Orosomucoid 1 and orosomucoid 2 types in the Taiwanese and Japanese: evidence for five new orosomucoid variants.

The genetic polymorphisms of orosomucoid, ORM1 and ORM2, were analyzed by isoelectric focusing on polyacrylamide gels and subsequent immunoprinting. Sera from 600 unrelated individuals in Taiwan and Japan were examined. Five new alleles, designated ORM1*7, ORM1*8, ORM1*4.2, ORM2*8 and ORM2*10, were observed together with common and rare alleles that have been found in the Japanese and the Filipinos. As compared with the Japanese, the Taiwanese have a higher frequency of ORM1*1, and a significantly lower frequency of ORM1*2.1.

The preventive and therapeutic effects of vagotomy for stress ulcer--experimental studies on the aspects of gastric submucosal blood flow and ulcer index.

The purpose of this study was to examine the preventive and therapeutic effects of vagotomy on the stress-induced ulcer in terms of gastric submucosal blood flow and ulcer index. Stress was induced in male Wistar rats by forced immersion in water, and the gastric submucosal blood flow (hydrogen clearance method) and ulcer index were determined in animals that underwent truncal vagotomy alone or truncal vagotomy + pyloroplasty as well as in untreated controls. Both truncal vagotomy alone and truncal vagotomy + pyloroplasty were effective in maintaining the blood flow during stress and preventing the development of ulcers, however, these effects were more notable in the truncal vagotomy + pyloroplasty animals. Truncal vagotomy alone or truncal vagotomy + pyloroplasty performed after the development of stress-induced ulcers had no therapeutic effects.

An ontogenic study of focal seizure induced by penicillin injection into sensorimotor area in rats.

The ontogenic development of focal seizure activity induced by penicillin injection into the unilateral sensorimotor area of rats was investigated in neonate, young, and weaning-through-adulthood groups. In the neonate group, the latency to the onset of repetitive isolated discharges (RIDs) was the longest. RIDs appeared in other structures simultaneously with the focus sensorimotor area, but their amplitude remained low compared to that in the focus. In the young group, RIDs and sustained high frequency discharges (SHDs) occurred. The latency to onset of RIDs was the shortest. RIDs propagated into subcortical structures more quickly than in the weaning-through-adulthood group, but their amplitude remained low in other structures. SHDs of high amplitude occurred in the ipsilateral subcortical structures simultaneously with those in the focus. In the weaning-through-adulthood group, the onset of SHDs was shorter and the frequency of SHDs was greater than those in the young group. RIDs and SHDs appeared in the focus sensorimotor area first and propagated to the ipsilateral caudate putamen, the medial nucleus of the thalamus, and the contralateral sensorimotor area at the early stage of seizures. Thereafter they propagated to other cortical and subcortical structures. These results suggest the following. A well-developed neocortex is needed for generation of SHDs as opposed to RIDs. In the young group, the ipsilateral subcortical structures play an important role in the generation of focal motor seizure. Following maturation, the bilateral sensorimotor areas, ipsilateral caudate putamen, and ipsilateral medial nucleus of the thalamus seem to increase in importance in producing the focal motor seizures.

An ontogenic study of amygdala seizures induced by penicillin in rats.

Amygdaloid seizures were induced by unilateral injection of penicillin into the amygdala in infant and young rats to investigate the ontogenic development of the amygdaloid seizure. "Repetitive isolated discharges" and "sustained high-frequency discharges" appeared in 3- to 4-day-old rats. In these rats, the repetitive discharges appeared in bilateral subcortical structures simultaneously with those of the amygdaloid penicillin focus, but sustained high-frequency discharges were localized to the ipsilateral subcortical structures. In 9- to 10-day-old and 14- to 15-day-old rats, repetitive discharges occurred simultaneously in the bilateral cortices and subcortical structures. In contrast to them, sustained high-frequency discharges propagated faster from the amygdala penicillin focus to the bilateral midbrain reticular formation and medial nucleus of the thalamus than to other regions. In 20- to 21-day-old and older rats, both events occurred simultaneously at all cerebral sites. These results indicate that the subcortical structures, especially the midbrain reticular formation and medial nucleus of the thalamus, play an important role in seizure propagation from the ipsilateral to the contralateral hemisphere in infant rats.

The effect of pictorial context on sentence memory in Broca's and Wernicke's aphasia.

Eighteen aphasic patients (8 Broca's and 10 Wernicke's aphasics), 11 right hemisphere damaged patients and 12 normal subjects were tested to assess the effect of pictorial context on verbal memory with a sentence recognition task. The subjects were read aloud a stimulus sentence describing a simple event and simultaneously shown a picture congruent or incongruent with the sentence. Immediately following or after an interval of 30 sec, the subjects were read aloud a second sentence and asked to judge whether this sentence was the same or different from the stimulus sentence. The results indicated that verbal memory was better retained in supportive situations than in distracting situations, and that this contextual effect was greater in aphasic patients than in any of the other groups. Verbal memory declined rapidly after an interval in aphasic patients, but not in normal subjects. The Broca's and the Wernicke's aphasics demonstrated different performance patterns when semantic analysis of sentences was critical.

Application of the sum peak method to determine the perturbed angular correlation parameters in biological specimens.

The counting rate of a sum peak formed by two gamma-ray emissions in radioactive decay of a nuclide depends on its integral attenuation factor G22. Therefore, in the "sum peak method", the G22 value for the specimen in question can be obtained merely from sum peak measurements using one radiation detector when the resolution time and the geometry are fixed. The procedure to determine the G22 values is described for specimens containing 111In, together with the detailed relationships between the sum peak counting rates and the values of G22.

A comparative study of absence status epilepticus between children and adults.

Based on the clinicoelectrographic data of 28 patients (14 children and 14 adults) with absence status epilepticus thoroughly documented by CCTV/EEG, it was found that there were significant differences between the children and adults. In childhood, absence status tended to occur in those who had experienced individual short-lived atypical absence seizures and also other types of generalized seizure. In contrast, there was a general tendency for absence status in adulthood to occur in females without individual absence seizure. With respect to the clinicoelectrographical manifestations, absence status with a decreased postural tone was prone to be associated with a more profound clouding of consciousness, whereas in those with myoclonic components there was a less profound clouding. The former was found solely in children while the latter was both in children and in adults. During absence status, the focal motor features with or without secondarily generalized convulsions were observed in 8 adult patients (57%). On the other hand, no focal motor manifestations were observed during absence status in children. Absence status is composed of two modalities: either a prolongation or repetition of absence seizures. It was demonstrated that, in children, either the prolongation or repetition of individual absence seizures developed into absence status. The short-lived absence was of an atypical nature, whereas in adults, absence status consisted of the prolongation of an absence seizure which occurred in patients with no experience of individual absence seizures. With respect to the drug treatment, antiabsence drugs had some effect in children, while none of the adult patients responded either to the antiabsence drugs or other antiepileptic drugs. The long-term seizure prognosis was not necessarily poor in children but invariably poor in adults. Four adult patients showed diffuse, but unilateral frontally accentuated asymmetrical paroxysmal activity during the status. Three of them showed initial localized spike-wave discharges in the unilateral frontal region followed by a generalized spike-wave rhythm. Furthermore, all of these 4 patients with focally accentuated ictal EEGs have shown partial motor seizures intermingled with absence status.(ABSTRACT TRUNCATED AT 400 WORDS)

Mitral valve and its ring in hypertrophic cardiomyopathy--a mechanism creating surplus mitral leaflet involved in systolic anterior motion.

A large surplus portion of the mitral leaflets appears to be involved in the systolic anterior motion (SAM) of the mitral valve in hypertrophic cardiomyopathy (HCM). To understand the exact features and the mechanism of the surplus leaflet, we studies the mitral valve and its ring quantitatively with two-dimensional echocardiography in 17 patients with HCM (5 with large SAM, 6 with small SAM and 6 without SAM) and 10 normal subjects. In the large SAM group, the surplus length of the anterior mitral leaflet (AML) increased but that of the posterior mitral leaflet (PML) did not increase when compared to the other 3 groups. In the large SAM group, the length of AML was greater than in the normal group and the length of PML was greater than in the other 3 groups. The mid-systolic anteroposterior diameter of the mitral ring did not differ among the 4 groups. The mid-systolic position of the coaptation was more anterior in the large SAM group than in the other 3 groups. In conclusion, the surplus size of AML is actually increased in the patients with HCM and large SAM, which is caused by the elongation of both AML and PML with anteriorly displaced coaptation and with no dilation of the mitral ring.

Highly efficient procedure for production of human monoclonal antibodies: establishment of hybrids between Epstein-Barr virus-transformed B lymphocytes and heteromyeloma cells by use of GIT culture medium.

We describe a method for production of human monoclonal antibody by a combination of the capacity of Epstein-Barr virus (EBV) to transform human B lymphocytes with somatic cell hybridization, in which a new culture medium, GIT, is used. The transformed B cells from wells positive for anti-purified protein derivative (PPD) fused with a (mouse x human) heteromyeloma line (deficient in hypoxanthine-guanine phosphoribosyl transferase and ouabain-resistant) that had been cultured in GIT medium (Kudo et al. 1987) supplemented with geneticin (antibiotic G418) before cell fusion. The hybrids were selected in GIT medium containing HAT and ouabain (GIT-HAT-O) and cloned by limiting dilution technique by use of GIT medium. According to our method, we obtained higher fusion frequency (1/5.5 x 10(3) vs. 1/1.1 x 10(4)) and higher cloning efficiency (43.3-56.7% vs. 3.3-13.3%) compared with the regular method which used the culture medium containing fetal bovine serum (FBS). Six hybrid clones were consequently obtained and characterized. They produced large amount of specific antibodies (35-170 micrograms/ml) in GIT medium, while establishment of hybrid clones producing specific antibodies by the regular method was unsuccessful. This method will be applicable to any kind of human monoclonal antibody production.

[The phase IV studies with Estracyt in prostatic cancer].

Both anticancer and adverse reactions of Estracyt were investigated by oral administration to 200 patients with prostatic cancer. Two capsules of Estracyt were given twice a day and the administration was principally continued for more than six months. The 200 patients consisted of 68 and 132 patients who were previously untreated and treated, respectively. Thirty seven cases had been treated only with Estracyt and 132 cases also received other treatments. Seventy-five cases were of primary therapy, 71 cases were of maintenance therapy, and 27 cases were of the re-activated stage therapy and 27 cases were of other categories. In conclusion, among the 190 cases for which the due judgement of the effect was possible, Estracyt was markedly effective in 40 cases (21.1%), effective in 43 cases (22.6%), slightly effective in 38 cases (20.0%) and ineffective in 69 cases (36.3%). Adverse reactions were observed in 67 cases (33.5%), among which the administration was discontinued in 18 cases.
