Study on digestibility and energy availability of daily food intake in Japanese (Part 3. Cereals).

Four male Japanese were fed a semisynthetic diet including egg and soy powder as protein source for seven days (Basal-diet period), and in the following seven days 200 g of polished rice, wheat flour and buckwheat flour added at the expense of part of the corn starch and sugar in the basal diets (Test-diet period). Urine and feces were collected throughout both periods and the contents of nitrogen, fat and energy in these excreta were determined. Digestibility of protein (N), fat and carbohydrate (by difference) was calculated. The protein digestibilities of the polished rice (in the form of cooked grains), wheat flour (in the form of cooked powder) and buckwheat flour were 89.6 +/- 5.0%, 93.4 +/- 2.9% and 85.1 +/- 2.5%, respectively. The fat digestibilities of the polished rice, wheat flour and buckwheat flour were 93.6 +/- 1.8%, 70.8 +/- 13.5% and 103.1 +/- 8.4%, respectively showing relatively large variation (This results may be caused by an errors in measurement). The carbohydrate digestibility was close to 100%. The net energy availabilities of the polished rice, wheat flour and buckwheat flour were 100.6 +/- 1.4%, 96.5 +/- 1.1% and 96.0 +/- 1.1%, respectively.

[Determination of Mg and Zn contents of Naruto "wakame" (Undaria pinnatifida)].

The contents of Mg and Zn in the sea-weed "wakame" were determined and compared at different growth stages, in different parts, for different root cultivation intervals, and in processed foods. The results obtained were as follows: 1. Mg (2.09-2.60 g/100 g) and Zn (8.00-10.76 mg/100 g) contents were independent of growth stage with the exception of Zn (29.00-35.78 mg/100 g) in the natural young "wakame". 2. The mineral contents of leaves, stipes, fruiting bodies and rhizoids of "wakame" at different growth stages were determined. The contents of Mg (1.00-2.36 g/100 g) and Zn (3.93-12.01 mg/100 g) were richer in leaves and stipes than in fruiting bodies and rhizoids. 3. Densely cultured "wakame" showed higher contents of Zn (6.73 mg/100 g) than thinly cultured plants (5.99 mg/100 g), and Mg content was not affected by changes in environmental conditions. 4. The Mg/Ca ratios reached a maximum in the stipe (3.3) and at fruiting bodies (2.9) in the middle stage, and in the base of the stipe of the mature plant (3.5). 5. Contents of the minerals in processed foods were determined. "Suboshi" was rich in Mg (1.07 +/- 0.42 g/100 g), and "Haiboshi" was rich in Zn (10.90 +/- 1.20 mg/100 g). The mineral contents in "Enzo" were small.

Practical debittering using model peptides and related compounds.

In order to develop a practical debittering method for amino acids and peptides, several debittering methods were studied. The authors found that hooking acidic amino acids to and acetylation of bitter amino acids is very effective to remove the bitterness from their concentrated solution. For debittering by mixing with additives, skin milk and other peptide compounds were effective. Acidic amino acids were also effective to reduce the bitterness. Gelatinized starch was found to be useful for debittering because it takes bitter substances into its net structure.

Mechanism for the bitter tasting potency of peptides using O-aminoacyl sugars as model compounds.

In order to study the role of hydrophobicity in bitter peptides, several O-aminoacyl sugars, in which amino acids or peptides were attached to the 2- and 3-position of methyl alpha-D-glucopyranoside, were synthesized and sensory analyses were carried out. It was found that the bitterness increased as the hydrophobicity of compounds increased, implying that the bitterness receptor recognizes the hydrophobicity of bitter peptides. A structure for the bitterness receptor is also discussed.

[Acute promyelocytic leukemia developed in the course of systemic lupus erythematosus: a case report].

A-36-year old Japanese female who had been suffered from systemic lupus erythematosus (SLE) and treated with prednisolone for 3.5 years developed de novo acute promyelocytic leukemia (APL) without preleukemic state. She had a short period of complete remission in leukemia but she died with recurrence of leukemia. While malignant tumors including lymphoid malignancy have been shown to develop occasionally in the course of SLE, acute myelogenous leukemia (AML) following SLE is a very rare condition. Although combination of SLE and AML may be incidental, review of the literatures revealed some interesting insight into the pathogenesis of SLE and AML.

[A combination treatment of transcatheter arterial embolization (TAE) and irradiation for hepatocellular carcinoma (HCC)--evaluation for therapeutic efficacy in comparison with TAE or irradiation alone].

Therapeutic effects of a combination therapy (TAE and irradiation) and a radiation therapy alone were evaluated comparatively in 54 patients with HCCs. Twenty eight patients of them underwent external X-irradiation with a linear accelerator after TAE and the remaining twenty six did irradiation alone. The incidence of partial response was 82.1% in the combination therapy and 69.2% in the radiation therapy. The cumulative rates of survival in the combination therapy were 70.8% (1-year), 37.1% (3-year), and 27.9% (5-year), which were higher than those in the radiation therapy. The cumulative rates of recurrence in the former were lower than those in the latter. The prognoses after the both treatments were poor in the patients with advanced liver cirrhosis (Child C). In the patients with HCCs smaller than 5 cm in diameter or accompanied by portal vein thrombi, the combination therapy was superior to TAE alone in the cumulative rates of survival. It is considered from the above results that the combination therapy is useful in the patients with HCCs when properly selected.

[Tetany as a sole manifestation in a patient with Bartter's syndrome and a successful treatment with indomethacin].

37 year-old man presented to our clinic because of tetany that occurred during driving a car in the morning. He had no anorexia, chronic diarrhea or vomiting. He was normotensive and no edema was noted. On neurological examination, he was rather apprehensive and Trousseau sign was mildly positive. No muscle weakness was noted except for the moderate atrophies of bilateral anterior tibial muscles as a sequela of Guillain-Barré syndrome that he suffered at the age of 7. The results of laboratory studies included hypokalemia, metabolic alkalosis, hyperuricemia, increased plasma renin activity, increased plasma aldosterone concentration, reduced pressor response to infusion of angiotensin II. Based on these clinical and laboratory findings, the diagnosis of Bartter's syndrome was made. As far as we know, it is extremely rare that Bartter's syndrome manifests tetany alone as in the present case. The exacerbation of preexisting alkalosis by hyperventilation during driving might be the cause of tetany. Tetany was easily provoked by hyperventilation, during which serum ionized calcium remained in normal level despite the exacerbation of alkalosis. The results suggest that alkalosis per se can cause tetany in Bartter's syndrome. Indomethacin, 75 mg daily, effectively prevented tetany from recurring for nearly 4 years without any side effect.

[Correlation between vibratory detection threshold and conduction study of sural nerve in diabetic patients].

Vibratory detection threshold (VDT) was determined on the plantar side of the distal phalanx of the right great toe of 22 diabetic (NIDDM) patients. In addition, a neurological examination and a sural nerve conduction study were performed. Peripheral neuropathy, based on our criteria, was found in 41% of the patients. VDT was abnormally high in 23% of the patients. The combined frequency of the abnormality of the amplitude of the action potential of the sural nerve or the conduction velocity or both was 73%. The nerve conduction study showed the highest sensitivity in detecting the abnormality of the peripheral nerve in this study; this is in agreement with the result shown in the literature. Seven patients showed no response to electrical stimulation of the sural nerve, although VDT was obtained in these patients. Among the 6 patients that underwent the normal sural nerve conduction study, no one showed abnormally high VDT. The determination of VDT seems to be a useful examination for the follow-up study of diabetic neuropathy, although it is less sensitive than the sural nerve conduction study.

Purification of NADH-ferricyanide dehydrogenase and NADH-quinone reductase from Escherichia coli membranes and their roles in the respiratory chain.

The respiratory chain-linked NADH-quinone reductase (NQR) and NADH-ferricyanide dehydrogenase (NFD) were extracted from membranes of Escherichia coli by n-dodecyl octaethyleneglycol monoether detergent and purified by DEAE-Sephacel, DEAE-5PW and Bio-Gel HTP column chromatography. The purified NQR contained FAD as a cofactor, catalyzed the reduction of ubiquinone-1 (Q1) and reacted with NADH, but not with deamino-NADH (d-NADH), with an apparent Km of 48 microM. On the other hand, the purified NFD contained FMN as a cofactor, reacted with both NADH and d-NADH, and catalyzed the reduction of ferricyanide but not Q1. NFD showed a high affinity for both NADH and d-NADH with a Km of 7-9 microM. NFD was inactivated, whereas NQR was rather activated, by preincubation with an electron donor in the absence of electron acceptor. These properties were compared with those of activities observed with inverted membrane vesicles with special reference to the generation of inside-positive membrane potential (delta psi). It was found that d-NADH-reactive FMN-containing NFD is a dehydrogenase part of energy-generating NADH-quinone reductase complex. The FAD-containing NQR was very similar to that purified by Jaworowski et al. (Biochemistry (1981) 20, 2041-2047), and reduced Q1 without generating delta psi.

[Study of multiregression analysis on NK activities and related clinical factors in patients with head and neck neoplasms].

NK activity in patients with head and neck neoplasms treated with radiation and chemotherapeutics was studied on the influence of 19 factors by using the method of multiregression analysis. Of these factors, BUN, WBC, the size of radiation portal, chemotherapy, radiation doses, total protein (T.P.), age and GOT were significantly influenced on the fluctuation of NK activity (p less than 0.05). In the studies of the size of radiation portal and its doses, NK activity showed a tendency to increase with radiation dose in small size of radiation portal and a tendency to reduce in large size of it.

A study of drug efficacy in the treatment of ovarian cancer.

One hundred and nine cases of common epithelial ovarian cancer treated in our department during the past decade were entered into this study, which dealt with direct therapeutic effects and prognosis, comparing the groups administered cisplatin with those administered other anticancer drugs. 1) In 38 patients with measurable lesions, high response rates were achieved through the administration of anticancer drugs containing cisplatin in both the primary cancer and recurrent cancer groups. 2) Preventive chemotherapy was performed in patients in whom complete surgery was achieved at the initial operation. No significant difference in prognosis was observed through the administration of cisplatin and other anticancer drugs. 3) In patients in whom reduction surgery was carried out to the maximum possible degree at the initial operation, an improvement in prognosis was observed through the administration of cisplatin. These results imply that a cisplatin-based regimen should be aggressively used, except in cases that underwent complete debulking surgery.

Study on digestibility and energy availability of daily food intake by the Japanese. Part 2. Horse mackerel and milk.

Four male Japanese students were fed a semisynthetic diet including rice and whole egg as protein for seven days (basal-diet period), and in the following seven days milk and then, for seven days, horse mackerel were added at the expense of part of the corn starch and sugar in the basal diet (test-diet period). Urine and feces were collected throughout all periods and the contents of protein, fat and energy in these extra were determined. The results obtained were as follows: 1) Digestibility of protein was 97.3 +/- 4.1% for milk and 96.8 +/- 2.8% for horse mackerel. 2) Digestibility of fat was 96.7 +/- 1.9% for milk and 98.6 +/- 2.6% for horse mackerel. 3) The ratio of the total available energy to intake energy (Net Energy Availability) was 92.5 +/- 2.9% for milk and 88.4 +/- 5.2% for horse mackerel.

Radiation therapy in Kimura's disease.

Kimura's disease is a rare disorder which predominantly involves the head and neck region and causes eosinophilia in peripheral blood. It often responds well to corticosteroid therapy but some patients can be resistant; in these patients symptomatic radiation therapy can be of value. We reviewed 10 patients with Kimura's disease who received radiation therapy from 1975 through 1981 in the Department of Radiology, Chiba University Hospital. Nineteen tumors were irradiated and 15 of them locally controlled. In 5 patients, steroid therapy could be withdrawn. For local control, 25 to 30 Gy seemed to be adequate.

[Fundamental study of 99mTc nanocolloids for inflammation imaging].

The biodistribution of nanometer-sized colloidal particles (nanocolloids, Solco-Basle Ltd.) was examined in six rabbits with turpentine-induced abscesses in the musculature of their hind legs. 99mTc labelled colloids with a mean particle size less than 100 nm were administered to each rabbit intravenously. The biodistribution of the tracer was studied with a gamma camera 1, 3, and 24 hours after injection. Prominent uptakes were demonstrated in the liver, spleen, bone marrow, kidneys, and urinary bladder. On the other hand, the abscess and surrounding inflammatory edema were depicted as unclearly margined faint hot spots. Subsequently, the rabbits were sacrificed and activity per gram tissues were counted with a well-type gamma counter. Although the results were almost in agreement with a gamma camera study, it was noticed the activity counts in the abscess were significantly higher than that of control muscles.

[Difference in susceptibility between auditory and equilibrium function in rabbits with experimentally-induced transient ischemic attack].

Patients with transient ischemic attack (TIA) in the vertebrobasilar artery more often complain of disturbance of equilibrium, such as vertigo or dizziness, than of auditory disorders, such as hearing impairment or tinnitus. The author induced TIA in rabbits by injecting adenosine diphosphate (ADP) into the right vertebral artery. Observations of rotatory nystagmus have shown that a peak level of directional preponderance appeared 1 minute after injection of ADP (0.3mg/kg b.w.) and continued for more than 4 minutes. On the other hand, the amplitude of auditory brainstem responses (ABRs) presented no significant changes following the injection of ADP (0.5mg/kg b.w.). However, greater amounts of ADP (1.0 and 2.0 mg/kg b.w.) were found to reduce the amplitude of ABR-waves, although significant reduction was observed for only less than 1 minute. This change was represented by flattening of the later part of the waves, with the first wave much less affected. Electrocochleogram (E. Coch. G.) have also demonstrated a transient reduction in amplitude 10 seconds after injection of ADP (4.0mg/kg b.w.) with correspondingly rapid recovery. In cases of hypertension due to noradrenaline load or trimethaphan-induced hypotension, reduced amplitudes were more profound and continued longer, returning to its normal range within 1 minute. Differences in susceptibility between the equilibrium system and auditory system in TIA of the vertebrobasilar artery are discussed.
