[Sperm-immobilizing antibodies in serum, follicular fluid and ascites].

Antisperm antibodies appear to impair reproduction. Follicular fluids, ascites and sera taken at the time of oocyte recovery from patients undergoing gamete intrafallopian transfer (GIFT) were examined for the presence of sperm immobilizing antibodies and their immunoglobulin classes were determined. Three out of ten patients were found to have sperm immobilizing antibodies in their sera. The immunoglobulin classes of the antibodies were IgG and/or IgM, and no IgA class antibodies were detected. The same classes of antibodies found in sera also existed in follicular fluids and ascites in these patients. Furthermore, titers of the antibodies in follicular fluids and ascites were consistent with those of patients' sera.

[Identification and characterization of a human sperm coating antigen isolated from sperm cell membrane].

An antigenic spermatozoal polypeptide was identified from nonionic detergent solubilized membrane material of sperm cells by using a human seminal plasma specific rabbit antiserum. The radiolabeled sperm cell membrane preparation was subjected to gel filtration and gel fractions were assayed for the binding activity with antiseminal plasma antiserum. The immune complex of the peak fraction of the binding was applied on SDS-PAGE, and the antigen was estimated to be a single polypeptide of approximately 12,000 daltons. The inhibition assay of the binding revealed that the antigen is present in sperm, seminal plasma, and milk, but is absent from any other organs and tissues except male genital tracts. Seminal plasma showed the largest amount of the activity, suggesting that this antigen is a sperm coating antigen. In seminal plasma, the antigen molecule present in a large form with a molecular weight of approximately 50,000 daltons. The antigenicity was stable when treated with heat, acid, alkali and various protein denaturants. The characteristics of the present antigen are very similar to those of lactoferrin and ferrisplan, the other sperm coating antigens common to milk. However, they are quite different in molecular weight, and no cross reactivity was found between the present antigen and lactoferrin or ferrisplan.

Visual seizures in children.

Twenty-one children (11 boys, 10 girls) suffering from visual seizures were studied. The visual seizures manifested as loss of vision, flickering light, blurring or figurative hallucinations. Ictal EEGs during visual seizures were recorded in 4 cases; the ictal discharges all originated in the occipital area. Visual seizures appeared in association with two or more non-visual ictal signs. In 18 cases, interictal EEGs revealed epileptiform discharges in the occipital area. In 3 cases, all girls, interictal EEGs showed photoconvulsive responses and focal spikes outside the occipital areas. They experienced clinical seizures when watching television and were thought to have photosensitive partial seizures.

Long-term prognosis of tuberous sclerosis with epilepsy in children.

A follow-up study was performed on 40 children with tuberous sclerosis and epilepsy. In 68% of the patients with infantile spasms and 73% of those without them, complex partial seizures were observed. Thus, patients with tuberous sclerosis are subject to not only infantile spasms but also complex partial seizures. Patients with infantile spasms showed mental retardation more often than those without. Seizures which had evolved from infantile spasms were more difficult to control than those which had not. The prognosis of infantile spasms associated with tuberous sclerosis was better than that of prenatal group other than tuberous sclerosis. Forty percent of the 40 cases showed asymmetry on EEG, and the asymmetry tended to disappear as the patients grew up.

Synthesis and structure-activity relationships of 1-substituted 4-(1,2-diphenylethyl)piperazine derivatives having narcotic agonist and antagonist activity.

Racemates and enantiomers of 1-substituted 4-[2-(3-hydroxyphenyl)-1-phenylethyl]piperazine derivatives (3-18) were synthesized, and their analgesic and other pharmacological activities and structure-activity relationships were investigated. The S-(+) enantiomers of 2a, 5, 7, 9, 10, and 15-18 had a stronger analgesic activity than their R-(-) enantiomers; analgesic activity of the strongest one [(S)-(+)-10] was 105 times as potent as that of morphine. The S-(+) enantiomers of these compounds had the opposite configuration to that of morphine with respect to its (C-9) asymmetric center but the same configuration to that of the tyrosine residue of Met5-enkephalin. The R-(-) enantiomers of 16 and 18 showed narcotic antagonist activity, but the S-(+) enantiomers did not. (R)-(-)-18 had analgesic and narcotic antagonist activities comparable to pentazocine but showed no significant physical dependence liability. From these results, it is suggested that these compounds show an uncommon enantioselectivity in comparison with morphine and its surrogates, and belong to a new series of compounds having a potent analgesic activity.

Effect of a soluble factor secreted from cultured human trophoblast cells on in vitro lymphocyte reactions.

The immunoregulatory role of trophoblast cells in cell-mediated immunity was investigated. Trophoblast cells were obtained from 8-10-week human placentae by treatment with collagenase followed by differential centrifugation. The cells were cultured for 48 hr, and the culture supernatant was examined for immunosuppressive activity in vitro. The supernatant when added to cultures of peripheral blood lymphocytes from healthy donors suppressed both their reactivity to different lectins (PHA and PWM) and their activity in one-way mixed lymphocyte reaction. The degree of suppression was dose-dependent. Furthermore, the supernatant was able to reduce the natural killer cell activity against K562 target cells. On the other hand, the supernatant had no inhibitory effect on the effector phase of lymphocyte-mediated cytotoxicity activity against tumor cell lines RPMI 8866 and Daudi. In all cases, the suppression observed was not due to lymphocytotoxicity or tumor cell mortality. The results indicate that trophoblast cells release a soluble suppressive factor that is a potent inhibitor of cell-mediated immunity.

Complex partial seizures in children: ictal manifestations and their relation to clinical course.

We analyzed complex partial seizures in 38 children aged 0 to 13 years, using simultaneous EEG-VTR recording. In infants, seizure duration was longer, automatisms were less purposeful and more common in the oral area, and convulsive movements were frequently seen and more extensive in comparison with older children. In ictal EEG, spiky components frequently appeared in infants. Parietal or occipital ictal foci were more often associated with mental or physical abnormalities than were frontal or central foci. Extensive convulsive movements were inversely proportional to seizure prognosis in older children. Interictal paroxysmal discharges were observed less frequently in infants.

Determination of ISF-2405, an active metabolite of cadralazine, in plasma and tissues by radioimmunoassay.

A radioimmunoassay has been developed which makes possible sensitive and specific determination of an active metabolite of cadralazine, (+/-)-6-[ethyl(2-hydroxypropyl)amino]-3-hydrazinopyridazine (ISF-2405), in plasma and tissues. On account of its lability in a sample solution, ISF-2405 in biological samples was selectively derivatized to a stable form, (+/-)-6-[ethyl(2-hydroxypropyl)amino]-3-(3,5-dimethyl-1-pyrazolyl) pyridazine (ISF-3349), by treatment with acetylacetone. The concentration of ISF-2405 was determined by the assay of the resultant ISF-3349. Antiserum against ISF-3349 was elicited in guinea pigs immunized with the ISF-3349 derivative coupled with bovine serum albumin. The obtained antiserum was highly specific for ISF-3349, and did not cross-react with either cadralazine or its metabolites. [Pyrazole-4-3H]labeled ISF-3349 with a specific activity of 13.9 Ci/mmol was used as the radioligand. Assays of ISF-2405 in plasma and tissues were possible over the concentration range from 0.4 to 6.4 ng/ml with 1 ml of plasma, and from 2 to 64 ng/g with 100 mg of tissue. Plasma and blood vessel levels of ISF-2405 in rats after single oral administration of cadralazine have also been determined by the present method.

Antitumor activity of streptococcal acid glycoprotein produced by Streptococcus pyogenes Su.

Streptococcal acid glycoprotein (SAGP) was purified from the cultured cells of Streptococcus pyogenes Su, and its in vitro and in vivo antitumor activities were investigated in comparison with those of OK-432, a cell preparation of S. pyogenes Su which is used clinically as a potent antitumor agent. SAGP inhibited the growth of several tumor cell lines in vitro at less than 0.1 microgram/ml, while it did not affect the growth of the other tumor and normal cell lines even at 10 micrograms/ml. This selective cytotoxicity is a unique characteristic of SAGP. OK-432 did not show cytotoxicity in vitro. SAGP also showed a considerable life-span-prolonging effect on mice bearing Meth A tumor and inhibited the growth of sarcoma 180 tumor implanted im. The comparison of antitumor activities between SAGP and OK-432 definitely suggested a difference in the mechanisms of their actions, even though they were derived from the same bacterial strain.

[ELISA for antisperm antibodies in unexplained infertile women].

It is commonly believed that autoimmune or isoimmune responses to human sperm antigens are associated with human infertility. We examined an enzyme linked immunosorbent assay (ELISA) kit used in the detection of antisperm antibodies in the sera of women with unexplained infertility. 1) For the ELISA assay, an absorbance at 405nm greater than 0.300 was considered positive. Of the 83 sera of infertile women studied, 20(24.1%) were positive. This value was significantly higher than that of the control group: 10% of pregnant women, 11.1% of unmarried women and 10% of healthy men (p less than 0.05). 2) The results obtained with ELISA were compared with those of the sperm immobilization test. Comparison of the results of ELISA with those of the sperm immobilization test indicated that these methods detect a different, though often overlapping, spectrum of antibody activity. 3) The results obtained with ELISA did not always correlate with those of the Huhner test. However the ELISA method may be useful in the clinical screening of antisperm antibodies because of its simplicity and rapidity.

Development of the beta activities in children: an EEG-topographical analysis.

We investigated the characteristics of beta activities during drowsiness and very light sleep in 61 children using topographical analysis. The average power of beta activities was posterior-dominant in infancy, and it became frontal-dominant as the age increased. These topographical changes occurred earlier in the low frequency bands than the high frequency bands. The total power of the high frequency bands was larger in infants, and with increasing age that of the low frequency bands predominated. The developmental changes of the beta activities considered to be induced by drugs were similar to those of children without drugs.
