Actin organization in migrating corneal epithelium of rabbits in situ.

We have found previously that fibronectin enhances the migration of rabbit corneal epithelium both in vitro and in vivo. In this paper we report a change of actin localization in migrating corneal epithelium as determined by immunofluorescent microscopy. Rabbit cornea was cut into small blocks and cultured in TC-199 medium. In the normal cornea, actin was detected as diffuse fluorescence at each epithelial layer. After 8 hr of cultivation epithelial cells had not started to migrate significantly, but actin had accumulated at the cell membrane. After 24 hr, epithelial migration had begun, and actin-specific fluorescence was detected mainly in the basal cell layer at the leading edge. When fibronectin or epidermal growth factor was added to the culture medium, epithelial migration began 8 hr after initiation of culture, and at 24 hr actin-specific fluorescence at the basal side of the migrating epithelial cells appeared stronger than that of a control group cultured in TC-199 unsupplemented medium. At the same time, fibronectin-specific fluorescence was more intense beneath the migrating epithelial cells. It is known that fibronectin has an affinity to collagen, and thus it might coat the cut surface of the stroma. Epithelial cells may attach then to the stroma via coated fibronectin. When a large quantity of exogenous fibronectin is added or when fibronectin is synthesized by the addition of epidermal growth factor, it may further stimulate the organization of intracellular actin from globular form (G-actin) to fibrilar form (F-actin). As a result, the change of intracellular localization and appearance of organized actin molecule might lead to cellular migration.

Clinical evaluation of fibronectin eyedrops on epithelial disorders after herpetic keratitis.

The clinical efficacy was investigated of fibronectin eyedrops on the nonhealing corneal epithelial defect after herpetic keratitis. Fibronectin eyedrops were prepared from the patient's own blood plasma by an affinity chromatography using gelatin-coupled agarose gel and by gel filtration technique. Twenty eyes with metaherpetic trophic ulcer with persistent epithelial defect that had not responded to conventional therapy were treated by the instillation of fibronectin eyedrops 6 times a day. The mean +/- standard deviation duration of epithelial defect was 61.5 +/- 66.1 days. The epithelial defects healed within 15.6 +/- 12.4 days. These results suggest that fibronectin eyedrops are a possible effective therapeutic approach in nonhealing corneal epithelial defect after herpetic keratitis.

Relationship between the concentration of myoglobin and parvalbumin in various types of muscle tissues from chickens.

The concentration of parvalbumin was determined in various types of chicken muscle by immunological analysis and was compared with that of myoglobin. Parvalbumin was present specifically in skeletal muscle and absent in cardiac and gizzard muscle; exceptionally, neither parvalbumin nor myoglobin was detected in white breast muscle. The wing and leg red muscles, which had larger amounts of myoglobin, contained smaller quantities of parvalbumin. In these muscles, the concentration of parvalbumin was inversely related to that of myoglobin (correlation coefficient = -0.69). Both myoglobin and parvalbumin were observed in the legs of 18-d-old embryos; the parvalbumin content exceeded that of myoglobin until the birds were 4 to 6 weeks old, but the relationship was reversed thereafter. Myoglobin in gizzard muscle was present in 18-d-old embryos and increased markedly at hatching; it was already present in cardiac muscle at an early embryonic stage, increasing gradually until 14 weeks after hatching.

Correlation between cellular ATP level and bile excretion in the rat liver.

The influence of the cellular level of adenosine triphosphate (ATP) in the liver on bile excretion was studied in rats. In ischemia, the cellular ATP level decreased rapidly--and, concomitantly, bile flow stopped within 5 min. Administration of L-ethionine i.p. to rats reduced the bile flow rate with decrease in the cellular ATP level. The correlation between the bile flow rate and the cellular ATP level was confirmed in a liver perfusion system. On anoxic perfusion, the ATP level and bile flow rate changed in the same manner as in ischemia. The recovery rates of both on reoxygenation decreased with increase in the anoxic perfusion period. During perfusion under oxygenated conditions, decrease in cellular ATP to various levels by infusion of various concentrations of potassium cyanide, an inhibitor of respiration, resulted in corresponding and concomitant suppression of bile excretion. Kinetic analysis of the bile flow rate revealed a Michaelis-Menten-type curve for the cellular ATP level. The apparent Kms for ATP of bile flow rate in L-ethionine-treated rat liver and liver perfused with potassium cyanide were 1.0 and 1.6 mM, and their Vmax values were 4.1 and 2.5 microliter/min/g liver, respectively. The concentrations of main bile components, such as phospholipids, cholesterol, and taurocholate increased, but their total outputs decreased with decrease in the ATP level, and returned to the normal range with recovery of the ATP level. Thus, it was shown experimentally that the extent of hepatic injury can be assessed simply by monitoring the bile flow rate, which reflects the cellular level of ATP.

Phagocytosis of spermatozoa and latex beads by epithelial cells of the ampulla vasis deferentis of the rabbit: a combined SEM and TEM study.

The fine structure in the ampullary region of the vas deferens of the rabbit was observed by scanning and transmission electron microscopy, with emphasis on the occurrence of epithelial spermiophagy. The epithelial cells were cuboidal or low columnar and contained moderately developed organelles. These cells, like luminal macrophages, were found to be not only involved in spermiophagy but also capable of actively taking up latex beads administrated intraluminally. Taking into consideration the previous findings in some other species, the epithelial spermiophagy seems to be a common event in the vas deferens of mammals.

Phagocytosis of spermatozoa and latex beads by the epithelial cell of the cat oviduct: combined SEM and TEM study.

The adult cat oviduct was viewed by SEM and TEM as spermatozoa of the same species and inert latex beads were injected into its lumen. The present study demonstrates that nonciliated epithelial cells in extensive areas of the oviduct as well as luminal macrophages are capable of actively taking up both the spermatozoa and the latex beads in the manner of phagocytosis. The epithelial cells are thus able to eliminate foreign bodies including degenerating spermatozoa in order to scavenge the lumen, though the cellular mechanisms involved in their phagocytosic activity remain to be elucidated.

[Four cases of cystitis induced by the anti-allergic drug tranilast].

First, the cases of two patients with intractable cystitis are presented. The first case was a 72-year old man who was admitted to our clinic with bladder symptoms (pollakisuria, pain on urination, hematuria and so forth) for about one month. These symptoms were not relieved by several kinds of antibiotic therapy. For the previous three months he had been suffering from asthma bronchiale. Rizaben (tranilast) was administered for more than two months. An excretory urogram showed normal renal function, but a small bladder with trabeculation. Voiding cystourethrograms revealed bilateral VUR. Cystoscopy disclosed remarkable red areas in the bladder mucosa. A bladder biopsy was done and the pathological finding was cystitis chronica. Intravesical installation of AgNO3, steroid administration, anti-histamic and anti-allergic therapy did not resolve these symptoms. However, Chinese medicines (Chorêto and Ryutanshakantõ) were effective. The second case was a 39-year old woman who visited our clinic complaining of bladder symptoms with hematuria for more than two months. In the past, she had suffered from asthma bronchiale. Rizaben (tranilast) was administered for three months. On cystoscopic examination, a small amount of intravesical coagula and erythematous, edematous areas with petechiae, were observed. The intravesical installation of AgNO3, anti-histamic and antiallergic therapy and gamma-globulin were not effective, but Chinese medicine was effective. Two other cases of cystitis due to Rizaben are presented briefly. Including our cases, 24 cases of cystitis chronica or eosimophilic cystitis due to Rizaben have been reported recently.

[A case of male breast cancer with special reference to the hormonal environment during chemoendocrine therapy].

Chemoendocrine therapy was performed on a man with advanced breast cancer, and partial response was observed for 11 months. Estrogen receptor in cancer tissue was detected before and after the therapy. Progesterone receptor, however, was not detected after the treatment, whether it had been there or not prior to it. The serum estrone level was continuously high during the treatment, but the serum testosterone level was obviously decreased after that.

Intrasplenic grafting of 7,12-dimethylbenz (a) anthracene-treated ovarian piece.

Immediately after a direct application of a chemical carcinogen, 7,12-dimethylbenz (a) anthracene (DMBA), the ovarian piece containing DMBA was implanted into the spleen of Wistar strain rats with or without gonadectomy. Six months after these treatments, an intrasplenic neoplasm was found in four of nine gonadectomized rats and in two of ten rats without castration. The volume of the induced tumor in the former group was 1.85 cm3 in average and that in the latter was 0.25 cm3. Histologic examination indicated that all of the induced tumors were not in the sex cord-stromal category, but adenocarcinomas. No morphological differences between the tumors in the castrated group and those in the noncastrated group could be detected by light microscopy. From the difference in the volume and the incidence of the induced tumors in the two groups, it appears that the hyperstimulation of the pituitary gonadotropins in the castrated rats promoted the tumorigenic process of the induced 'epithelial' ovarian cancer caused by the DMBA application to the grafted ovarian pieces.

Scanning and transmission electron microscopic evidence of epithelial phagocytosis of spermatozoa in the terminal region of the vas deferens of the cat.

Scanning and transmission electron microscopic observations have been made in the terminal region of the vas deferens of the cat, with emphasis on the occurrence of spermiophagy. The present study has revealed that epithelial cells as well as luminal macrophages are extensively and actively involved in phagocytosis of spermatozoa. The mechanism of the spermiophagy is discussed, in relation to a possible role of the epithelial cells, as one function of the vas deferens.

Peripheral K-lymphocyte population in head and neck cancer.

Peripheral K-lymphocytes in head and neck cancer were measured by assay of plaque forming-cells. In the cancer patients, the percentage (4.37 +/- 0.87%, mean +/- SD, N = 42, P less than 0.01) and absolute counts (68 +/- 28/mm3, P less than 0.01) of K-lymphocytes were significantly lower than those in normal controls (8.04 +/- 1.41%; 175 +/- 53/mm3, N = 29). The untreated group showed decreased K-lymphocyte counts (63 +/- 22/mm3, N = 13) as compared with the treated, disease-free group (83 +/- 27/mm3, N = 13, P less than 0.05). There were significant correlations between absolute counts of K-lymphocytes and T-cells or B-cells within the untreated group (r = 0.79 in T-cells, P less than 0.01; r = 0.64 in B cells, P less than 0.01). Moreover, absolute counts and percentage of K-lymphocytes in the patients having regional lymph node metastasis (58 +/- 25/mm3; 4.14 +/- 0.77%, N = 22) were significantly lower than those in the negative node group (80 +/- 26/mm3, P less than 0.01; 4.63 +/- 0.91%, P less than 0.05; N = 20). In a total of 25 patients with squamous cell carcinoma who were grouped into grade I, II, and III according to classification of the histologic differentiation of the World Health Organization, the absolute counts and percentage of K-lymphocytes in the grade I group (93 +/- 33/mm3; 5.14 +/- 1.08%, N = 7) showed significant increases in comparison to those in the grade II group (62 +/- 19/mm3, P less than 0.02; 4.06 +/- 0.62%, P less than 0.02, N = 14). Moreover, the change of the K-lymphocyte population in the treated, disease-free eight patients revealed a gradual increase of K-lymphocytes. These results led us to suggest that the measurement of peripheral K-lymphocytes is a useful method of characterizing host defense in head and neck cancer.

Fibronectin enhancement of corneal epithelial wound healing of rabbits in vivo.

The effect of topical administration of fibronectin on corneal reepithelialization in vivo was investigated in rabbits. Rabbit plasma fibronectin was purified by affinity chromatography using gelatin-coupled agarose gel (Sepharose 4B). The concentration of purified fibronectin was adjusted to 0.75 mg/mL. Corneal epithelial cells were removed by iodine vapor in 13 rabbits (26 eyes), and the size of the epithelial defect was calculated from the printed photographs in each eye. Fibronectin or phosphate-buffered saline eye drops were instilled. Healing rate was calculated by linear regression. The mean epithelial healing rate in fibronectin-treated eyes was 1.62 +/- 0.33 sq mm/hr, whereas in the control eyes it was 1.30 +/- 0.35 sq mm/hr. This study shows that fibronectin facilitates corneal epithelial wound healing in vivo.

Peripheral T gamma lymphocyte population in head and neck cancer.

Peripheral T gamma lymphocytes were measured in head and neck cancer patients and controls. The percentage was significantly higher in the 59 cancer patients than in the 46 normal controls (P less than 0.001). The 12 patients with recurrent disease had elevated percentages of T gamma lymphocytes compared with the untreated group (n = 31; P less than 0.05) and the treated, disease-free group (n = 16; P less than 0.05). Moreover, the percentage of T gamma lymphocytes was significantly higher in the 31 patients with regional lymph node metastasis than in the node-negative group (n = 28; P less than 0.05). In a total of 37 patients with squamous cell carcinoma histologically graded I, II, and III, the absolute counts and percentages of T gamma lymphocytes in the grade I group (n = 13) showed significant decreases compared with those in the grade III group (P less than 0.05; n = 6). Moreover, postoperative serial determinations of the percentage of T gamma lymphocytes in the 14 treated, disease-free patients revealed a gradual decrease of T gamma lymphocytes, whereas the five patients with recurrent disease had a tendency to increases in the percentage of T gamma lymphocytes.

Effects of dietary butter fat on fecal bile acid excretion in patients with Crohn's disease on elemental diet.

The excretion rate of fecal bile acids was determined by gas-liquid chromatography in nine patients with Crohn's disease and six healthy volunteers under two dietary conditions with different amounts of fat: during nasoduodenal tube feeding with a fat-restricted elemental diet containing approximately 1.5 g/day of fat, and during the addition of 50 g/day of butterfat by peroral administration. The fecal bile acid excretion rate on the elemental diet was significantly greater in the patients with Crohn's disease than in the healthy controls. With ingestion of the additional fat, the excretion rate was significantly increased in the patients with Crohn's disease, but not significantly changed in the healthy controls. The bile acid excretion rate in Crohn's disease correlated with fecal fat excretion, but not with either fecal weight or disease activity. These studies show that the amount of dietary fat represents an important consideration in the evaluation of bile acid malabsorption in Crohn's disease.
