Double-stranded cDNAs of hop stunt viroid are infectious.

Restriction fragments composed of only hop stunt viroid (HSV) cDNA were prepared from recombinant clone pHS-P4P, which carries four tandemly repeated HSV cDNAs, and inoculated into cucumber cotyledons. The results showed that double-stranded cDNAs consisting of 1 to 3 units of HSV sequences were infectious. In cucumber plants inoculated with these double-stranded cDNAs, infectious RNA molecules indistinguishable from authentic HSV were propagated.

Major-histocompatibility-complex-restricted male skin graft rejection responses in rats.

Normal DA (RT1a) female rats rejected male skin isografts when preimmunized with RT1-compatible DA, or ACI (RT1a) male bone marrow cells (BMC), but failed to reject them when preimmunized with RT1-incompatible PvG/c (RT1c), F344 (RT1lvl), or BN (RT1n) male BMC--as well as when they were untreated. DA female rats rendered neonatally tolerant to RT1-incompatible PvG/c female tissues failed to reject, not only DA, but also PvG/c male skin grafts when preimmunized with PvG/c male BMC. DA female rats tolerant of PvG/c female tissues that had rejected DA male skin grafts following immunization with DA male BMC failed to reject PvG/c male skin grafts. These results indicate that in rats, unlike mice, male skin graft rejection responses are MHC-restricted, at least in the DA strain.

Malignant lymphoma, small non-cleaved, non-Burkitt's type.

An autopsy case of "Malignant lymphoma, small non-cleaved, non-Burkitt's type" with hypercalcemia is presented. A 57-year-old Japanese man suffering from epigastralgia and "retroperitoneal tumor" for nine months was found at autopsy to have two large tumor masses in the retroperitoneum and pelvic cavity. Tumor cell infiltration into the liver, spleen, both kidneys, and both adrenal glands was observed on microscopic examination. The tumor cells were lymphoid cells with the characteristics of "Malignant lymphoma, small non-cleaved, non-Burkitt's type". The importance of recognizing non-Burkitt's type is pointed out. The cause of the presence of hypercalcemia is also discussed.

Histopathological and immunohistochemical study of neuroendocrine tumors of the rectum.

A clinicopathological and immunohistochemical study was carried out on 32 cases of neuroendocrine tumors of the rectum. Typical carcinoids consisted of 27 cases, histologically showing uniform round to columnar cells forming solid alveolar nests and ribbon-like or trabecular arrangement. Neuroendocrine carcinomas consisted of 5 cases in which tumor cells with prominent nuclear atypism were arranged in a ribbon-like or trabecular fashion and formed gland-like structures. There were also small round tumor cells resembling lymphocytes. The prognosis of neuroendocrine carcinomas is very poor with marked tumor invasion of lymphatics and veins resulting in liver metastases and death within one year after operation. Thirty cases out of the 32 showed a positive argyrophil reaction, while immunohistochemistry of 29 cases revealed more than one peptide hormone in 23 cases. The most common hormone was somatostatin being present in 18 of the 23 tumors and glucagon in 16 of the 23 tumors. Gastrin/CCK and calcitonin were proven in 6 of the 23 tumors and in 4 of the 23 tumors, respectively. On the other hand, more than two hormones was present in 15 of the 23 tumors examined. Histologically, neuroendocrine tumors have a very wide spectrum. Histogenetically, typical carcinoids and neuroendocrine carcinomas are considered to be of the same origin with the former showing morphological and functional differentiation to endocrine cells and the latter being more undifferentiated.

[Peplomycin sensitivity of various types of human gynecological cultured tumor cells].

Various types of human gynecological cultured tumor cells were tested for the sensitivity to Peplomycin (PEP), an effective antitumor antibiotic for squamous cell carcinomas, by the regrowth assay method together with morphological observation. Bleomycin-hydrolase activity of these cell lines was also compared in cell-free extracts by assaying the conversion of Bleomycin into its deamidated from (HPLC method). SKG-I, SKG-II, SKG-IIIb cells derived from squamous cell carcinoma of the cervix and RKN cells derived from myosarcoma of the ovary were much more sensitive to PEP than other cell lines. PEP was found to be mainly a time-dependent drug, but also concentration dependent. The effect of PEP on cell morphology was characterized by the appearance of enlarged cells and swelling nuclei. The specific activities of Bleomycin-hydrolase in SKG-I, SKG-II, SKG-IIIb cells were shown to be relatively lower than that in other cell lines. These results suggested that cervical squamous carcinoma cells and ovarian myosarcoma cells were sensitive to PEP and Bleomycin-hydrolase activity was one of factors which decided the PEP sensitivity of human cultured tumor cells.

A case of pleomorphic adenoma of the trachea.

An unusual tumor of the trachea was found in a 76-year-old woman. The tumor resembled an adenoid cystic carcinoma in having many cystic spaces, but the fairly uniform tumor cells gradually merged with the myxomatous or hyalinous stroma, and foci of squamous metaplasia and glycogen-rich adenoma-like components were also observed. S-100 protein was demonstrated immunohistochemically in the tumor cells. This tumor was diagnosed as a pleomorphic adenoma (benign mixed tumor) of the trachea.

A two-site sandwich radioimmunoassay of human fibroblast (beta-) interferon with monoclonal antibodies.

Monoclonal antibodies were produced against human fibroblast (beta-)interferon (IFN-beta). Four of them were directed to the determinant designated a, while the remaining 3 to another determinant named b. An IFN molecule was found to bear one each of a and b determinants arranged in such a manner that the occupation of a with the corresponding antibody did not interfere with the binding of b to its corresponding antibody, and vice versa. This allowed the development of a 2-site sandwich radioimmunoassay in which antibody to b was immobilized on wells of a microtiter plate and the bound antigen was detected by the radiolabeled antibody to a. The 2-site sandwich radioimmunoassay detected, with a high sensitivity, IFN-beta either induced from fibroblast or produced by Escherichia coli harboring the gene of IFN-beta.

Influence of lipids metabolism on platelet activation in vivo.

Platelet aggregability and plasma factor VIII-related antigen (F. VIIIR:AG) level in 16 ischemic heart disease (IHD) patients were increased by isometric exercise and these changes were prevented by administration of a lipid lowering agent, simfibrate, a derivative of clofibrate. Serum total cholesterol (TC) level decreased and the high density lipoprotein-cholesterol (HDL-C)/TC ratio increased with the treatment. Another 7 hyperlipidemics were administered with simfibrate. Platelet malondialdehyde (MDA) production decreased with improvement in lipid profile. In an in vitro study, platelet aggregability and the plasma level of von Willebrand factor (vWF) and F.VIIIR:AG of normal citrated blood were increased by passing it through a glass bead column. Combining above results of the three separate studies, it would be suggested that hyperlipidemia might enhance platelet activation in vivo, which occurred through contact of platelets to atherosclerotic rough vessel surface. The anti-platelet effect of simfibrate might be mediated through its effect on arachidonic pathway in platelets.

Immunoreactive somatostatin and vasoactive intestinal polypeptide in adrenal pheochromocytoma. An immunochemical and ultrastructural study.

An adrenal pheochromocytoma producing somatostatin (SRIF) and vasoactive intestinal polypeptide (VIP) in a 17-year-old boy is presented. High concentrations of immunoreactive (IR)-SRIF were found in plasma taken from the antecubital vein (31.0-33.0 pg/ml) and the inferior caval vein near the tumor (54.6 pg/ml), but after removal of the tumor the values became normal (11.0-15.2 pg/ml). In two portions of the resected tumor, considerable but different amounts of IR-SRIF (151.7 and 12.1 ng/g wet wt) and IR-VIP (13.0 and 5.5 ng/g wet wt) were demonstrated with size heterogeneities. Immunohistochemically, many IR-SRIF cells and a few IR-VIP cells were observed, but no cell reacting with both anti-SRIF and anti-VIP sera was found. Electronmicroscopically, many tumor cells had catecholamine-like granules (250-350 nm in diameter) while some others had VIP-like granules (110-140 nm in diameter). However, no granules resembling the SRIF granules seen in the pancreatic D cells were found. This seems to be the first report of an adrenal pheochromocytoma that produces SRIF and VIP simultaneously. It provides information on the histogenesis of hormone-producing neurogenic tumors.
