Antiepileptic effects of globulin-N, an intact human immunoglobulin and its tissue-distribution in kindled cats.

The antiepileptic effects of globulin-N, an intact human immunoglobulin was examined, using the amygdaloid kindled animals prepared by the method of Goddard et al. [1969] and Wada et al. [1974 b]. In the non-treated kindled cats, kindled convulsion and after-discharge (AD) were simultaneously created at 24-hour intervals by electrically stimulating generalized seizure-triggering threshold (GST) in amygdala, hippocampus and neocortex. The kindled cats, treated intravenously with 200 mg/kg of globulin-N, the generalized convulsion and AD produced by GST-stimulation disappeared entirely from 30 min to 8 days in 8 out of 10 cases. Furthermore, kindled cats treated with phenobarbital and phenytoin showed inhibition of GC and AD by GST-stimulation for 24-48 hours. Globulin-N was determined to have a plasma half-life of about 10 days in the kindled cats, and was presented immunohistochemically in the visceral organ, brain and spinal cord. The administered globulin-N passed through the BBB easily in the kindled cats with GST stimulation induced epileptic seizures, and was apparently taken up by the nerve and glial cells in the cerebral cortex and other deep structures of the central nervous system. These results may not only yield clues for the elucidation of the antiepileptic mechanisms of globulin-N, but may also support the clinical use of globulin-N in patients with intractable epilepsy [Ariizumi et al. 1982], including epileptic psychosis.

[Cortical deafness following bilateral hypertensive putaminal hemorrhage].

A case of cortical deafness is reported, which was caused by two episodes of bilateral hypertensive putaminal hemorrhage. Such a case has not appeared in literature up to this time. The patient was a right-handed, 69-year-old man, admitted to us after the stroke with right hemiparesis and disturbance of consciousness. CT scans showed right putaminal hemorrhage. After recovery of consciousness, the patient complained of hearing difficulty of the right ear. The right hemiparesis improved and he was followed ambulatorily after two months hospitalization. He returned to his work and was able to drive a car. His hearing difficulty of the right ear progressed and became stone deaf six months after onset of the initial stroke. Two years after the first stroke, he was attacked by the second stroke with left hemiparesis. CT scans showed left putaminal hemorrhage larger than the previous one in the right. When the patient recovered consciousness, he complained of total deafness. Audiometry test showed that hearing was lost completely to both high and low notes, and there was absence of bone conduction bilaterally. Auditory brain stem response test revealed normal pattern from wave I through wave V bilaterally. Middle latency response showed normal component Po on the right ear and it was absent on the left. The component Pa was not identified on both ears. He showed skill and reliance on lip reading two years after the onset of the complete deafness. Now he can speak fluently and communicate with lip reading and written language, although no improvement of the complete deafness being noticed.(ABSTRACT TRUNCATED AT 250 WORDS)

[Studies on the cellular immune response in patients with urinary bladder carcinoma. XI: Evaluation of specific ADCC activity].

We previously reported that the activity of antibody dependent cell-mediated cytotoxicity (ADCC) of lymphocytes from the patients with urinary bladder carcinoma was lower than that of normal subjects. We performed the specific ADCC assay using SRBC as the target cells, coated with tumor extract from the urinary bladder carcinoma. At the same time, the antiserum was collected from the immunized rabbit by the same tumor extract. The addition of the antiserum from the immunized rabbit gave the highest value for the patient sub-group that produced a precipitin band (Ouchterlony gel diffusion assay). Therefore it is suggested that the reaction of lymphocytes to tumor associated antigens is maintained to a considerable extent, although ADCC activity of the patient group was decreased. However, the antibody producing phenomenon toward the tumor associated antigen might be restrained.

[Frontal SEPs and parietal SEPs following median nerve stimulation--clinical correlation and consideration of their generation sites].

In order to know the characteristics of frontal and parietal SEP components following median nerve stimulation, 25 patients with unilateral cerebral lesions above the thalamus were examined, and their SSEPs were carefully compared with the clinical and radiological findings. In 10 normal subjects, there were three cortical components of the frontal SEPs (P 20-N 28-P 44) and four those components of the parietal SEPs (N 18-P 22-N 26-P 42). In patient's group, central conduction times (CCTs) between components P 13 and each cortical component were measured and the latency differences between normal side and affected side were calculated. When the latency differences increased over 3 S.D. from the mean of the control values or the some cortical components disappeared, they were regarded as abnormal. According to the combination of the abnormalities in frontal and parietal SEPs, three groups were classified as follows: group 1; frontal and parietal SEPs were normal (n = 10), group 2; frontal and parietal SEPs were both affected (n = 10), group 3; parietal SEPs were affected but frontal components were preserved in normal range (n = 5). CT scan showed that the region from internal capsule to cortex around the central sulcus remained intact in the patients of group 1, while this region was involved in various degrees in all cases of the group 2. In patients of group 3, frontal or parietal regions were variously affected. Both the motor and sensory functions were mainly intact in group 1, and disturbed in group 2.(ABSTRACT TRUNCATED AT 250 WORDS)
