Dermatoglyphs in duplication of the thumb.

The dermatoglyphs of 13 Japanese (eight males and five females) with duplication of the thumb were studied. The dermal patterns on the thumb were compared with those on the extra digit. A whorl on the thumb was associated with a whorl on the extra digit in two cases, a radial loop in four cases, and an arch in two cases. Where the thumb pattern was a loop the pattern on the extra digit was a loop in all three cases. Where the thumb and extra digit are both well developed the pattern tends to be the same on both digits. When the extra digit is poorly developed it tends to have a different pattern from the thumb.

Persistent infection of mouse tumor cells with mumps virus.

A mouse tumor cell line (MCT) persistently infected with mumps virus (strain Urabe) was obtained. No antibody or chemicals were required for establishment or maintenance of the infected cell line (M-MCT). No difference was observed by light microscopy between MCT and M-MCT cells, except for cytoplasmic inclusion bodies in 10--20% of the M-MCT cells. Hemadsorption of chicken erythrocytes, which did not necessarily coincide with inclusion positive cells was shown by 10--20% of the M-MCT cells. Immunofluorescence staining revealed that almost all M-MCT cells contained mumps virus antigens, which were seen mainly in the cytoplasm as fine granules, dots, or large clumps. Infectious mumps virus was consistently detected in the culture fluid and the released virus showed some temperature sensitivity when assayed at 34 degrees C and 40 degrees C. M-MCT was resistant to superinfection with homologous virus and showed some resistance to heterologous viruses. Thirty one clones of M-MCT were isolated by the soft agar method. Some, but not all, clones had viral antigen and all those with antigen released virus into the culture medium. The growth rates of MCT and M-MCT cells as monolayer cultures in vitro were similar, but the transplantability of M-MCT cells in syngeneic C57BL/6 mice was lower than that of MCT cells.

Cell kinetic analysis of the quiescent cell compartment in cultured renal carcinoma of Syrian hamster.

Using a culture line of renal carcinoma of Syrian hamster (HKC-400), the distribution of DNA content in individual cells was analyzed by autoradiography after pulse and continuous labeling with 3H-thymidine in order to investigate the behavior of the non-proliferative cell compartment. Additional data used were the cell cycle parameters obtained by the fraction of labeled mitoses (FLM) method and the growth rate. The principle of analysis is to compare the cell fractions having G1 and G2 DNA contents with the fractions of G1 and G2 phase cells. It was concluded that even in the exponential phase of growth 31% of cells were blocked in either the G1 or G2 DNA region (Q2 = 22% and Q2 = 9%, and the cell loss rate from Q1 was higher than from Q2.

Primary systemic amyloidosis. A case permitting pathological and biochemical investigations.

In this 72-year-old male patient, large amounts of amyloid substances were deposited in the mesenchymal organs, but not in the parenchymal organs. Histochemical and electron microscopic studies revealed large amounts of amyloid substances and amyloid fibrils in the skeletal and smooth muscle tissues. In the skeletal muscle tissues, where large amounts of amyloid substances were deposited, glycosaminoglycan was also markedly deposited, especially chondroitin-4-sulfuric acid. The concentrations of silicon were also markedly increased in the amyloid fibrils. The concentrations of nickel and cobalt were increased but those of zinc were decreased in all organs measured, although both nickel and cobalt in another case of amyloidosis experienced were within normal values.

Ultrastructure of cartilage in heritable disorders of connective tissue.

Ultrastruct of cartilage were examined in Marfan syndrome, Menkes kinky hair syndrome, achondroplasia, asphyxiating thoracic dysplasia, mild diastrophic dysplasia and mucopolysaccharidoses I and III. Ruthenium red staining revealed decrease of proteoglycans in cases with Marfan syndrome and kinky hair syndrome, and increase in cases with osteochondrodysplasia and mucopolysaccharidosis III. This morphologic tendency coincided with the result obtained by biochemical analysis of glycosaminoglycan contents in cartilage matrix from cases with Marfan syndrome (decreased content) and asphyxiating thoracic dysplasia (increased content). It was postulated that proteoglycan content in cartilage matrix might be related to excessive or reduced skeletal growth in Marfan syndrome or osteochondrodysplasia.

Proton correlation nuclear magnetic resonance study of anaerobic metabolism of Escherichia coli.

Proton correlation nuclear magnetic resonance has been used to investigate anaerobic metabolism of glucose in Escherichia coli cells. The time course of the concentrations of six metabolites (ethanol, lactate, acetate, pyruvate, succinate, and formate) has been followed at the very early state of fermentation, and used to discuss dynamical aspects of the mixed-acid fermentation of glucose by E. coli.

Metabolic studies of adrenoleukodystrophy.

Two series of metabolic studies were prompted by the previous finding that the brain and adrenal tissues of patients with adrenoleukodystrophy, an X-linked genetic disorder, contain unusually long-chain (C22--C32) fatty acids in cholesterol esters and gangliosides. Postmortem brain tissues from three patients were assayed for activities of the three distinct cholesterol ester hydrolases, using [4-14C]cholesterol oleate, lignocerate and cerotate as the substrates. No deficiency of the crude mitochrondrial (pH 4.2), the microsomal (pH 6.0), or the myelin-localized cholesterol ester hydrolases was detected, although the activities of the myelin-localized cholesterol ester hydrolase against cholesteryl lignocerate and cerotate were too low for reliable assays. The activities of the microsomal and myelin-localized hydrolases were actually higher in adrenoleukodystrophy than in controls. Uptake and exclusion by cultured fibroblasts of [1-14C]stearic, [1-14C]lignoceric and [1-14C)cerotic acids were also examined. All fatty acids were avidly taken up by the fibroblasts. Stearic acid was excluded from the cells much more rapidly than lignoceric or cerotic acid. No difference was observed in the uptake and exclusion of fatty acids between the controls and adrenoleukodystrophy, except that cells from some cases of adrenoleukodystrophy consistently took up the very long chain fatty acids at greater rates than the control cells. Neither did the distribution of the label among individual lipids reveal differences between the controls and adrenoleukodystrophy, although there were interesting and dramatic differences in the metabolism of lignoceric acid and cerotic acid. Cerotic acid appeared largely inert with 90--95% remaining intact over eight days, while lignoceric acid was mostly incorporated into complex lipids. This series of studies did not uncover the fundamental genetic defect underlying adrenoleukodystrophy.

Endolymphatic stromal myosis with autoradiographic observation.

A biopsy case of endolymphatic stromal myosis, a rare neoplasm of the uterus, which occurred in a 33-year-old female was described. The histology was typical and resembled stromal cells of proliferative phase endometrium. The tumor extended in the endometrium and diffusely infiltrated into the myometrium and the parametrium. However, 3H-thymidine labeling index was low suggesting slow growth of the tumor.

Atypical GM1 ganglioside accumulation in a case of juvenile amaurotic idiocy.

The brain and liver from a 7-year-old Japanese girl with juvenile amaurotic idiocy were examined neuropathologically and biochemically. Visceromegaly and skeletal abnormalities were absent. Nerve cells in the central nervous system were swollen and contained fine fat granules. Electronmicroscopically, there were large numbers of irregular bodies in the perikarya and these corresponded to the curvilinear and membranous cytoplasmic bodies. Lipid analysis of the brain revealed that GM1 ganglioside was increased in the parietal and occipital areas, while the frontal lobe showed a normal ganglioside pattern. N-Acetyl neuraminic acid (NANA) content in all areas was not elevated. Determinations of beta-galactosidase activity were within normal ranges. The liver had no accumulation of GM1 ganglioside and showed a normal beta-galactosidase activity. These unusual findings in GM1 gangliosidosis were discussed.
