Antiviral effect of N-phenylacetoaminomethylene-DL-p-nitrophenylalanine (A-101) on murine leukemia viruses.

N-Phenylacetoaminomethylene-DL-p-nitrophenylalanine (A-101), when administered ip to male DDY mice infected with Friend leukemia virus, significantly inhibited virus-induced splenomegaly, reduced viable virus titers in spleen and plasma, and significantly prolonged survival time. A-101 also inhibited multiplication of the Friend and Moloney viruses in tissue culture systems.

"Pseudolysogenization" by RNA phage Q beta.

We isolated fairly stable lysogenic-like bacteria from a lysogenic state established between an amber mutant for the maturation protein gene of RNA phage Q beta (Q beta am 205) and its nonpermissive host BE110. These bacteria contained few mature phages intracellularly (less than 10(-3) plaque forming unit per cell), continued to grow with a potentiality to produce Q beta am 205 spontaneously, and showed an immunity-like response against homologous phage infection. These characteristics were maintained by growth in liquid medium containing anti-Q beta serum. We designated these cells as pseudolysogenic bacteria. The relative amounts of RNA genomes in these pseudolysogenic cells (about 10(2) infectious RNA strands per cell) indicated that the RNA genomes could replicate in nonpermissive cells and be distributed in daughter cells synchronizing well with cell division.

A treatment of detrusor-sphincter dyssynergia: pudendal nerve block by phenolglycerin under the guide of direct nerve stimulation.

While applying direct electrical stimulation to the pudendal nerve and recording of electromyographic reactions of bulbocavernosus muscle and/or transverse perineal muscle, pudendal nerve block by phenol was performed for the treatment of dysuria due to detrusor-sphincter dyssynergia, resulting in a favourable response. The technique and the cases treated with this nerve block are presented.

Congenital generalized lipodystrophy with insulin-resistant diabetes.

Hyperglycemia, glucose intolerance, hyperinsulinemia and resistance to exogenous insulin were found in a 10-year-old Japanese boy diagnosed as having congenital generalized lipodystrophy. Studies on insulin receptors of circulating mononuclear leucocytes indicated that insulin-resistant diabetes combined with congenital generalized lipodystrophy may be due to disturbance of insulin binding to membrane receptors. No insulin-binding antibody or antibody that impairs insulin-receptor binding was found. Plasma glucagon showed an exaggerated response to L-arginine before treatment. After treatment with a controlled diet and an oral sulfonylurea (500 mg/day) for 4 weeks, there was improvement in the plasma glucagon response to L-arginine. Improvement in the hyperglycemia, hyperinsulinemia and acanthosis nigricans was also observed. On the other hand, on completion of a 7-day high-fat diet, a marked increase in serum free fatty acids, triglycerides and beta-lipoproteins was observed. The total plasma post-heparin lipolytic activity during the high fat diet was within the normal range. However, the level of protamine-inactivated activity was 3 times that of the control.

Psychotropic agents. 3. 4-(4-Substituted piperidinyl)-1-(4-fluorophenyl)-1-butanones with potent neuroleptic activity.

A series of 1-(4-fluorophenyl)-4-(1-piperidinyl)-1-butanones substituted with benzimidazole, benzotriazole, or quinoxaline at the 4 position of the piperidine ring was synthesized and subjected to neuroleptic tests. Neuroleptic activities of several compounds were comparable to those of haloperidol. In particular, 4-[4-(2,3-dihydro-2-thioxo-1H-benzimidazol-1-yl)-1-piperidinyl]-1-(4-fluorophenyl)-1-butanone (10) was characterized by having a potent neuroleptic activity with less liability to the extrapyramidal side effect.

Effects of di-(2-ethylhexyl)phthalate administration on carbohydrate and fatty acid metabolism in rat liver.

1. Rats were fed on a diet containing 0, 1, 2, or 4% di-(2-ethylhexyl)phthalate (DEHP), a widely used plasticizer, for four weeks. 2. The level of plasma triglyceride was decreased in both the 1% DEHP and the 2% DEHP groups maintained for 1 to 2 weeks, but that of cholesterol was not changed. Plasma free fatty acid and ketone bodies increased in all DEHP groups. 3. The level of blood glucose was depressed to about 80% in the 2% and the 4% DEHP groups. Liver glycogen decreased markedly. Isotopic incorporations of 14C from [2-14C]pyruvate into blood glucose and liver glycogen were decreased. The changes in the contents of intermediates in the liver indicate that gluconeogenesis is inhibited at the reaction(s) between 3-phosphoglycerate and fructose 1,6-diphosphate. 4. Hepatic fatty acid synthesis increased about 2-fold on DEHP administration. The content of phospholipid in the liver was increased, whereas that of triglyceride was decreased. The rate of phospholipid synthesis was increased, but that of triglyceride was not changed.

Distribution of ribonucleic acid coliphages in south and east Asia.

We investigated the distribution of ribonucleic acid (RNA) coliphages in the Philippines, Singapore, Indonesia, India, and Thailand by collecting sewage samples from domestic drainage in November 1976. Of the 221 samples collected from domestic drainage, 50 contained RNA phages (52 strains). By serological analysis, 46 of the 52 strains were found to belong to group III. It can thus be said that the most prevalent RNA phages in Southeast Asia (at least, in the Philippines, Singapore, and Indonesia) were group III phages. Investigations of sewage samples collected from domestic drainage in Japan indicate that the most prevalent RNA phages in mainland Japan (north of Kyushu) are group II phages, whereas group III phages are predominant in the southern part of Japan (south of Amamiohshima Island). We therefore propose a borderline between Kyushu and Amamiohshima Island for the geographical distribution of RNA coliphages in the domestic drainage of South and East Asia. Moreover, one strain (ID2) was inactivated to some extent with the antisera of four groups of RNA phages. This is thought to be significant from the evolutionary viewpoint.

Surveillance and follow-up of patients with implanted cardiac pacemaker by telephone transmission.

A follow-up study on pacemaker function in 15 patients with implanted cardiac pacemaker has been performed by telephone transmission. The transmitting set consisting of electrocardiograph, pacemaker pulse modulator and acoustic coupler was manipulated by the patients themselves. ECG, pacemaker pulse, and pulse rate were simultaneously transmitted through 1 channel to the receiver in our clinic. Of 56 patients with pacemaker, 15 patients were surveyed by telephone transmission. In these patients battery exhaustion was detected in 3, competition in 2, and lead fracture in 1. The fact that surveillance and follow-up of the patients with implanted cardiac pacemaker could be carried out by telephone transmission as well as by clinic visitation tells us that the telephone transmission is a useful method and plays an important role in a pacemaker clinic. Moreover our study established its further usefulness in the patient's preference due to difficulties in travelling to the pacemaker clinic and in reducing patient's anxiety.

Effect of pyridoxal administration on the contents of pyridoxal phosphate and gamma-aminobutyric acid in mouse brain.

Many mice treated with a high single dose of PL died in convulsions. The convulsions were very similar to those induced with some anti B6 in several ways: (1) a long latent period occurs betfore convulsions; (2) PLP levels in brain decrease; (3) GABA levels in brain, especially in synaptosomal fraction, decrease. The possibility that a deranged GABA metabolism in nerve ending may be a major cause of the convulsions is discussed.
