In order to clarify the mechanism of alpha-naphthylisothiocyanate (ANIT) induced cholestasis in rats, a few hepatic changes seen after administration of ANIT 80 mg/kg body weight were examined in relation to time. To clarify changes in the bile production system and bile flow, the concentrations of bile acid and bicarbonate, as well as of sodium in the bile, were measured after infusion of secretin and taurocholic acid. The canalicular bile flow was estimated by measuring the biliary clearance of erythritol-C14. To assess hepatocellular injury, the levels of cytochrome P-450 in the microsome of hepatocytes and protein synthesis in the liver were estimated. It seems that ANIT is activated in the drug metabolising system through cytochrome P-450 in the liver, and significant changes are produced in both the bile acid metabolism and its transport system. Bile acid-independent bile flow in the canaliculus was inhibited, too. Hence, both the bile ductules and the bile ducts are functionally and histologically disturbed.

The author described an epidermoid which revealed homogenous high density in CT scan without contrast enhancement. Sixteen reported cases of intracranial epidermoid with CT scan including our 4 cases were classified in three types; Type I; classical pearly tumor of low density with no contrast enhancement. Type II; low density mass with marginal high density ring by contrast enhancement. Type III; homogenous high density mass with no contrast enhancement. The reason of high density in CT scan was considered mainly due to the peculiar cystic content, which was brown in color and gelatinous. Chemical analysis of the cystic content disclosed various heavy metals and high protein content, however, it was indicated that the high protein concentration was solely attributable to the high density EMI number.
Segments of surgically resected human adult bronchi were xenotransplanted into the subcutaneous tissue of 35 BALB/c genetic background athymic nude mice with 3-methylcholanthrene placed in their lumina. Bronchial segments were removed 8 to 20 weeks after transplantation and preservation of the bronchial mucosa was confirmed in 26 segments. The most frequent change observed in the mucosa was goblet cell hyperplasia, followed by squamous metaplasia and dysplasia. Severe dysplasia or questionable squamous cell carcinoma in situ was noted at the site of the bronchial gland in a segment removed 8 weeks after transplantation, and invasive squamous cell carcinoma in a segment removed 15 weeks after transplantation. A difficulty in this work was the frequent earlier development of malignant tumors of mouse origin.
The activities of arylsulfatases A and B were determined in human primary and secondary tumor tissues (total, 53 cases) of various histological types. Significantly higher activities of these sulfatases were found in almost all the primary lung carcinomas as compared to their corresponding uninvolved tissues. No significant correlation was demonstrated between the enzyme activities and histological figures (stroma amounts, etc.). Lung adenocarcinoma and squamous cell carcinoma showed the presence of an additional arylsulfatase component (B1) which was not detected in normal human lung. The tumor arylsulfatase B1 had an isoelectric point (pI) of 6.7 and was clearly distinguished from arylsulfatase A (pI 4.9) and arylsulfatase B (pI 9.1 to 9.2) in normal lung and lung tumor. The tumor B1 enzyme was demonstrated to be most probably an isoenzyme of arylsulfatase B, since this unusual enzyme was indistinguishable from arylsulfatase B in terms of Ag+ inhibition; its kinetic parameters of Km for p-nitrocatechol sulfate, which was 2.9 mM with B1; optimum pH of 6.3 for B1; heat stability; and substrate specificity for three synthetic and two physiological substrates.

Serum carcinoembryonic antigen (CEA) and calcitonin were assayed in 8 patients with medullary carcinoma of the thyroid (MCT) and 14 unaffected family members, from 4 pedigrees of Sipple's syndrome and one pedigree with inherited MCT. Serum CEA ranged from 5.4 to 44.5 ng/ml in the patients, but less than 2.5 ng/ml in the unaffected. This is in contrast with retinoblastoma and cancer family syndrome, where not only the patients but also unaffected family members show high serum CEA, and cytoplasmic or selective maternal inheritance of serum CEA level is presumed. In inheritable MCT, serum CEA increases in association with the development of MCT, and serum CEA level per se is not inherited.
We have reported 5 cases of fulminant hepatic failure who were treated with a combination of glucagon and insulin. Marked improvement of hepatic coma was characteristically noted in all cases.
The F-P border and strain phenomenon of the gastric mucosa were investigated by the application of methylene blue dye spraying method in endoscopy to 105 asymptomatic control volunteers and the following results were obtained; 1. The pyloric metaplasia is observed in the fundic gland area from the twenties in age and becomes increasing in its number and move widely spreading from the lesser curvature to the anterior and/or posterior wall of the corpus with advancing age. 2. The intestinal metaplasia arises from the thirties in age. 3. The intestinal metaplasia is observed either in the pyloric gland area of following the pyloric metaplasis in the fundic gland area. 4. Histologically, the strain phenomenon of the gastric mucosa is closely related to the intestinal metaplasia. Then, methylene blue dye spraying method is reevaluated to be useful for a precise endoscopic diagnosis of intestinal metaplasia.

An ip transplantation of 3-methylcholanthrene-induced, transplanted fibrosarcoma KMT-17 cells (1 X 10(8)) grew rapidly and killed syngeneic WKA rats in 3-4 days. Agglutinability induced by concanavalin A (Con A) and antigenic expression of KMT-17 cells were investigated in relation to days after ip transplantation. Agglutinability was highest in 1-day-old cells and lowest in 3-day-old cells. The agglutinability of 3-day-old cells increased again when these cells were transplanted into normal rats. The cytotoxic sensitivity of tumor cells to antiserum against tumor-associated surface antigen (TASA) changed simultaneously with the degree of Con A agglutinability. This phenomenon disappeared after artificial infection of tumor cells with Friend murine leukemia virus. The result of the quantitative absorption test at 4 degrees C overnight was that 1- and 3-day-old cells did not differ in their absorbing capacities to anti-TASA sera. However, when the absorption test was done at 37 degrees C for 60 minutes, 1-day-old cells had approximately 16 times more absorbing capacity than 3-day-old cells. However, the cytotoxic sensitivity to antiserum against histocompatibility antigen did not change, regardless of the number of days after ip transplantation. Analysis based on the quantitative absorption test revealed no difference in antibody-absorbing capacities between 1- and 3-day-old cells at both 4 degrees C and 37 degrees C. The relationship between Con A agglutinability and cytotoxic sensitivity to anti-TASA serum is discussed from the viewpoint of "lateral receptor mobility" on the cell surface.

Androphilic proteins in the cytosol from the human benign prostatic hypertrophy are separated into two fractions by Sephadex chromatography; void volume fraction and IgG fraction which was eluted near the site of hIgG. In the present study, properties of these two androphilic proteins were compared. Association constants of these proteins were in the order of 10(9) M-1. However, the binding capacity of the former was smaller than that of the latter. These two androphilic proteins well bound to nuclei, and the high-affinity and saturable binding to nuclei was observed in the 3H-dihydrotestosterone-IgG fraction complex, while binding of 3H-dihydrotestosterone-void volume fraction complex to nuclei was low affinity and unsaturable. The binding of the complexes to chromatin seems to be of low affinity and nonsaturable. These androphilic proteins did not bind to calf thymus DNA. Salt extractability of the bound void volume fraction after incubation with nuclei was not different from that of the bound IgG fraction. It was observed that the chromatographic behavior of the androphilic protein in IgG fraction was changed after incubation with nuclei.
