Pretreatment with acebutolol or propranolol at high concentrations had an inhibitory effect on the contractile response to 5-hydroxytryptamine (5-HT) in most vascular smooth muscles such as rabbit aorta and basilar, mesenteric, renal, femoral arteries and cat coronary artery. The inhibitory actions of both agents were generally greater than on the responses to excess Ca2+ and potassium. In rabbit renal arteries, acebutolol had no effect on the response to 5-HT but inhibited the responses to excess Ca2+ and potassium. Propranolol had a marked inhibitory effect on the response to 5-HT. In all preparations used, the contractions induced by norepinephrine (NE) and histamine showed a much greater resistance to the effect of acebutolol and propranolol than the contractions induced by 5-HT, Ca2+ and potassium. Nifedipine had no inhibitory effect on the response to 5-HT in most of the preparations. Nifedipine inhibited the response to 5-HT only in the basilar arteries. The inhibitory actions of propranolol on the response to 5-HT was greater than that of acebutolol. The inhibitory action of acebutolol and propranolol on the response to 5-HT may be related to mechanisms other than the beta-adrenoceptor blocking action of the drugs. The possible mechanisms of inhibitory action of both beta-adrenoceptor antagonists on 5-HT are discussed.

Laboratory and clinical studies were carried out with aspoxicillin (TA-058, ASPC), a new semisynthetic penicillin, in pediatric infectious disease, and following results were obtained. The average serum concentrations of ASPC after intravenous injection were 53.6, 151.2 micrograms/ml at 15 minutes, 28.0, 72.8 micrograms/ml at 30 minutes, 17.6, 43.3 micrograms/ml at 1 hour, 7.0, 19.9 micrograms/ml at 2 hours, 0.3, 1.8 micrograms/ml at 6 hours, when the doses were 20 mg/kg and 40 mg/kg, respectively. The mean half-lives of ASPC in blood after injection were 0.87 hour and 1.1 hours when the doses were 20 mg/kg and 40 mg/kg, respectively. The mean recovery rates in the urine during 6 hours after intravenous injection of 20 mg/kg and 40 mg/kg were 62.0 and 62.8%, respectively. The antibacterial activity of ASPC against clinically isolated organisms was determined. ASPC had good activity against H. influenzae, H. parainfluenzae, S. pneumoniae and S. pyogenes. Thirty-four patients; 26 cases of respiratory tract infections, 6 cases of urinary tract infections, 1 case of bacterial meningitis and 1 case of bacterial lymphadenitis, were treated with 57.0-159 mg/kg daily dose of ASPC for 4-19 days. The rate of satisfactory clinical response was 85.3%. As to side effects, loose stool was observed in 3 cases, slight elevation of GOT & GPT and elevation of platelet were noted, in 2 cases, respectively. All were transient and considered to be minor.

Hanganutziu-Deicher (HD) antigen-active N-glycolylneuraminic acid (NeuGc)-containing gangliosides were isolated and characterized from human colon cancer tissues. The antigenic gangliosides were detected by thin-layer chromatography by our newly developed method (H. Higashi, Y. Fukui, S. Ueda, S. Kato, Y. Hirabayashi, M. Matsumoto, and M. Naiki. J. Biochem., 95: 1517-1520, 1984) of enzyme immunostaining using affinity-purified chicken antibody against hematoside containing NeuGc (II3NeuGc-LacCer) and horseradish peroxidase-conjugated rabbit anti-chicken lgG. One to six species of the antigenic gangliosides were isolated from seven of 16 cases of colon cancer, whereas no antigenic compound was detected in all apparently normal colorectal tissues from 17 individuals without colorectal cancer. Tissues from different patients showed different patterns of molecular species of the antigenic gangliosides. Densitometric determination indicated that HD antigenic sialic acid, NeuGc, accounted for about 1% or less of the total lipid-bound sialic acids. Four species of antigenic gangliosides were identified as hematoside and hematoside-containing O-acyl ester (II3NeuGc-LacCer and II3 4- or 7-O-acyl-NeuGc-LacCer), GM2-containing NeuGc (II3NeuGc-GgOse3Cer), and sialylparagloboside (IV3NeuGc-nLcOse4Cer) by their behaviors on 2-dimensional thin-layer chromatography, and the effects of mild alkaline treatment, sialidase treatment, periodate oxidation, and endo-beta-galactosidase treatment.
Serum pancreatic elastase 1 content in 45.9% of 203 samples from 32 patients with acute pancreatitis was found to be greatly elevated to greater than 1000 ng/dl, much higher than the control values (90 to 270 ng/dl), whereas serum alpha-amylase activity, in 62.3% of the same samples, was within the control range (100 to 460 IU/L). The increased pancreatic elastase 1 content persisted for several days after serum alpha-amylase activity returned to normal, suggesting that, compared with serum alpha-amylase activity, pancreatic elastase 1 content truly reflects the clinical course of acute pancreatitis. The measurement of pancreatic elastase 1 content appears to be more valuable in the diagnosis and prognosis of acute pancreatitis than that of serum alpha-amylase activity. In 41 patients with chronic pancreatitis, on the other hand, pancreatic elastase 1 content and serum alpha-amylase activity were within the normal range in 38.3% and 56.2% respectively, of 336 samples.
This study was based on our model of experimental autoimmune hepatitis produced by immunizing C57BL/6 (B6) mice with syngeneic liver proteins and Freund's complete adjuvant. Spleen cells obtained from these hepatitis mice were transferred to syngeneic normal recipient mice, and histological changes occurring in the liver and the role of cellular immunity in producing liver damages in recipient mice were investigated. Sensitized spleen cells from these immunized (donor) mice were fractionated by a nylon wool column and injected intravenously into normal B6 mice. Histology of the liver of the recipient mice showed mild infiltration of mononuclear cells around the periportal area 7 days after the transfer of sensitized spleen cells, and changes were most prominent in the mice injected with the fraction of nylon wool column adherent spleen cells. Induction of these liver lesions in the recipient mice was blocked by treatment of sensitized donor spleen cells with anti-Thy 1,2 monoclonal antibody and guinea pig complement before injection. Lymphocyte reactivity to liver-specific lipoprotein (LSP) in recipient mice was studied by a lymphocyte transformation system, and a high immune response was demonstrated with the fraction of nylon wool column non-adherent (T-enriched) spleen cells. These data seem to indicate T-cell interactions between donor and recipient mice in the transfer study using our experimental autoimmune hepatitis model.

Fundamental and clinical trials were carried out with cefminox (CMNX, MT-141) in pediatric infections. Results were as follows. The mean serum concentrations of CMNX following intravenous injection of 10, 20 and 40 mg/kg were 73.1, 112.5 and 181.4 micrograms/ml at 15 minutes after injection, respectively. The serum half-life times were 1.37, 1.20 and 1.53 hours, respectively. Average recovery rates in the urine until 6 hours from the start of injection were 91.4, 59.4 and 85.8%, respectively. The antimicrobial activity of CMNX against clinically isolated organisms was measured; CMNX was more active than CMZ and CEZ against H. influenzae. CMNX was equal to or more active than CMZ and CEZ against E. coli. CMNX was administered clinically to 32 pediatric patients with various infections; 19-pneumonia, 5-bronchopneumonia, 3-bronchitis and 5-pyelonephritis. Overall efficacy rate was 93.8%. Slight elevation of S-GOT and S-GPT was observed in 2 patients. No other serious side effect was observed.

The effects of chemotherapy on living tumor tissue in hamsters and rats were investigated by measuring the 31P nuclear magnetic resonance spectra using topical magnetic resonance. Human neuroblastoma, human glioblastoma, and rat glioma tumor cells were inoculated s.c. in the lumbar region of the animals. After the diameter of the tumors increased to 1.5 cm, in vivo 31P nuclear magnetic resonance spectra were measured selectively in the tumors with a TMR-32 spectrometer. Adenosine triphosphate, inorganic phosphate (Pi), phosphodiester, and phosphomonoester peaks were observed. The phosphocreatine peak was hardly detectable, adenosine triphosphate and phosphomonoester peaks were high, and tissue pH, calculated from the chemical shift of Pi, declined. Regardless of the tumor origin or the histological type, the spectral pattern of each neuroectodermal tumor was found to be essentially the same. After i.v. injection of a large dose of a chemotherapeutic agent, adenosine triphosphate peaks decreased and Pi increased gradually, resulting in a dominant Pi peak pattern after 6 to 12 hours. However, during the same period, there were no observable changes in the spectra of normal organs. These findings indicated that the drugs have a selective and direct action on the energy metabolism of tumor cells. With lower drug doses, no remarkable changes were seen in the spectrum. Measurement of in vivo 31P nuclear magnetic resonance spectra is valuable not only to investigate the energy metabolism in tumor tissue but also to evaluate the effects of chemotherapy.

Fundamental and clinical studies on BRL 25000 granules, containing 2 parts amoxicillin (AMPC) and 1 part clavulanic acid (CVA) (a beta-lactamase inhibitor) were carried out in the pediatric field. Serum concentrations and urinary excretion rates were determined after oral administration of BRL 25000 granules at a dose of 20 mg/kg to 2 children. The mean peak serum concentrations of AMPC and CVA were 4.89 and 2.85 micrograms/ml at 1 hour after administration, with serum half-lives (T 1/2) of 1.15 and 0.89 hours respectively. Mean cumulative urinary excretion rates of AMPC and CVA in the 6 hours after administration were 24.91% and 10.19%, respectively. BRL 25000 granules were also administered at daily doses of 25.1-60.4 mg/kg in 3 divided doses, to 20 pediatric patients with bacterial infections (4 acute tonsillitis, 2 acute pharyngitis, 3 suspected scarlet fever, 3 acute bronchitis, 8 urinary tract infection). The efficacy rate was 100% clinically and 70% bacteriologically. No adverse reactions were observed, however, abnormal laboratory findings were observed in 4 cases (slight elevation of GOT in 2, GPT in 1, eosinophilia in 1).
