochemistry of myoglobin in rhabdomyosarcomas.

Immunoperoxidase staining for myoglobin was performed on formalin-fixed, paraffin-embedded tissues of normal human skeletal muscle and rhabdomyosarcomas in order to assess its potential diagnostic value. Myoglobin-positive cells were observed in normal adult and fetal muscles, and in 13 of 26 rhabdomyosarcomas. In normal skeletal muscle, the intensity of staining for myoglobin varied in individual cells. Of the 13 myoglobin-positive rhabdomyosarcomas tested, 6 showed cross striations in tumor cells, whereas the others did not. Myoglobin-positive tumor cells were not demonstrated in two rhabdomyosarcomas in which cross striations were observed. In other types of tumors examined that sometimes resembled rhabdomyosarcomas, no myoglobin was detected. Thus tests for myoglobin by the immunoperoxidase method seem helpful in identifying rhabdomyosarcomas and confirming their histological diagnosis.

Anaplastic carcinoma of the esophagus.

The present report includes the histological, electron microscopical and immunohistochemical findings observed in two cases of anaplastic carcinoma of the esophagus. Anaplastic carcinoma of the esophagus revealed histological features similar to oat-cell carcinoma and carcinoid. Positive argyrophil reaction, neurosecretory granules, and hormonal activity suggest that both cases come under the category of neuroendocrine cell tumors. Anaplastic carcinoma of the esophagus may be considered as a pluripotential tumor. The biological behavior of this tumor is quite different from that of carcinoid and is a highly malignant tumor.

Fetal and maternal plasma levels of immunoreactive somatostatin at delivery: evidence for its increase in the umbilical artery and its arterio-venous gradient in the feto-placental circulation.

The concentrations of immunoreactive somatostatin (IR-SRIF) in plasma samples taken from the umbilical artery (UA), umbilical vein (UV), and maternal antecubital vein (MV) were measured in 23 cases of normal delivery at term. High concentrations of IR-SRIF were detected in the plasma from the UA (mean +/- SD, 73.2 +/- 40.6 pg/ml), the value being about 2.5 times that in the UV (29.5 +/- 17.5 pg/ml; P less than 0.001). An arterio-venous gradient was observed in all 23 subjects. The plasma level of IR-SRIF in the MV (10.9 +/- 4.6 pg/ml) was significantly lower than those in the UA (P less than 0.001) and UV (P less than 0.001) and was almost the same as that in nonpregnant women (12.7 +/- 5.4 pg/ml). Chromatographic analysis of an extract of plasma from the UA gave only one peak of immunoreactive material, which was eluted in the same position as synthetic SRIF-14. A similar result was obtained in nonpregnant women. No correlation was found between the plasma level of IR-SRIF and that of GH, insulin, glucagon, or gastrin in the UA. The present findings suggest that the high concentration of IR-SRIF in the feto-placental circulation originates from the fetus and reflects a particular role of this peptide in the process of functional maturation of the neuroendocrine system in early human life.

New alpha-amylase inhibitor, trestatins. I. Isolation, characterization and biological activities of trestatins A, B and C.

Trestatin complex which exhibited a potent inhibitory activity on various alpha-amylases has been isolated from the culture filtrate of Streptomyces dimorphogenes nov. sp. NR-320-OM7HB. Three major components, trestatins A, B and C, have been purified by adsorption and ion-exchange chromatography. Their spectral and chemical properties suggested that trestatins were novel basic oligosaccharide homologues each characterized by the possession of a trehalose moiety at the non-reducing end of the molecule.

[Evaluation of radioimmunoassay of the blood tobramycin level for the purpose of setting the dosage based on the clinical pharmacokinetic theory and a comparison of radioimmunoassay with other assay methods].

With the purpose to use for therapeutic drug monitoring (TDM), blood concentrations of tobramycin (TOB) in each patient were measured by radioimmunoassay (RIA). A RIA kit of TOB (Clinical assay-Japan Travenol) was evaluated for precision and recovery, in that partial improvement of the method was made, in order to measure low level of TOB. The RIA was compared with high-performance-liquid-chromatography (HPLC), bioassay (BA) and 2 kinds of enzyme immunoassay (EIA) (EMIT and SLFIA). The RIA of TOB revealed high precision (1.8-2.4% in C.V.) and high reproducibility (5.0-6.9% in C.V.). It was found that this RIA kit can be used for measuring low level of serum TOB concentrations by a modification of the method. The total range of measurable blood level is from 0.1 to 16.0 micrograms/ml. The nearly one to one correspondence was observed between RIA and other 4 methods, when 154 samples obtained from 18 cases were measured. A representative case of TDM for TOB was demonstrated, in which predicted concentrations agreed fairly well with actual measured values at steady state. It was concluded that the RIA kit is useful for clinical application of TDM for the adequate dosage regimen of TOB. Modification of the method for rapid assay of a small number of samples will increase the clinical usefulness.

Distribution of histaminase in human tumor tissues. An immunohistochemical study.

Tissue samples from 125 cases of a variety of human malignancies and benign conditions were examined for the presence of histaminase by the immunoperoxidase method on conventional pathological specimens. Histaminase was found diffusely in the cytoplasm of cancer cells of primary and metastatic tumors of lung, thyroid gland, pancreas, stomach, esophagus, ovary and pharynx. Histaminase-positive cases were also found in carcinoids, dysgerminomas and in melanoma. The incidence of positive histaminase staining in epithelial malignancies overall was 28/84, and those in sarcoma as well as in benign conditions were 1/22 and 0/19, respectively, excluding placental decidual cells and eosinophils intermingled in the tissues. No significant difference in the incidence of histaminase was found by histological type or by degree of cellular differentiation. The present results thus provided another direct evidence for the production of histaminase by many kinds of tumor cells.

[Evaluation of left ventricular asynergy by two-dimensional echocardiography: comparison between fixed external axis system and floating axis system].

Detection and quantitative estimation of segmental asynergy in the infarcted area has been impeded by the rotation and anterior movement of the heart during systole. In the present communication, we compared the two measurements determined by the floating and fixed external axis systems. The former takes into account of the effect of cardiac movement. Furthermore, we assessed which system is superior by the left ventriculographic and coronary arteriographic analyses. The materials consisted of 24 cases of myocardial infarction. There was no significant difference between the two systems in the estimation of the number of asynergic segments or between echocardiographic and ventriculographic findings. The floating axis system had a tendency to underestimate asynergy, so that the number of hypokinetic segment estimated by the floating system was greater than that by the fixed external axis system. Therefore, the fixed external axis system was superior to the floating axis system in determining the causative coronary artery.

[Clinical significance of colorectal cancers arising after partial or total gastrectomy].

Among colorectal cancer patients operated at the 2nd Department of Surgery, Kyushu University Hospital between 1974 to 1980. 6 developed this cancer after partial or total gastrectomy for gastric cancer or ulcer. The interval between the gastric operation and colorectal surgery ranged from 15 months to 17 years. The colorectal cancers arose in the transverse colon (1) the sigmoid colon (2), there were 3 rectal cancers. Therefore, not only the occurrence of recurrent or primary cancer in the remnant stomach, but also the occurrence of colorectal cancer should be monitored carefully after partial or total gastrectomy for gastric cancer or ulcer.

Organic acid metabolism in a patient with ornithine transcarbamylase deficiency.

Serum and urinary organic acids in a 9-year-old girl with ornithine transcarbamylase deficiency were investigated. The urinary excretion of alpha-ketoglutarate (alpha-KG) and citrate were abnormally high. An inverse linear correlation was found between the plasma ammonium and alpha-KG levels when the plasma ammonium level was less than 200 micrograms/dl, but not at higher levels, suggesting that on overloading with ammonium, its route of metabolism and compensating mechanism are not functionally normal. After administration of NH4Cl, plasma and urinary levels of alpha-KG, pyruvate and citrate increased, but the urinary succinate level decreased slightly. After oral administration of citrate the plasma alpha-KG level increased and the ammonium level decreased transiently. Our results suggest that administration of citrate or alpha-KG is not effective for reducing hyperammonemia in ornithine transcarbamylase deficiency.

Immunoreactive somatostatin in catecholamine-producing extra-adrenal paraganglioma.

A patient with catecholamine-producing extra-adrenal paraganglioma was found to have a high concentration of immunoreactive somatostatin (IR-SRIF) in the peripheral blood plasma (47.0 pg/ml, normal; 13.3 +/- 5.3 pg/ml, mean +/- SD). After removal of the tumor, the plasma SRIF level fell within normal range (8.7 pg/ml). In the resected tumor, a considerable amount of IR-SRIF (71.0 ng/g wet weight)( was also detected and SRIF-positive tumor cells were demonstrated by the immunoperoxidase technique. Chromatographic analysis of an extract of the tumor showed the presence of two IR-SRIF components of large molecular size besides a component consistent with authentic tetradecapeptide SRIF (SRIF 14), and one of these had the same molecular size as octacosapeptide SRIF (SRIF 28). This seems to be the first report of a catecholamine-producing extra-adrenal paraganglioma that produces IR-SRIF with molecular heterogeneity.

Determination of the antibody to hepatitis B e antigen by the inhibition on electrophoresis of free e antigen.

A new method is described for the rapid and sensitive detection of the antibody to hepatitis B e antigen (anti-HBe) in serum. Twenty-five microliters of the test serum were incubated with 10 microliter of the purified small 'free' e antigen, and then the consumption of the added antigen was detected by the failure to precipitate with standard anti-HBe in counter-electrophoresis. When a total of 444 serums from asymptomatic carriers of hepatitis B surface antigen was tested for anti-HBe by this method, 348 (78.4%) were found to be positive with a detectability much higher than that of the passive hemagglutination method which detected the antibody in only 264 (59.5%) of the same serums.
