Immunohistological studies of masked indoleamine cells in the area postrema of the rat.

Masked indoleamine cells (MICS) in the area postrema and adjacent areas in the rat were immunohistochemically studied (the peroxidase-antiperoxidase method) using a serotonin antiserum. After pretreatment with nialamide (200-300 mg/kg), immunoreactive MICS could be observed. They were small cells (about 12 micron in diameter) with several processes and were distributed in nearly all parts of the area postrema and also in the nucleus tructus solitarii. Following a single intraventricular injection of 75 micrograms 5,6-dihydroxytryptamine, the immunoreactivity of these cells conspicuously decreased for several days. The submicroscopical structure of the cells was investigated using immunoelectron microscopy. Immunoreactive products were observed in the cytoplasm as particles with a diameter of 25-40 nm and high electron density, but these were not found in the nucleus or cell organelles.

Extraosseous osteogenic sarcoma arising from the retroperitoneum: a case report.

A 44-year-old housewife with a mass in the left upper quadrant of the abdomen was admitted. The tentative diagnosis was cystadenocarcinoma arising from the pancreatic tail, as determined by CT and selective angiography. Excision of the mass with removal of the spleen, pancreatic tail, left renal capsule and surrounding lymphnodes was performed. Photomicroscopic sections revealed proliferation of malignant cells and malignant osteoids. The final diagnosis was osteogenic sarcoma arising from the retroperitoneum. Among the extraosseous osteogenic sarcoma arising from the somatic soft tissue, involvement of the retroperitoneum is rare. Only 12 cases including our own case have been reported as being of retroperitoneal origin. No patient survived for five years and in our patient, there are signs of recurrence one year after the surgery.

Immunohistochemical investigations of the influence of reserpine on the serotonin neuron system in the rat brain.

A peroxidase-antiperoxidase immunohistochemical method with serotonin antiserum was employed to investigate the influence of reserpine on serotonin neurons of rats which were sacrificed at various times after injection (10 mg/kg i.p.). The disappearance of serotonin immunoreactivity induced by reserpine was detected only in the perikarya after 15 min, and then rapidly proceeded to the terminals. Between 2 and 4 h, immunoreactivity completely disappeared throughout the brain. The immunoreactivity reappeared in the perikarya after 6 h, and progressed toward the terminals gradually. However, there was an obvious difference in the rate of recovery of immunoreactivity between areas. After 7 days, the immunoreactivity returned to control levels.

Dopamine-induced potentiation of the contractile response to arachidonic acid in guinea-pig aorta.

In guinea-pig aorta, the potentiating effect of dopamine on the response to arachidonic acid was examined. The potentiation by dopamine was completely abolished by nordihydroguaiaretic acid, but not by indomethacin, aspirin, corticosterone, haloperidol, SK & F 83566, propranolol, PGI2 or imidazole. Dopamine did not have any significant effect on the response to PGF2 alpha U-46619, KCl, 5-hydroxytryptamine or histamine.

Observations of energy metabolism in neuroectodermal tumors using in vivo 31P-NMR.

The energy metabolism of living tumors in rats and hamsters were investigated by obtaining in vivo 31P-NMR spectra, and the effects of chemotherapy on tumors were evaluated by observing the changes of these spectra. Tumor cells of rat glioma, human glioblastoma and human neuroblastoma were inoculated subcutaneously in the lumbar region of the animals. After the tumor grew to over 1.5 cm in diameter, in vivo 31P-NMR spectrum data was obtained selectively from the tumor with a TMR-32 spectrometer (Oxford Research Systems, U.K.). Several peaks (ATP, inorganic phosphate (Pi), phosphodiesters and phosphomonoesters (PME) were observed in the tumors. The heights of these peaks varied widely corresponding to the tumor growth. However, the spectrum pattern of each tumor in an active stage was found to be essentially the same regardless of histological type or tumor origin. The phosphocreatine (PCr) peak was small, ATP and PME peaks were large and tissue pH calculated from the chemical shift of Pi was low in each tumor group. After intravenous injection of a large dose of a chemotherapeutic agent, ATP peaks decreased and the Pi peak increased gradually, resulting in a dominant Pi peak pattern after several hours in all groups. With lower drug doses, spectrum changes were temporarily seen in the tumors. These findings indicated that drugs with a high dose have a selective and a direct action on the energy metabolism of tumor tissues. In vivo 31P-NMR spectra measurement is very valuable not only to investigate the energy metabolism in tumor tissue but also to evaluate the effects of chemotherapy on the tumor.

Comparison of the effects of primary prostaglandins on isolated human urinary bladder.

The effects of primary prostaglandins (PGs) on the dome and trigone of the human urinary bladder were investigated in vitro. In the dome preparations, PGE1 (10(-9) to 10(-6) gm./ml.) showed a weak contractile effect, while PGE2 (10(-9) to 10(-6) gm./ml.) produced dose-dependent contractions. PGF2 alpha (10(-9) to 10(-6) gm./ml.) induced contractions greater than those induced by PGE1 and PGE2. In the trigone preparations, PGE1 (10(-9) to 10(-6) gm./ml.) had no effect, and PGE2 (10(-9) to 10(-6) gm./ml.) produced only a weak contractile response. PGF2 alpha (10(-9) to 10(-6) gm./ml.) showed a greater contractile potency than PGE2. Contractile responses of both preparations to PGE1, PGE2 and PGF2 alpha were not affected by pretreatments of atropine, hexamethonium and phentolamine. These results suggest that the order of potency of primary PGs to induce contractile responses is PGF2 alpha greater than PGE2 greater than PGE1 in both the dome and the trigone preparations, and that the contractile effects of PGE1, PGE2 and PGF2 alpha may not be mediated by activation of muscarinic, nicotinic or adrenergic alpha-receptors.

Treatment of juvenile laryngeal papilloma with a combination of laser surgery and interferon.

The therapeutic results obtained from the treatment of juvenile laryngeal papillomas with combined laser surgery and alpha-type IFN treatment are reported. Three cases of papilloma occurring under four years of age were found to be of the fulminant type with a strong tendency for recurrence. The results were complete remission of papilloma in two cases and a residual trace in one case. Thus, at present, control of the disease has been successful. A combination of several modes of treatment may be necessary for a more lasting therapeutic effect. Currently, no single method has been used to treat the disease. Laser surgery and IFN are both powerful therapies for this tenacious disease.

A truncated immunoglobulin epsilon pseudogene is found in gorilla and man but not in chimpanzee.

Molecular genetic analyses of the young pseudogenes of the immunoglobulin C epsilon genes were carried out to obtain qualitative evidence for the phylogenetic branching pattern of hominoid primates. We found that Old World monkeys had two C epsilon genes, one of which was processed. Among the hominoids examined only the gorilla and human genomes contained three C epsilon genes: an active, a truncated, and a processed gene. Other hominoids so far examined, including chimpanzee, contained two C epsilon genes: one active and the other processed. These results suggest that the processed C epsilon pseudogene was generated before the divergence between Old World monkeys and hominoids and that the gorilla is more closely related to man than the chimpanzee is, unless the chimpanzee has lost the C epsilon 2 gene after the divergence of this species.

Differences in the processing of secreted glycoprotein A induced by Marek's disease virus and herpesvirus of turkeys.

The steps from the precursor to the processed forms of a secreted glycoprotein (gA) of Marek's disease virus (MDV) and of herpesvirus of turkeys (HVT) were examined by two-dimensional gel electrophoresis of immunoprecipitates from the cell lysate and medium of infected cultures with monoclonal antibodies. Differences between MDV- and HVT-gA were particularly observed in isoelectric points of the glycosylated or unglycosylated precursor forms.

Correlation between plasma fibrin-fibrinogen degradation product values and CT findings in head injury.

Plasma fibrin/fibrinogen degradation product (FDP) determinations and CT scans were performed in 26 patients on the first, third, and seventh day after head injury. Plasma FDP concentrations were increased in the patients with cerebral contusion shown on CT scans, while the plasma FDP values were normal in the patients with various sized epidural haematomas. Furthermore, the plasma FDP elevation appeared to be more marked and more prolonged in the patients with severe contusion than in those with mild contusion. These findings suggest that the degree of the increase in plasma FDP is proportional to the amount of brain tissue destruction.

Immunohistochemical studies of the serotonergic supraependymal plexus in the mammalian ventricular system, with special reference to the characteristic reticular ramification.

Distributional and morphological features, especially characteristics of the ramification of serotonin-containing supraependymal fibers (SEF), were studied in the ventricular systems of mammals (mouse, rat, guinea pig, rabbit, cat, dog, monkey) by means of a modified peroxidase antiperoxidase technique, using antiserotonin antiserum prepared in our laboratory. SEF were present in all ventricular systems, except on the third ventricle floor and in the choroid plexus. The density of SEF was higher in the smaller species. In the rat, light- and scanning electron microscopical SEF were almost completely abolished 1 week after intraventricular administration of 5,6-dihydroxytryptamine. Ramification of SEF was complicated; the SEF formed a true network with frequent anastomosing. In the ventricular system of rats rendered hydrocephalic by kaolin administration, the mode of axonal branching in the supraependymal plexus could best be analyzed by the scanning electron microscope because the meshes of the plexus were spread out.

Hazard to endotracheal tubes by CO2 laser beam. Experimental report.

Ignition or combustion of the endotracheal tube during CO2 laser microlaryngo surgery may cause fatal lung disorders. We undertook an experiment on ignition and combustion of endotracheal tubes by CO2 laser beam and came to the following conclusions: (a) the silicon endotracheal tube was nonflammable; (b) the spiral endotracheal tube was also nonflammable; (c) a pulse wave was recommended and 3-second intervals were necessary; (d) four sheets of moist gauze were necessary to prevent cuff trouble, and (e) it was advisable to set the O2 concentration below 25% when using the silicon endotracheal tube.

Applications of anticardiac myosin monoclonal antibodies in the diagnosis of heart disease.

We have developed anticardiac myosin antibodies, especially monoclonal antibodies, for helping in the diagnosis of heart disease. Our investigations were divided into three research projects. We visualized the distribution of myosin isozymes in human atrial and ventricular myocardium by an immunofluorescence staining method using monoclonal antibodies specific for individual human cardiac myosin isozymes. We also revealed the redistribution of these cardiac myosin isozymes in an overloaded condition. The isozymic pattern of cardiac myosin was changed from the atrial type to the ventricular type in the overloaded atrium. This isozymic redistribution can be considered as physiological adaptive mechanism to meet increased cardiac work during overload. We developed a new method of imaging for myocardial infarction by single photon emission tomography using labelled monoclonal antibody specific for cardiac myosin heavy chain. Specific localization of the labelled antibody was demonstrated in the infarcted area and no accumulation of radioactivity was shown in the bone matrix as observed in 99mTc pyrophosphate images. We developed a sensitive radioimmunoassay of cardiac myosin light chain I (LCI) and demonstrated that peak serum levels of LCI in the patients with acute myocardial infarction correlated well with the left ventricular ejection fraction. Furthermore, LCI release from the infarcted myocardium was not affected by coronary reperfusion due to intracoronary thrombolysis. Thus, serial determinations of serum LCI better quantify the extent of myocardial damage even after coronary reperfusion in acute myocardial infarction.
