Effect of 1 alpha-hydroxycholecalciferol administration on Mg metabolism in sheep.

The effect of 1 alpha-hydroxycholecalciferol (1 alpha-OH-D3) administration on Mg metabolism in sheep was studied. Three wethers were intramuscularly injected with 0.2 microgram/kg body weight of 1 alpha-OH-D3 for 3 days. Plasma Mg concentration was decreased and plasma Ca concentration was increased by 1 alpha-OH-D3 administration. However, the duration of hypomagnesemia was shorter than that of the increment in the plasma Ca concentration. The administration of 1 alpha-OH-D3 increased apparent Ca absorption and urinary Ca excretion. However, apparent absorption and urinary excretion of Mg was not changed by 1 alpha-OH-D3 administration. The amount of glomerular filtrated Mg was reduced concomitantly with the reduction of plasma Mg concentration and glomerular filtration rate when 1 alpha-OH-D3 was injected. The tubular reabsorption of Mg tended to be suppressed and the ratio of reabsorbed to filtrated Mg was significantly lowered by 1 alpha-OH-D3 administration. These results suggested that the stimulation of urinary Mg excretion might not be one of the major factors inducing hypomagnesemia although 1 alpha-OH-D3 suppressed reabsorption of Mg in the renal tubule.

Effects of nutritional conditions on plasmid stability and production of tryptophan synthase by a recombinant Escherichia coli.

Effects of nutritional conditions and insertion direction of the tryptophan synthase (TSase) gene into a plasmid vector on the plasmid stability and the production of TSase in high cell concentration cultures were examined using recombinant Escherichia coli (E. coli K12 IFO 3301) harboring pBR322trpAB(1) (the TSase gene was inserted at the EcoRI site of pBR322 in the clockwise direction) and pBR322trpAB(2) (counterclockwise direction). As to the effects of the insertion direction, the cells harboring pBR322trpAB(2) were slightly lower in the growth rate and the plasmid stability than those harboring pBR322trpAB(1). However, the former was higher in the productivity of TSase and the final cell concentration attained than the latter. On the other hand, the addition of organic nutrients, especially yeast extract, to TK-25 medium was very effective to improve the plasmid stability. Among the components of yeast extract, L-glutamic acid was found to be effective to improve both the plasmid stability and the production of TSase. When 1 g/l of L-glutamic acid was added to TK-25 medium, a mineral synthetic medium developed for a high concentration culture, 115g (dry basis)/l of recombinant cells were obtained in 14 hr and the expression of TSase was maintained at 240-300 U/mg-protein during the cultivation.

In vivo changes of hemopoietic progenitors and the expression of the interleukin 5 gene in eosinophilic mice infected with Toxocara canis.

It has been demonstrated that purified recombinant interleukin 5 (rIL-5) supports the terminal differentiation and proliferation of eosinophilic precursors in vitro and plays an important role in increasing the functional activities of eosinophils. In this study, we examined the hemopoietic changes and analyzed murine (m) IL-5 mRNA expression in eosinophilic mice infected with the helminth Toxocara canis. In eosinophilic mice, eosinophils increased in number in both bone marrow and spleen. However, the number of eosinophilic precursors increased markedly in spleen cells of eosinophilic mice but remained relatively constant in the bone marrow. In the presence of granulocyte colony-stimulating factor (G-CSF), the number of granulocytic precursors increased in the spleen cells of eosinophilic mice. From these findings, the condition of eosinophilopoiesis in eosinophilic mice is accompanied by an increase in granulocyte-macrophage progenitors as well as eosinophil progenitors. Using Northern blot analysis, a weak but definite band corresponding to mIL-5 mRNA was detected in spleen cells of mice 4 and 5 days after helminthic infection. In addition, these data were confirmed by in vitro polymerase chain reaction (PCR) amplification of mRNA obtained from these spleen cells. Finally, injections of a monoclonal antibody against mIL-5 completely suppressed the blood eosinophilia in mice infected with T. canis. In conclusion, IL-5 is suggested to play a major role in eosinophilopoiesis in vivo.

Egg signals for triggering the acrosome reaction in starfish spermatozoa.

Upon encountering the jelly coat of an egg, starfish spermatozoa undergo the acrosome reaction. To induce the acrosome reaction, 3 jelly components act in concert on the spermatozoa: a sulphated glycoprotein named acrosome reaction-inducing substance (ARIS), a group of steroidal saponins named Co-ARIS, and an oligopeptide presumably having an ability to increase the intracellular pH of the spermatozoon. All three are required to mimic the full ability of jelly coat to induce the acrosome reaction instantaneously. A combination of ARIS and Co-ARIS is enough for induction in normal sea water, although its action is almost 2 min slower than the jelly. ARIS can induce the acrosome reaction by itself in high Ca2+ or high pH sea water. When spermatozoa meet the jelly coat, the acrosome reaction is eventually induced because ARIS and Co-ARIS co-operatively increase the intracellular Ca2+ by stimulating verapamil- and maitotoxin-sensitive Ca2+ channels and the oligopeptide increases the intracellular pH by stimulating Na+/H+ exchange systems.

[Evaluation of left ventricular mechanical energy efficiency and ventriculo-arterial coupling in humans using the conductance catheter technique].

To evaluate left ventricular mechanical energy efficiency and ventriculo-arterial coupling in humans, left ventricular pressure-volume relations were determined using the conductance catheter technique in 20 patients undergoing cardiac catheterization. The results were as follows: 1. A convex, curvilinear relationship was observed between end-systolic pressure-volume relations (Emax) and left ventricular ejection fraction (EF), as shown in the equation of EF = 28.5 x log (Emax) + 39.6 (r = 0.67, p less than 0.01, n = 20); EF remained nearly constant in the range of Emax greater than or equal to 4 mmHg/ml/m2, whereas, EF decreased markedly under the Emax of 4 mmHg/ml/m2. 2. A convex, curvilinear relationship was observed between Emax and mechanical energy efficiency (EW/PVA), as shown in the equation of EW/PVA = 30.5 x log (Emax) + 54.8 (r = 0.83, p less than 0.01, n = 16). 3. A concave, curvilinear relationship was observed between Emax and ventriculo-arterial coupling (Ea/Emax), as shown in the equation of Ea/Emax = -0.8 x (Emax)0.5 + 2.6 (r = -0.85, p less than 0.01, n = 16). Accordingly, EW/PVA was markedly decreased with changes in Emax in the range less than 4 mmHg/ml/m2, although it remained slightly increased above 4 mmHg/ml/m2. Ea/Emax was maintained constant (p less than 0.5) in the range of Emax above 4 mmHg/ml/m2 but was abruptly decreased when Emax was reduced below 4 mmHg/ml/m2. These results indicate that depressed left ventricle attempts to work effectively at the risk of mechanical energy efficiency and with suitable matching by aortic property. Application of pressure-volume relationships provides a new framework for evaluation and treatment of the failing heart.

[Resin adhesion on the primary ground enamel. (Report 1). Influence of the etching times].

The purpose of this study was to investigate the influence of the etching times on the adhesion of the resion on the primary ground enamel. Fifty extracted and frozen bovine mandibular primary incisors were used. The etchant used in this study was 40% phospholic acid gel and the etching times were 0, 10, 20, 30 and 60 seconds. All of the specimens were washed with an air water spray for 30 seconds after etching. The bonding agent and composite resin used in this study were Photo Bond and Photo Clearfil A (Kuraray Co.). Shear bond strength on the ground enamel were measured. After the shear bond strength test, all the test surfaces of the enamel and resin specimens were observed using the SEM. The following conclusions were observed. 1) The etching time which showed the highest bond strength was 30 seconds (80.10 +/- 13.01 MPa). 2) When the enamel was etched with all of the etching times, the bond strengths were significantly higher than that of the enamel without etching. 3) The bond strength with 10 seconds of etching time was significantly higher than that with 20 seconds of etching time, and the bond strength with 30 seconds of etching time was significantly higher than that with 10 seconds of etching time, and the bond strength with 30 and 60 seconds of etching time was significantly higher than that with 20 seconds of etching time. 4) On the enamel specimens after the shear bond strength test, clear prism structures were observed in the group which showed a higher bond strength. 5) On the enamel specimens after the shear bond strength test, the frequency of the resin tags were fractured inside of the enamel prisms was lower in the group which showed a higher bond strengths. 6) Thirty seconds is a sufficient time for etching on the primary ground enamel using Photo Bond and Photo Clearfil A.

[Resin adhesion on the primary ground enamel. (2) Influence of the etched enamel].

The purpose of this study was to investigate the influence of the etching times on the adhesion of the resin on the primary ground enamel. Fifty extracted and frozen bovine mandibular primary incisors were used. The etchant used in this study was 37% phospholic acid gel and the etching times were 0, 10, 20, 30 and 60 seconds. All of the specimens were washed with an air water spray after etching. The bonding agent and composite resin used in this study were Dual Cured Scotch Bond and Silux (3M Co.). Shear bond strengths on the ground enamel were measured. After the shear bond strength test, all the test surfaces of the enamel and resin specimens were observed using the SEM. The results of this study were compared with the previous results using Photo Bond and Photo Clearifil A (Kurarey Co.). 1) The etching time which showed the highest bond strength was 20 seconds (76.77 +/- 12.99 Mpa). 2) When the enamel was etched with all of the etching times, the bond strength were significantly higher than that of the enamel without etching (p less than 0.01). 3) In the etched groups, there was no significant difference among the bond strengths with all of the etching times categories. 4) In the etched groups, the longer the etching times, the lower the standard deviations were. 5) On the enamel specimens after the shear bond strength test, the frequency of the resin tags fractured inside of the enamel prisms was higher in the group which showed a lower bond strength. 6) For the same etching times, significant difference between the bond strengths of Silux and those of Photo Clearfil A was obtained only with the 20 seconds of etching time (p less than 0.01) and the bond strength of Silux was higher.

[Experiences in general anesthesia of patients with scoliosis presupposed to have difficulties in airway maintenance].

We experienced general anesthesia of two patients with cerebral palsy presupposed to have difficulties in tracheal intubation by reason of scoliosis. Case 1 is a 26-year-old woman. Chest X-ray photography and respiratory sounds in both pulmonary areas indicated neither airway stricture nor respiratory distress, but severe scoliosis presupposed difficult tracheal intubation. However, under the impression of possible intubation obtained by preoperative laryngoscopy, orotracheal intubation was tried with rapid induction. Epiglottis deviation to the left noted upon developing larynx made the glottis direct invisible, but some compression of cricoid from outside barely succeeded in intubation. The fixation of the endotracheal tube found much difficulties in its proper positioning so as to make stethoscopy uniform in both pulmonary areas, but trial rotations of the tube both in various directions and at various depths barely managed to find a position of uniform stethoscopy in both pulmonary areas, in which position the tube was fixed as proper positioning. Anesthesia was performed by nitrous oxide.oxygen.halothane; peroperative hemodynamics remained stable and arterial blood gas analysis presented no problems. Case II is a 16-year-old man. Resting respiration presented stridor and chest X-ray photography indicated scoliosis and laryngeal stricture. Patient's lack in the degree of cooperation made laryngoscopy impossible. Thus, in view of a high possibility of difficult tracheal intubation, orotracheal intubation was tried under the control of spontaneous respiration. While the intubation was being carried out by means of a stylet without developing larynx, severe bronchostriction was palpable at the point when the tube barely passed through the glottis, making the intubation impossible. However, the tube barely managed to be inserted while rotating with the stylet being extracted. Anesthesia was carried out by nitrous oxide.oxygen.halothane; peroperative hemodynamics underwent no remarkable change and arterial blood gas analysis presented no problems.

The value of push-type jejunal endoscopy in familial adenomatosis coli/Gardner's syndrome.

Push-type jejunal endoscopy using a long forward-viewing duodenofiberscope was performed in 10 patients with familial adenomatosis coli/Gardner's syndrome. Jejunal polypoid lesions were detected in nine of the 10 patients (90%). The lesions appeared sessile, whitish, and measured 3 mm or less in diameter. Histologic findings of the biopsy specimens from these polypoid lesions revealed tubular adenoma in all nine patients. In addition, this method led to the detection of duodenal adenomas in one patient, in whom duodenofiberscopy with the side-viewing instrument revealed no polypoid lesions in the bulb and second portion of the duodenum. In view of these results, we recommend that push-type jejunal endoscopy, as well as side-viewing duodenofiberscopy, should be added to the usual preoperative examinations done in patients with this disease.

New ligands for human C-reactive protein: calcium-dependent binding with epsilon-aminocaproic acid-agarose and calcium-independent binding with omega-aminohexyl-agarose.

Human C-reactive protein (CRP) is an acute phase protein which increases in concentration in response to inflammation. CRP has been known to bind with phosphorylcholine in a calcium-dependent manner. In this study, CRP is found to bind to affinity chromatography of negatively-charged epsilon-aminocaproic acid-agarose when calcium ions are present, and to the affinity chromatography of positively-charged omega-aminohexyl-agarose if a normal ionic strength of NaCl exists but calcium ion is not present.

[A case of successful closure of multiple lung fistulae by omental pedicle flap after intrapleural sponge plombage 34 years before].

A case of successful closure of pulmonary fistulae by omental pedicle flap is reported. A 58-year-old female with chronic empyema as a sequela of pulmonary tuberculosis was referred with complaints of fever and purulent sputa. After extirpation of spongy "Plombs" (Vinylsponge) which had filled pleural space in previous operation done 34 years before, omental pedicle flap was fixed by sutures to multiple lung fistulae. Omental pedicle was not filled up all the empyema cavity. We anticipate that an omental pedicle flap covering the lung fistulae will be a simpler but promising method of closing lung fistulae, while simultaneously preventive of infection of residual cavity.

Microsurgical transvaso-vasostomy.

We report a case of an infertile man treated successfully with microsurgical transvaso-vasostomy. The patient had a normal testicle with an obstructed vas deferens on one side and an atrophied testicle with a normal vas on the other side. We anastomosed the proximal vas from the functioning testicle to the opposite intact vas through the median scrotal raphe. As a result, the patient fathered a child 16 months after operation.

[Ulcerative colitis with localized clustered inflammatory polyposis; report of a case].

We reported a case of 21 years old male with ulcerative colitis in whom barium-enema follow-up study demonstrated two localized narrowing areas with clustered inflammatory polyps in the transverse colon although there was no severe attack during the 3-year observation period. In this case, the possibility of malignant changes was considered radiographically, but denied endoscopically and histologically by examining the nature of the polyps.

An efficient directional cloning system to construct cDNA libraries containing full-length inserts at high frequency.

We have developed a high efficiency cDNA cloning system which can direct the orientation of inserts in lambda-plasmid composite vectors with large cloning capacities. Cleavage of the vector DNA by SfiI creates two different nonsymmetrical 3' extensions at the ends of the vector arms. Using a linker-primer and an adaptor, cDNA is prepared so it has two different sticky ends which can be ligated to those of the vector arms. When the cDNA fragments and the vector arms are mixed, both the molecules can assemble without self-circularization due to base-pairing specificity. Ligation of the cDNA-vector mixture produces the concatemers from which phage clones carrying a single cDNA insert in the desired orientation can be formed very efficiently by in vitro packaging. This system provides: (1) high cloning efficiency [10(7)-10(8) clones/micrograms poly(A)+ RNA], (2) low background (more than 90% of the clones contain inserts), (3) directional insertion of cDNA fragments into the vectors, (4) presence of a single insert in each clone, (5) accommodation of long inserts (up to 10 kb), (6) a mechanism for rescue of the plasmid part from the lambda genome, and (7) a straightforward protocol for library preparation. Screenings of cDNA libraries constructed by this method demonstrated that cDNAs of up to 6.4 kb, containing complete coding sequences, could be isolated at high efficiency. Thus, this cloning system should be useful for the isolation of cDNAs of relatively long transcripts, present even at low abundance, in cells.

The occurrence of mouse-type oligosaccharides in mouse-human chimeric immunoglobulin G.

The asparagine-linked sugar chains of mouse-human chimeric IgG which is composed of the variable regions derived from mouse and the constant regions derived from human and produced in mouse cells were quantitatively released as tritium-labelled oligosaccharides by hydrazinolysis followed by N-acetylation and NaB3H4-reduction. Paper electrophoresis in combination with sialidase digestion indicated that more than 70% of the oligosaccharides were neutral and the rest was sialylated oligosaccharides. Their structures were determined by sequential exoglycosidase digestions. The mouse-human chimeric IgG was shown to have same sugar chains as those of mouse IgG.

Early colorectal carcinoma with special reference to its development de novo.

The growth type of early colorectal carcinoma was classified into two types. The first type is intramucosal polypoid growth (PG-Ca) and the second type nonpolypoid growth (NPG-Ca) which shows mainly massive infiltration of tumor cells below the submucosal layer. The incidence of adenoma-carcinoma sequence was 72 of 75 lesions (96.0%) in pedunculated polypoid carcinoma, and 61 of 71 lesions (85.9%) in sessile and broad-based polypoid carcinomas. Their average sizes were 15.0 and 18.7 mm, respectively. Submucosal invasive carcinoma (SM-Ca) showed a low incidence. They were detected as microscopical or scattered lesions with a few lymphatic and venous permeation. The NPG-Ca contained 32 lesions. Intramucosal carcinoma without adenoma showing slight depression consisted of ten lesions of which the average size was 5.1 mm. The other 22 lesions showed massive submucosal invasion with marked lymphatic and venous permeation. The average size was 10.3 mm being smaller than PG-Ca. Histologically, NPG-Ca was not accompanied with adenoma. The NPG-Ca arose from de novo carcinoma less than 10 mm in diameter and invaded into the submucosal layer. In advanced carcinoma, the PG-Ca showed a low incidence (21.8%), and almost all cases were of the NPG type (78.2%). The NPG advanced carcinomas increased in those over the size of 20 mm. It is concluded that nonpolypoid early colorectal carcinomas easily progress to advanced carcinoma, and de novo carcinoma occupied about 80% of colorectal carcinoma.
