Serological and functional analysis of the epitope clusters on Leu3/T4 antigen.

Monoclonal antibodies (MoAbs) were generated against cells or cell membrane glycoproteins of a human T-cell line, HPB-ALL. Five, designated MCN 1, 3, 12, 19, and 29, were found to be specific to helper/inducer T-cells; they gave a positive membrane staining to approximately 48% of peripheral blood lymphocytes and 84% of thymocytes and these proportions did not change upon costaining with Leu 3a, a known anti-helper/inducer T-cell MoAb. Furthermore, their reaction pattern with a panel of human lymphoid cell lines was identical to that of Leu 3a. A reciprocal binding blocking test showed that the epitopes reactive with the MCN MoAbs are divided into three separate clusters. The MCN 3- and Leu 3a-reactive epitopes formed a cluster and they appeared to be the same epitope. This cluster was well separated from that represented by the MCN 1-reactive epitope. The MCN 12-, 19-, and 29-reactive epitopes could be assigned to a third cluster. MCN 12 and 19 were probably toward the same epitope. A sequential binding test indicated that the three epitope clusters reside on the molecule carrying Leu 3a-defined epitope, i.e., the Leu3/T4 antigen. On the functional analysis, MCN 3 gave a profound inhibitory effect on T-cell proliferative response to MHC class II antigens, whereas other MCN MoAbs did not show any modifying effect on the T-cell function.

Improved method for the isolation and preliminary characterization of human DAF (decay-accelerating factor).

DAF (decay-accelerating factor) is one of the integral membrane proteins of erythrocytes, and is considered to play an important role in the regulation of complement activation. The purification of DAF has been impeded by the difficulty in removing glycophorin. We devised an effective method for removing glycophorin. Through the limited trypsinization of stromata prior to the extraction of DAF, glycophorin was readily digested so that the DAF could be purified free of glycophorin by DEAE-Sephacel and Bio-Gel A 0.5 m chromatographies. On SDS-PAGE, DAF from trypsinized stromata showed the same mobility as that from native stromata: its molecular weight was estimated to be about 70 kDa. Amino acid analysis of DAF showed high contents of serine and glutamic acid. The amino-terminal sequence of DAF prepared by the present method, determined for the 29 residues, did not show significant homology with that of glycophorin.

Further characterization of a human sperm coating antigen (gp12).

In our previous paper, we identified a novel sperm-coating antigen with molecular weight of approximately 12,000 daltons that is highly specific to sperm, seminal plasma, and milk from the plasma membrane fraction of human spermatozoa by using rabbit antiseminal plasma antiserum. In the present study, this 12,000-daltons component, termed gp12, has been investigated for its tissue distribution and antigenic stability. The largest amounts of the antigen are found in seminal plasma, although individual variation is rather high. In seminal plasma, the gp12 molecule presents in a large form with a molecular weight of approximately 50,000 daltons. Its antigenicity is stable when treated with acid, alkali, heat, and various protein denaturants.

A cDNA clone encoding a glycinin A1a subunit precursor of soybean.

A cDNA clone covering the whole coding region for a glycinin subunit precursor containing the A1a acidic subunit, one of the A2 family, has been identified from a library of soybean cotyledonary cDNA clones using a mixed oligonucleotide probe. Analysis of the cDNA insert revealed that it contained 1746 nucleotides of mRNA sequence with a 5'-terminal nontranslated region of 54 nucleotides, a signal peptide region corresponding to 19 amino acids, an acidic subunit region (A1a) corresponding to 291 amino acids followed by a basic subunit region corresponding to 185 amino acids, and a 3'-terminal nontranslated region of 207 nucleotides. By comparing the predicted protein sequence of this precursor with that of the legumin A precursor of pea, it was found that glycinin A2 subunit family appeared to be more closely related to the legumin than to the A3 subunit family, and that the evolutional rearrangement of glycinin genes has occurred.

Glycinin A5A4B3 mRNA: cDNA cloning and nucleotide sequencing of a splitting storage protein subunit of soybean.

The complete nucleotide sequence of a cloned cDNA, designated pGA5A4B3822, corresponding to glycinin A5A4B3 mRNA was determined. Analysis of the cDNA insert revealed that it contained 1899 nucleotides of mRNA sequence with a 5'-terminal non-translated region of 31 nucleotides, a signal peptide region corresponding to 23 amino acids, an acidic subunit region (A5) corresponding to 97 amino acids, an acidic subunit region (A4) corresponding to 257 amino acids followed by a basic subunit region (B3) corresponding to 185 amino acids, and a 3'-terminal non-translated region of 182 nucleotides. These results show that the glycinin A4 subunit, which is not found to be linked to a basic subunit via a disulfide bond, is synthesized as a full-sized precursor, i.e. the A5A4B3 subunit complex, from a single mRNA, followed by post-translational processing to generate an intermediary subunit complex (A5-B3), covalently linked by a disulfide bond, and the mature A4 subunit, which may associate with the above subunit complex by non-covalent interactions. From the results obtained by the Chou-Fasman rules we speculated that the two post-translational cleavage sites of this subunit precursor might be processed by the same proteolytic enzyme.

Self-induced photogenic epilepsy in infants.

We studied two infants with self-induced photogenic epilepsy and investigated their seizures with simultaneous EEG and videotape recording. A 2-year-old boy showed peculiar head-nodding behavior towards bright light before he manifested myoclonic absence or myoclonic seizures. A 14-month-old infant girl showed blinking in front of the television set before myoclonic jerks developed. Neither head nodding nor blinking was associated with paroxysmal discharges. We concluded that such preictal behavior was not part of the seizure.

Auditory brainstem response: a comparative study of ipsilateral versus contralateral recording in neurological disorders of children.

The auditory brain stem response (ABR) was studied in 103 normal and 90 neurologically abnormal children. The ABR was recorded simultaneously in Cz to the mastoid both ipsilateral and contralateral to the stimulated ear. A comparison between the ipsilateral and contralateral recording demonstrated that contralateral recording resulted in a small increase in the latency of peak V, whereas peak III showed a small latency reduction in the large majority of normal and neurologically abnormal children despite relatively large intersubject variability. There were some cases (cerebral palsy, infantile spasms) which demonstrated abnormal ABR only on contralateral recordings. Since most averaging systems have two channels, it is desirable to record the ABR simultaneously on bilateral recordings.

Aicardi syndrome. Report of 6 cases and a review of Japanese literature.

This report presents 6 cases of Aicardi syndrome (infantile spasms, agenesis of the corpus callosum, chorioretinopathy), and a review of Japanese literature. Two patients developed Lennox-Gastaut syndrome following infantile spasms. Visual evoked potential was examined in every case with binocular and monocular flash stimulations. Normal response was obtained in 5 cases, and no response in 1. Auditory brainstem response was examined in 2 cases. Central conduction time was prolonged in 1 case.

A human sperm coating antigen isolated from sperm cell membrane.

A human sperm cell membrane antigen that is highly specific to sperm and seminal plasma was isolated from plasma membrane fraction of spermatozoa using rabbit antiserum against human seminal plasma. In addition to the high specificity to sperm and seminal plasma, the isolated antigen showed the following characteristics: (1) It is a glycoprotein of approximately 12,000 daltons that has an affinity to lentil lectin; (2) it is distributed in human milk other than in sperm and seminal plasma, but is not found in any other organs and tissues including testis; (3) seminal plasma contains the largest amount of the antigen activity, 60-fold greater than spermatozoa and 900-fold greater than milk, suggesting that this antigen could be a sperm-coating seminal plasma antigen.

Predictors of long-term outcome of convulsive disorders in the first year of life: clinical usefulness of five risk factors.

The clinical usefulness of 5 risk factors (delayed development before the onset of seizures, abnormal EEG, symptomatic etiology, abnormal past histories, nonbrief symmetric generalized tonic and/or clonic convulsions) used to indicate long-term prognosis was investigated in 295 children who had convulsive disorders in the first year of life. All patients were followed up to the age of 6 years or older. Patients without any of these risk factors showed mentally and physically normal development, and 89% became seizure-free. However, only 10% or less of the high-risk group with 3 or more of these factors became mentally and physically normal. The score of these 5 factors was significantly related to the long-term prognosis and was assumed to be a valuable index to prognosis in the early stages.

EEG topography in porencephaly and arachnoid cyst.

With the topographic EEG mapping method, the correlation between the morphologic abnormality of the brain and its functional changes was investigated in 13 patients with porencephaly and arachnoid cyst verified by CT scan. The spatial distribution of the EEG activity was displayed in the delta, theta, alpha-1, alpha-2, beta-1, and beta-2 frequency bands. The abnormality of EEG topographic images (e.g., an increase of the power of delta activity and/or a decrease of the power of alpha activity in the involved side of the brain) was more marked in cases with porencephaly than those with arachnoid cyst, and in cases with mental retardation or paralysis than among those without.

[Syntheses of [prolyl-U-14C]alacepril and its related compounds].

In order to study the metabolic fate of alacepril, an anti-hypertensive agent, the 14C-labeled compound of alacepril and its related compounds were synthesized. [Prolyl-U-14C]alacepril was synthesized in over-all yield of 32.7-38.0% by the mixed anhydride condensation of L-phenylalanine with [prolyl-U-14C]DU-1163, which had been prepared from L-[U-14C]proline and N-(S-3-acetylthio-2-methylpropanoyloxy)succinimide. [Prolyl-U-14C]captopril and [prolyl-U-14C]DU-1227 were prepared in high yields by hydrolysis of [prolyl-U-14C]DU-1163 and [prolyl-U-14C]alacepril, respectively. [Prolyl-U-14C]captopril-cysteine was synthesized by condensation of [prolyl-U-14C]captopril with cystine S-monoxide in 55.0% yield.
