Transformation of rabbit lymphocytes with T-cell leukemia virus.

Co-cultivation of rabbit lymphocytes with a human cell line carrying adult T-cell leukemia (ATL) virus (ATLV) resulted in the establishment of an ATLV-producer rabbit cell line. This cell line expressed ATL-associated antigens and reacted with antisera to the structural proteins of human T-cell leukemia virus which is identical or closely related to ATLV.

Dimorphism of sister chromatid exchange in Bloom's syndrome B- and T-cell lines transformed with Epstein-Barr and adult T-cell leukemia viruses.

The present study describes the establishment of both B- and T-cell lines from the peripheral blood cells of two Bloom's syndrome (BS) patients and one healthy female by using Epstein-Barr (EBV) and adult T-cell leukemia viruses (ATLV). The cell lines from normal and BS subjects exhibited cell surface markers compatible with B- and T-cell origin; in addition, the BS B- and T-cell lines retained the original cytogenetic characteristics of the syndrome. Even though phytohemagglutinin-stimulated BS lymphocytes from the two BS patients studied all showed high levels of sister chromatid exchange (SCE), the established BS B-lines with EBV yielded two separate lines each, i.e., one with increased SCE and another with normal levels of SCE; also, one of the BS T-lines retained high SCE levels in 100% of the cells, whereas the other BS T-line contained two populations, one with high SCE (70%) and the other with normal SCE levels (30%), at a relatively constant frequency over a period of 6 months. Neither EBV nor ATLV caused a significant increase in chromosome instability in the established lines compared to fresh lymphocytes. Reinfection of the BS B- and T-cell lines with EBV or ATLV did not alter the SCE or karyotypes. These results strongly suggest that BS patients have two populations in vivo, one with high and another with normal levels of SCE, at least in the lymphoid cell system.

[Establishment and characterization of ATLV-producing cell lines].

Peripheral blood lymphocytes from a Japanese monkey (Macaca fuscata) were co-cultivated with lethally irradiated MT-2 cells that carry abundant type C virus particles (ATLV) isolated from a patient with adult T-cell leukemia (ATL). After six weeks, a lymphoid cell line designated Si-1 was established from the simian lymphocytes. The Si-1 line was E-, SIg-, Leu-1-, OKI1+, EBNA-, ATLA+ and ATLV+. Co-cultivation of peripheral blood lymphocytes from three anti-ATLA positive and two negative healthy adults resulted in the establishment of three lymphoid cell lines derived either from an anti-ATLA positive donor or both donors. All three cell lines were E+, SIg-, Leu-1+, OKI1+, EBNA-, ATLA+ and ATLV+. This mixed lymphocyte culture technique provides a simple means for the isolation of ATLV from healthy ATLV carriers.

Characteristics of a brain lymphoma cell line derived from primary intracranial lymphoma.

A brain lymphoma cell line, designated TK, was established from biopsy material of a patient with primary intracranial lymphoma. The TK cell line grew in suspension with clumps of cells and consisted of primitive lymphoid cells. TK cells and original lymphoma cells from which the cell line was derived bore surface immunoglobulin and lacked Epstein-Barr virus-determined nuclear antigen. Transplantation of the TK cells into immunosuppressed newborn hamsters gave rise to invasive growth of tumors with metastasis to the brain and eyes. These findings provide definitive evidence for a B-cell origin of the brain lymphoma in this case.

Circadian rhythm of brain susceptibility to haloperidol during chronic administration.

Circadian fluctuation has been reported to exist to the effects of haloperidol after acute administration. In an attempt to clarify the viability of chronotherapy with haloperidol, the antiapomorphine effect of haloperidol after chronic administration was investigated in the present paper. Haloperidol was administered once daily at the same time for 21 consecutive days to rats which were kept under 12 hr lighting conditions with light onset at 19:30. Then the chronology of the antiapomorphine effect was investigated. The antiapomorphine effect was significantly stronger in the group treated at 19:30 than that treated at 13:30. These data agreed with the results found after the acute administration of the drug. After chronic administration, no difference was found in the plasma and brain level of haloperidol due to the time of administration. These experimental results seem to suggest that a circadian rhythm in the brain susceptibility to haloperidol exists even during chronic administration.

Characterization of xenoantiserum produced against B cell acute lymphoblastic leukemia cell line.

Antiserum was produced in white rabbit by intravenously injecting living cells of a B cell acute lymphoblastic leukemia (ALL) line (BALL-1). The reactivity of the antiserum against various lymphoid cell lines was examined by membrane immunofluorescence after appropriate absorption. Serum absorbed with non-T, non-B (NALL-1) and T-ALL (TALL-1) cells recognized B cell antigens distinct from Ia-like antigens on both normal and neoplastic B cells. After further absorption with tonsillar cells or normal B cell line (KO-HL-3), it reacted only with BALL-1 cells and did not react with other leukemia/lymphoma and normal B cell lines. The serum absorbed with tonsillar cells reacted only with BALL-1 and some B cell lines. Thus we were able to obtain antisera with specificity to B cell antigen, B-ALL antigen, and B cell line antigen.
