Immunological studies on adult T cell leukaemia virus (ATLV) carriers.

Adult T cell leukaemia associated antibody (ATLA-Ab) positive people who were considered to be adult T cell leukaemia virus (ATLV) carriers were found in 0.75% of the adult population in the non-endemic area of Nagoya, Japan. Immunological studies on these people revealed that T lymphocyte subpopulations, as defined by nine monoclonal antibodies reactive to T lymphocyte surface antigens including T cell activation antigen, showed no differences between ATLA-Ab positive and ATLA-Ab negative individuals. Only a slightly higher percentage of Ia positive T lymphocytes was found in ATLA-Ab positive persons. Furthermore, the serum IgG level and the antibody titre of cytomegalovirus were significantly increased in ATLA-Ab positive individuals, while serum IgA, IgM level and the antibody titre of herpes simplex virus and mumps virus, showed no differences in both groups. This data suggests the possibility that ATLV carriers have some mild immunological abnormalities.

A long-term follow-up study on the efficacy of further attenuated live measles vaccine, Biken CAM vaccine.

Antibody persistence was measured in 39 children in an open community 12-13 years after immunization against measles with further attenuated live vaccine, Biken CAM. Serum samples of the children taken every two or three years after vaccination had higher, lower, or the same HI antibody titers as those in samples taken 6 weeks after vaccination. These differences reflected a decrease in the titer in some children and subclinical natural reinfection in others. However, all the children still retained detectable antibody in 12 or 13 years after vaccination, indicating long-term persistence of immunity after immunization with Biken CAM vaccine. For evaluation of the protective efficacy of the vaccine, matched controls were studied during the same period. Serological examination revealed that 97.5% of the controls were infected with measles and contracted the disease. In contrast, none of the vaccinees developed clinical infection after close contact with measles patients.

Measurement using tracers of steady-state turnover and metabolic clearance of insulin in dogs.

In nine conscious dogs, the steady-state metabolic clearance rate (MCR) and the systemic appearance rate (Ra) of insulin were determined by the tracer dilution method. [3H-PheB1]insulin ([3H]insulin) was infused as a tracer from time 0 at a constant rate. After tracer equilibration was attained, unlabeled porcine insulin was infused at variable constant rates (10.6-279 mU/min) with somatostatin (0.3 microgram . kg-1 . min-1) to suppress endogenous insulin secretion. Glucose was infused to prevent hypoglycemia. Tritiated and immunoreactive insulin (IRI) concentrations were determined in plasma samples after extraction on a C-18 reverse-phase column. Tracer-determined basal Ra of insulin was 2.39 +/- (SE) 0.61 mU/min. The calculated steady-state Ra of insulin for plasma IRI from 20 to 2,300 microU/ml showed good agreement with insulin infusion rates. The mean ratio of these rates was 0.973 +/- 0.018. The MCR of insulin under basal conditions was 29.9 +/- 3.4 ml . kg-1 . min-1, and it decreased with increasing insulin concentrations. It is concluded that 1) insulin turnover rates can be measured accurately using [3H]insulin as a tracer and 2) insulin kinetics are nonlinear.

Virus-induced diabetes mellitus: mengovirus infects pancreatic beta cells in strains of mice resistant to the diabetogenic effect of encephalomyocarditis virus.

In mice, Mengovirus produces a fatal encephalitis. Plaque purification of the virus resulted in the isolation of a clone (Mengo- 2T ), which in addition to encephalitis caused diabetes. Microscopic examination of pancreases from infected mice revealed necrosis in the islets of Langerhans and infiltration of inflammatory cells. By immunofluorescence viral antigens were found in the islets, and radioimmunoassays demonstrated a substantial decrease in pancreatic immunoreactive insulin. Studies on susceptibility among inbred strains of mice showed that whereas the D variant of encephalomyocarditis virus caused diabetes only in SJL/J mice, Mengo- 2T caused diabetes in strains of mice resistant to encephalomyocarditis-induced diabetes (i.e., CBA/J, C3H/HeJ, CE/J, AKR/J, C57BL/6J). The ability of Mengo- 2T to induce diabetes in encephalomyocarditis-resistant mice was found to be due to the greater capacity of Mengo- 2T as compared to the D variant of encephalomyocarditis virus to replicate in and destroy the islets of these animals. Although Mengo- 2T and the D variant of encephalomyocarditis virus are antigenically indistinguishable by hyperimmune sera, our studies show that these viruses have different host ranges and tissue tropisms .

The effects of an alpha-glucoside hydrolase inhibitor on glycemia and the absorption of sucrose in man determined using a tracer method.

Acarbose, an alpha-glucosidase inhibitor, lowers the glycemic excursion following the ingestion of carbohydrates, in particular, sucrose. This was confirmed with increasing doses of acarbose (0, 50, and 100 mg) and the causes investigated. The absorption of the glucose moiety of sucrose was determined from plasma tracer concentrations when overnight-fasted normal subjects received a 100-g oral sucrose load labeled with sucrose [(1-14C]glucose and a simultaneous intravenous infusion of [3-3H]glucose. As the dose of acarbose given with the sucrose load was increased from 0 to 100 mg, the percentage of the load appearing in the peripheral circulation decreased from 90% to 62%. Malabsorption was confirmed by the appearance of breath hydrogen. Simultaneously, absorption time increased from 243 to 411 min. Maximal glycemic excursions were therefore lowered from 64 to 31 mg/dl. The plasma concentrations of gastric inhibitory polypeptide and insulin decreased with the acarbose dose so that the fractional disappearance rate of glucose also decreased. However, the concentrations of glucagon-like immunoreactivity (GLI) rose, confirming the ileal appearance of malabsorbed sucrose.

Cysteinyldopa isomers and dopa in lesions and urine of Japanese patients with malignant melanoma.

Cysteinyldopas and Dopa in the urine and tissues of Japanese melanoma patients were investigated quantitatively by means of high-performance liquid chromatography. Cysteinyldopa isomers were detected in the urine of eumelanic Japanese patients. The amount (X +/- SD%) of each isomer of cysteinyldopa in the urine was 80.26 +/- 4.66% in 5-S-cysteinyldopa, 9.39 +/- 1.64% in 2-S-cysteinyldopa, 7.07 +/- 3.33% in 2, 5-S, S-dicysteinyldopa, and 3.28 +/- 1.43% in 6-S-cysteinyldopa. The amount of cysteinyldopa in melanoma tissues was 26-314 times more than that of Dopa. The amount (X +/- SD%) of cysteinyldopa in the tissues was 80.34 +/- 1.75% in 5-S-cysteinyldopa, 11.06 +/- 1.91% in 2-S-cysteinyldopa, 6.27 +/- 1.43% in 2, 5-S, S-dicysteinyldopa, and 2.34 +/- 0.61% in 6-S-cysteinyldopa. The fact that the percentages of each isomer of cysteinyldopa in the urine and in the tissues were approximately constant suggests that the cysteinyldopas secreted from melanoma cells were excreted into the urine without being metabolized.

Studies on amelanotic melanoma with the fluorescence method (Falck and Hillarp) and biochemical analysis of 5-S-cysteinyldopa in the tissues.

Histochemical findings of primary and metastatic amelanotic melanomas were shown by the formaldehyde-induced fluorescence method (Falck and Hillarp). All or some of the amelanotic melanoma cells were discovered to emit green specific fluorescence. Results of the determination of 5-S-cysteinyldopa and DOPA in amelanotic melanoma tissues indicated that the specific fluorescence emitted by these cells is primarily due to the presence of 5-S-cysteinyldopa. The values of 5-S-cysteinyldopa in these tissues were lower than those in melanotic melanoma, but were approximately the same as those in pigmented nevus. When unpigmented tumors were histopathologically revealed to be malignant, amelanotic melanoma could be definitely diagnosed by the fluorescence method of Falck and Hillarp and the biochemical analysis of 5-S-cysteinyldopa in the tissues.
