Cells of origin of spinopontine fibers in the rat.

When wheat germ agglutinin-horseradish peroxidase conjugate was injected into the caudal part of the medial, lateral, ventral and peduncular pontine nuclei in the rat, retrogradely labeled spinal neurons were found contralaterally in the intermediate basilar nucleus (IBN) in the upper two cervical cord segments, and in laminae VI and VII in the lumbar cord segments. The IBN appeared to send fibers mainly to the caudomedial part of the pontine nuclei, while laminae VI and VII of the lumbar cord segments of the caudolateral part of the pontine nuclei.

Methylation pattern of human T-cell leukemia virus in vivo and in vitro: pX and LTR regions are hypomethylated in vivo.

The methylation patterns of the gag, pol, env, pX and LTR regions of proviral DNA of human T-cell leukemia/lymphoma virus type I (HTLV) in fresh leukemic cells and established cell lines were examined using HpaII/MspI endonuclease. Peripheral blood lymphocytes (PBL) isolated from patients with adult T-cell leukemia/lymphoma (ATL) did not express viral antigens of HTLV, but PBL that had been cultured for 2 days did express these viral antigens. Most parts of the gag, pol and env regions of the HTLV provirus in PBL isolated from 12 ATL patients and PBL cultured for 2 days were hypermethylated as reported by others. In contrast, in 10 established cell lines that harbored HTLV genomes and expressed viral antigens, HTLV proviruses were hypomethylated. In one cell line, ATL-IK, which harbored an HTLV genome but did not produce viral antigens, the gag, pol and env regions were hypermethylated. However, two HpaII sites, one in the middle of the gag region and the other in the middle of the pol region, were not methylated even in PBL from most ATL patients. Furthermore, the pX and LTR regions were hypomethylated not only in established cell lines but also in PBL of ATL patients. The hypomethylation of the pX and LTR regions detected in fresh leukemic cells of ATL patients may have some etiological significance in cell transformation by controlling the level of transcription of these regions, or modulating the binding of some factors to these regions.

Ultrastructural study of glycogen-rich oxyphilic adenoma of the nasopharyngeal minor salivary gland.

A glycogen-rich adenoma occurring in the minor salivary gland of the nasopharynx in a 41-year-old woman was studied ultrastructurally. The cytoplasm of the tumour cells was abundantly filled with glycogen particles. The tumour cells possessed many mitochondria, a great number of microvillous processes and microvilli and were joined to each other by desmosomes. These findings suggest that this adenoma is of salivary duct epithelial origin most probably from storing striated ductal cells, and is a variant of monomorphic oxyphilic adenoma.

Clinical features of OKT4+/OKT8+ adult T-cell leukemia.

Adult T-cell leukemia (ATL) has a range of clinical characteristics. Phenotypically the leukemic cells usually express the helper/inducer associated antigen OKT4 with lack of OKT8. We have observed three patients with acute ATL cytologically indistinguishable from OKT4+/OKT8- ATL but whose neoplastic cells had the unusual phenotype, OKT3+, OKT4+, OKT6-, OKT8+ OKT9+/-, OKT11+, Tac+/-, TdT-. All patients had abnormal karyotypes and antibodies against anti-ATL associated antigens as well as proviral DNA of human T-cell leukemia virus in the leukemic cells. The clinical course was complicated by skin eruptions, hypercalcemia, pulmonary infection and disseminated intravascular coagulopathy. All died of complications shortly after diagnosis. The clinical features of these patients were similar to those of OKT4+/OKT8- ATL. However, their acute course suggests that co-expression surface antigens OKT4 and OKT8 may be a sign of aggressive nature of the disease with poor prognosis.

Effects of nifedipine on forearm vascular resistance and venous capacitance in normal subjects and in patients with congestive heart failure.

The effects of oral nifedipine on limb hemodynamics were studied in 7 normal subjects, 8 patients with congestive heart failure and 2 patients who underwent sympathectomy of unilateral limb. Forearm venous capacitance remained unchanged both in normal subjects and in patients with congestive heart failure. In normal subjects, systemic vascular resistance decreased without change in forearm vascular resistance. On the other hand, both systemic and forearm vascular resistance decreased simultaneously in patients with congestive heart failure. In 2 patients with normal left ventricular function undergoing sympathectomy, limb vascular resistance decreased in the denervated side and increased in the contralateral innervated side. These findings indicate that the effects of nifedipine on forearm vascular resistance are dependent upon the circulatory state of the patient at the time the drug is administered, while venous dynamics were not changed by nifedipine, and that the difference in the density of sympathetic innervation results in a reordering of territorial blood flow by modifying the vasodilatation due to the calcium antagonistic action of nifedipine.

Manometric study on the intestinal motility in a case of megacystis-microcolon-intestinal hypoperistalsis syndrome.

We report on the intestinal motility in a case of megacystis-microcolon-intestinal hypoperistalsis syndrome (MMIHS) based on a total registered time of 9.5 h by means of a manometric method we recently developed for neonates. The characteristics of this case are as follows: (a) the frequency of the contractions in the stomach and duodenum was significantly lower than that of healthy infants; and (b) bands of rhythmic contractions, resembling phase III activity of the interdigestive migrating contractions in adults, were recorded in the duodenum, but their occurrence was less frequent and the amplitude was significantly lower than that of healthy infants. The present study indicates that "hypoperistalsis" in MMIHS means not only the low frequency, but also the low amplitude, of the contractions in the alimentary tract.

Normal ventricles in chronic schizophrenics.

The computed tomography (CT) scans of 46 chronic schizophrenic patients and 46 controls were studied using ventricular-brain ratio. Ewans' index, and cella media index. None of the indices used revealed significant differences between the patient and the control groups.

Value of heparinized fine-needle aspiration biopsy in liver malignancy.

The accuracy of diagnosis of hepatic malignancy by percutaneous aspiration biopsy was compared using heparinized and nonheparinized 22-gauge needles. When a heparinized instrument was used, it was able to accurately diagnose malignancy and cytologic types of exfoliative cells. Heparinization also made it possible to recover small tissue fragments and make ultrathin sections for histologic examination. In this way, material suitable for cytologic and histologic examination was obtained from 100% and 95%, respectively, of 59 patients suspected of having hepatic malignancy. A correct diagnosis was made cytologically in 92.5% of patients with hepatic malignancy and in 89.5% of those from whom histologic material was obtained. From a combination of histologic and cytologic results, the overall diagnostic rate for hepatic malignancy was increased to 95%. This procedure proved to be a reliable method for diagnosis of hepatic malignancy.

Epidemiological study of hemorrhagic fever with renal syndrome related virus infection among urban rats in two islands in Tokyo Bay, Japan.

A seroepidemiological survey of hemorrhagic fever with renal syndrome (HFRS) virus infection in 403 urban rats (Rattus norvegicus) captured on two different islands in Tokyo Bay was carried out using the indirect immunofluorescent antibody (IFA) test with the SR-11 strain of HFRS virus. Antibody positive sera were detected in 99 out of 355 (27.9%) rats from the island with reclaimed ground area, but none were detected in 48 rats from the other islands. IFA titers of the positive sera varied from 16 to 4,096 (mostly 128) with a peak at 1 : 128, and antibody positive ratios were significantly higher in the small rats (less than 50 g, probably of younger age) than in the large ones. A causative agent antigenically related to HFRS virus was isolated from lung tissue of seropositive rat using Vero-E6 cells. These results revealed the existence of HFRS related virus among urban rats in the coast of Tokyo Bay, Japan.

[Fundamental and clinical studies on aztreonam in the gynecological field].

Aztreonam (AZT), a new monocyclic beta-lactam antibiotic was studied on clinical efficacy for infectious disease in gynecological field. At about 80 minutes following intravenous injection of 1 g dose of AZT, it penetrated well into internal genital organs at therapeutic levels. Moreover it transferred very fast and enough into intrapelvic dead space exudate, and its level was kept still as high at 12 hours after administration. AZT was given to 20 women affected with gynecological infectious disease. The outcome of AZT therapy was as follows: effective in 5 out of 6 patients (83.3%) administered intravenously and in all of 14 patients (100%) received intramuscularly. Notable adverse effects or abnormal laboratory findings were not observed except 1 case of diarrhea and 2 cases of transient and slight elevation of serum CPK and transaminases. Based on these results, we may conclude that AZT is a highly effective and a very safe antibiotic for the treatment of infectious disease in gynecological field.

[Differential responses of tumor and normal skin tissue to low dose rate radiations].

HeLa S-3 cells in nude mice and the parabasal and basal cells of nude mouse skins were exposed to graded dose rates (37 or 55 rad/hour and 200 rad/minute) and doses of radiation and analyzed in terms of their cell cycle distribution using flow microfluorimetry. Standard procedures were HeLa cells, and the parabasal and basal cells were harvested from the skin using dispase and trypsin. In the case of the tumor cells, in vivo, 37 rad/hour proved to be the most effective in producing G2/M accumulation, which is the sensitive phase of the cell cycle. The same dose rate proved to be the most efficient in producing G2/M accumulation in the parabasal and basal cells of the skin, but sensitivity to this effect of radiation was far less than in the tumor. This differential sensitivity of normal and malignant cells to chronic radiation-induced accumulation in a radiosensitive phase of the cells might play an important role in increasing the therapeutic ratio.

[The cell population kinetics of established human uterine cervical carcinoma in nude mice, with comparative examination by types of tissues].

In order to clarify the cell kinetics as the biological features of human squamous cell cancer in uterine cervix, analysis was conducted on the cells of this cancer in nude mice to which various strains had been transplanted. The results of autoradiography revealed LI = 23 approximately 35.9%, Tc = 15 approximately 23 hours, and GF = 42.4 approximately 67.3%. The values for the small-cell non-keratinizing type were generally higher than in the other types. The results of FCM revealed G1 = 49.4 approximately 66.0%, S = 24.8 approximately 38.3%, and G2 approximately M = 5.8 approximately 17.7%. The results of comparison of tissue types were identical with those of paragraph 1. The results of calculation of the numbers of non-proliferative cell compartments on the basis of the results of autoradiography and FCM revealed Q1 = 26.7 approximately 44.9%, and Q2 = 0.84 approximately 10.5%. From these findings, the Q1/G1 ratio was found to be 50 approximately 70%. The comparison by types of tissues revealed the order small cell non-keratinizing less than large cell non-keratinizing less than keratinizing. From the above findings, it was regarded as very likely that the types of tissues and the cell kinetics in the cases with squamous cell cancer in the uterine cervix are closely related to each other.
