Induction of deoxypyrimidine kinase activity in human embryonic lung cells infected with varicella-zoster virus.

Deoxypyrimidine kinase (deoxythymidine [TdR] kinase and deoxycytidine kinase) activity was induced in human embryonic lung cells after infection with varicella-zoster virus (VZ virus). Increased enzyme activity was also produced by using cell-associated virus as inoculum instead of cell-free virus. Anti-VZ virus serum inhibited both the appearance of cytopathic effect and the induction of enzyme activity. The induced TdR kinase activity was more thermostable than that induced by herpes simplex virus type 1. Also, the TdR kinase activity of VZ virus-infected cells was inhibited by dTTP less than in mock-infected cells and more than in herpes simplex virus type 1-infected cells.

Compliance of the peripheral capacitance vessels in patients with congestive heart failure.

A method for measuring compliance of the peripheral capacitance vessels is reported, and the values of compliance in patients with congestive heart failure are compared with those in normal subjects. Peripheral venous compliance and right brachial venous pressure in both patients with congestive heart failure and in normal subjects, when plotted graphically, showed a relationship described by many hyperbolic curves. The mean value of compliance of the peripheral capacitance vessels in patients with congestive heart failure was only half that in normal subjects. This difference was statistically significant (P less than 0.001). After the congestion in the patients with congestive heart failure disappeared following treatment with digitalis and other drugs, the values of compliance became nearly the same as in normal subjects. It seems that the increase of compliance of the peripheral capacitance vessels is not due to an organic change but to a functional change of the vessels caused by distention.

A case of acute anterolateral infarction complicated with "left posterior hemiblock".

A case of transient left posterior hemiblock and various intraventricular conduction disturbances associated with acute anterolateral infarction is reported. The earliest manifestation was an appearance of ventricular premature systoles originated in the area of the left posterior fascicle. On following recordings left posterior hemiblock with periods of high grade A-V block developed. A few minutes later regular alternation of left posterior hemiblock and left anterior hemiblock was recorded in the presence of 2:1 A-V block. The mechanism for such rare manifestation is analysed. After representing complete left bundle branch block, spontaneous recovery of normal conduction was obtained without any change in heart rate.

Deoxycytidine kinase from rabbit kidney cells infected with herpes simplex virus type 1 or 2.

When rabbit kidney cells were infected with herpes simplex virus type 1 (strain Seibert) or herpes simplex virus type 2 (strain 316D), deoxycytidine kinase (CdR kinase) activity, assayed at 38 degrees, increased 5- to 15-fold relative to controls. The CdR kinase activity induced by type 2 virus was more thermolabile than the enzyme activity induced by type 1 virus. When CdR kinase activity was assayed at various temperatures between 0.5 and 38 degrees, maximum activity for type 1 enzyme was obtained at 16 degrees while maximum activities for host and type 2 enzymes were obtained at 38 degrees. Both type 1 and type 2 induced CdR kinase activities eluted at the same positions as deoxythymidine kinase activities on a Sephadex G-100 column. The estimated mol wt for HSV-1 (Seibert) and HSV-2 (316D) induced CdR kinases are 67,000 and 60,000, respectively.

Studies on live rubella vaccine. VI. Seroepidemiological surveillance and immunization of adolescents in high school.

The rubella seroimmunity status of a total of 1,204 students aged 12 to 19 in a junior and a senior high school in Osaka district was surveyed. Among these, 487 students (40.5%) were found to be seronegative (less than 1 : 8) by the hemagglutination inhibition (HI) test. A total of 287 students were immunized with live rubella vaccine, Biken lot No.7233. This caused an increased titer in all except one of the 262 seronegative students, while among 25 students with an initial HI antibody titer of 1 : 8 it caused more than 4-fold increase in 20 and 2-fold increase in 5. The vaccine caused no clinical manifestations, such as fever, rash or arthralgia.

A new method for measurement of right ventricular residual ratio using 121I-MAA.

A new technique of precordial counting with 131I-MAA for measurement of right ventricular residual ratio (RVRR) was described. 131I-MAA was rapidly injected injected into the right ventricle at the time of right heart catheterization. The fraction of isotope discharged from this chamber per beat was determined with a crystal detector with tapered collimator which was pointed to the center of right ventricle. The radioisotope dilution curve was corrected by using the lung built up curve recorded by the other detector pointed to the lung field. Then, the right ventricular end-systolic volume (ESV) and the right ventricular end-diastolic volume (EDV) were calculated from the RVRR and stroke volume. In 6 patients with normal hemodynamics, the RVRR averaged 57.8 plus or minus 4.9%, the ESV 52.2 plus or minus 13.6 ml/M(2) and the EDV 89.4 plus or minus 15.1 ml/M(2). In 16 patients with heart diseases, the RVRR averaged 60.6 plus or minus 7.1%, the ESV 80.6 plus or minus 34.0 ml/M(2) and the EDV 127.2 plus or minus 43.6 ml/M(2). In 11 patients with chronic pulmonary diseases, the RVRR averaged 70.7 plus or minus 9.9%, the ESV 94.7 plus or minus 42.4 ml/M(2) and the EDV 133.0 plus or minus 46.5 ml/M(2). The RVRR in the last group increased significantly.

Studies on live rubella vaccine. V. Quantitative aspects of interference between rubella, measles and mumps viruses in their trivalent vaccine.

Rubella vaccine combined with measles and mumps vaccines was administered in a single injection to children of 1 to 5 years of age. All three vaccines were serologically effective, and the clinical reactions caused by measles vaccine were considerably alleviated, when 6 x 10(3) PFU of rubella and 10(4) TCD50 per dose of mumps vaccines were combined with 5 x 10(4) TCD50 of measles vaccine. When a larger amount of mumps vaccine (3 x 10(5) TCD50/dose) was used, it caused interference with the rubella and measles viruses, i.e., the antibody response to rubella virus became poor and the incidence of clinical reactions to measles virus decreased. On the other hand, when 5 x 10(5) TCD50/dose of measles vaccine was combined with 10(4) TCD50/dose of mumps vaccine, the clinical reactions to measles virus were decreased but were almost the same as those induced by this vaccine alone.
