Delayed type hypersensitivity responses in mice transplanted with rat hepatocytes.

The delayed type hypersensitivity (DTH) response to xenogeneic hepatocytes (HCs) was investigated in mice received subcutaneous (s.c.), intrasplenic (i.s.), or intravenous (i.v.) transplantation of rat HCs. The DTH response in mice preimmunized i.s. or i.v. with rat HCs (1 x 10(6) cells) was significantly lower than that in mice preimmunized s.c. with the same doses of rat HCs. Co-transfer of spleen cells from i.s. immunized mice with spleen cells from s.c. immunized mice to naive recipient mice did not suppress the DTH response induced by transfer of spleen cells from s.c. immunized mice. On the other hand, co-transfer of spleen cells from i.v. immunized mice with spleen cells from s.c. immunized mice suppressed the DTH response to rat HCs in recipients. Furthermore, the levels of DTH responses in recipients transferred with spleen cells from mice sensitized i.s. or i.v. with rat HCs, immunized s.c. with rat HCs 6 hr after transfer, and challenged with rat HCs 7 days later was almost similar to those in recipients transferred with spleen cells from s.c. immunized mice. These results suggest that antigen-specific suppression of DTH responses to rat HCs in mice is associated with the presence of suppressive spleen cells induced by i.s. or i.v. immunization with rat HCs.

[Thermal insulation of clothing for seated and standing postures].

The purpose of this study is to examine the effects of body posture on thermal insulation of clothing. Clothing ensembles including skirts were mainly investigated. A new thermal manikin was developed for this study. This manikin was able to change her body posture without increasing or decreasing her surface area. By changing posture from standing to seated, the total thermal insulation of clothing (It) was decreased by around 1.4%. The surface thermal resistance at nude (Ia) was increased by 8%. As a result, the basic thermal insulation of clothing (Icl) was decreased by 14%. The main reason for the decrease of basic thermal insulation (Icl) when the posture of thermal manikin changes from standing to seated was the increase of Ia at nude. The total thermal insulation of clothing for each body posture (Iti), when standing and seated, were compared. The parts affected by the changed posture were abdomen, hip and thighs, and especially under the skirt of 61cm length, the knees were most affected.

Effect of twinning and supplemental iron-saturated lactoferrin on iron status of newborn calves.

Data from 53 calves by embryo transfer were collected to determine the effect of twinning and supplemental Fe-saturated lactoferrin or FeSO4 for 5 d after parturition on iron status of calves. Blood hematocrit and hemoglobin of multiple calves born from multiparous cows at d 1 postpartum were lower than those of single calves born from multiparous cows, although those of their dams were not different. Plasma Ca, inorganic P, Mg, Zn, and Cu were similar for single and multiple calves or for calves born from primiparous and multiparous cows. Blood hematocrit and hemoglobin of untreated calves decreased from 1 to 6 d postpartum. Blood hematocrit of calves treated with 40 mg of Fe/d as FeSO4 increased from 1 to 10 d postpartum, and blood hemoglobin of calves treated with 20 or 40 mg of Fe/d as ferrous Fe increased from 1 to 10 d postpartum. Blood hematocrit and hemoglobin of calves treated with 20 mg of Fe/d as Fe-saturated lactoferrin remained nearly constant for 10 d postpartum. Serum Fe of calves increased immediately after the treatment with 20 or 40 mg of Fe as FeSO4, but serum Fe of calves treated with lactoferrin was not affected. Thus, ferrous Fe may be more effective than Fe-saturated lactoferrin in elevating blood hematocrit and hemoglobin in newborn calves.

[A case report of interstitial pneumonia caused by granulocyte colony-stimulating factor].

Several clinical trials have demonstrated that granulocyte colony-stimulating factor (G-CSF) accelerates the recovery of neutropenia in chemotherapy-induced bone marrow suppression. In this report, we describe a 46-year-old female with glioblastoma multiforme who developed interstitial pneumonia due to administration of G-CSF during the phase of immunochemoradiotherapy-induced neutropenia. Thirty-three days after starting immunochemoradiotherapy (ACNU, VCR, IFN -beta, radiation), she developed neutropenia (1,000/microliters). Administration of G-CSF at doses of 125-250 micrograms/day led to an increase of peripheral neutrophil counts. Eleven days later, the patient developed sudden severe respiratory failure and cyanosis with worsening of lung shadows. Blood gas levels on room air were PaO2 49.3mmHg, PaCO2 28.0mmHg, and pH 7.46. At this time, her neutrophil count had risen to 26,080/microliters. LDH and alpha - HBD had also increased to 1,439 IU/l and 1,117IU/l respectively. Chest radiograph and CT scan demonstrated interstitial pneumonia. After treatment with methyl prednisolone, her respiratory symptoms were gradually resolved. A number of side-effects have been reported with granulocyte-macrophage colony-stimulating factor (GM-CSF). These include fluid retention with pericardial and pleural effusion, fever, bone pain, fatigue, and rash. This report also suggests that G-CSF might be a cause of interstitial pneumonia during the phase of immunochemoradiotherapy-induced neutropenia.

[Clinical analysis of the fatal cases of adult malignant gliomas after aggressive treatment].

Six patients operated on for supratentorial malignant astrocytomas and seven patients operated on for glioblastoma multiforme were analyzed to evaluate the effect of aggressive surgical resection on the length of survival and causes of death. Early postoperative contrast enhanced CT scan was used to assess the extent of surgical resection. A gross total resection was considered to have been accomplished when there was no evidence of any residual enhanced mass. When 10% or less of the preoperative enhanced mass remained, the resection was classified as a subtotal resection. Subsequent follow-up CT scan showed that a gross total resection was accomplished in nine patients, and a subtotal resection was attained in four patients. The patients' ages ranged from 40 to 78 years (mean, 59 years). The median survival after the first aggressive surgical resection was 18.0 months in patients with malignant astrocytoma and 13.6 months in those with glioblastoma multiforme. The median duration between first operation and recurrence of tumor was 8.8 months in patients with malignant astrocytoma and 11.5 months in those with glioblastoma multiforme. A second aggressive surgical resection for recurrent malignant astrocytoma or glioblastoma multiforme was carried out in four patients (40%) of the evaluated ten patients. The median survival of these patients after reoperation was 8.25 months. Accordingly, aggressive surgical resection of malignant astrocytoma and glioblastoma multiforme is correlated with longer survival and is advocated in the treatment of recurrent tumors. Leptomeningeal dissemination was diagnosed in nine patients (90%) of evaluated ten patients.(ABSTRACT TRUNCATED AT 250 WORDS)

[Cerebellar infarcts that require differentiation from tumors: diagnosis with MRI].

It is often difficult to differentiate cerebellar infarct with cerebellar swelling from neoplastic disorders, because former can be shown as cerebellar mass with marked contrast enhancement on CT scan. We analyzed radiologically three cases with cerebellar infarction by using MRI, conventional CT scan and angiography. Two cases in the acute stage could be diagnosed as cerebellar infarction by MRI alone based on the following findings: 1) the lesion was distributed in the territory of cerebellar arteries; 2) the normal pattern of cerebellar folia and fissures was preserved in Gd enhancement MRI image; 3) characteristics of MRI intensity were compatible with hemorrhagic infarction. The other case in the chronic stage showed peculiar enhancement, which was unusual for infarction. It was diagnosed as cellular infarction with reference to the angiographic findings. MRI is generally useful to obtain early diagnosis of tumor-like cerebellar infarcts, and proper treatment should be started as early as possible.

[Morphological changes of Kock pouch investigated by autopsy two years nine months postoperatively].

A 62-year-old male patient consulted us because of gross hematuria and was diagnosed as having a bladder tumor. Total cystectomy was performed and a Koch pouch was utilized. Two years and nine months later, he died as a result of recurrence of bladder tumor and pathological autopsy was performed. A microscopic section of the reservoir mucosa showed a reduction in the number of crypts and an increase in the number of goblet cells. These morphological changes seemed to be caused by chronic exposure to urine, but have a favorable effect upon metabolic alterations following the utilization of the Kock pouch.

[A case of dural type of histiocytosis X presenting as a mass lesion in the tentorium cerebelli].

Histiocytosis X is a disease of unknown etiology, characterized by a mass of proliferating histiocytes, plasma cells and inflammatory cells foaming a granuloma within the reticuloendothelial elements of any organ in the body. In the central nervous system (CNS), hypothalamic disorder of histiocytosis X is often found, but histiocytosis X in other regions is quite rare. We report a case of a 5-year-old girl with histiocytosis X of the zygoma presenting as a mass lesion in the tentorium cerebelli. A computed tomographic (CT) scan demonstrated a tumor at the left tentorial region, extending along the dura mater of the tentorium cerebelli. Magnetic resonance imaging (MRI) revealed a low signal intensity region on both T1 and T2-weighted images. MRI with Gd-DTPA showed a homogeneous enhanced mass extending to right and inferior sites with a thickened tentorium. As the thickened dura matter continued from the left middle fossa to the mass lesion, the tumor was considered to arise from the left zygoma and extend to the tentorium cerebelli. CNS extension of histiocytosis X is manifested either as (1) the cerebral type or (2) the dural type. Many cases of cerebral type histiocytosis X including hypothalamic disorder have been reported. Only 6 cases of the dural type of histiocytosis X have been described. Although the lesions of the cerebral type of histiocytosis X show prolonged T1 and T2 values on MRI, the MRI findings of the dural type have not been reported. The present case is the first report of the appearance of the lesion on MRI.(ABSTRACT TRUNCATED AT 250 WORDS)

[A case of unroofed coronary sinus with persistent left superior vena cava].

A 44-year-old female with left to right shunt and persistent left superior vena cava (PLSVC) suspected of unroofed coronary sinus defect with patent right SVC and innominate vein was treated surgically. PLSVC was ligated and coronary sinus ostium was closed with a EPTFE patch. Roof defect of coronary sinus was left open and allowed the coronary venous blood to drain into the left atrium. Patient recovered well and post-operative cardiac catheterization revealed no significant right to left shunt nor desaturation of arterial blood. In the correction of unroofed coronary sinus defect and PLSVC and no other intra-cardiac defect, ligation of PLSVC and closure of coronary sinus ostium was simple and effective, although it was not anatomically corrected.

Determination of bound sulfur in serum by gas dialysis/high-performance liquid chromatography.

A sensitive high-performance liquid chromatography (HPLC) method has been developed for the determination of bound sulfur in mammalian serum. In this work, bound sulfur is defined as divalent sulfur that is easily liberated as sulfide by reduction with dithiothreitol. Released sulfide was treated by flow gas dialysis and converted into a fluorescent derivative, thionine, through the reaction with p-phenylenediamine and ferric ion. Thionine was then determined by HPLC using fluorometric detection. The calibration curve for bound sulfur is linear in the range from 0.1 to 10 microM, and the sensitivity of this method allows detection of less than 10 nM bound sulfur (0.5-ml sample). Recoveries of low-molecular-weight and high-molecular-weight bound sulfur added to human serum at 1.0-5.0 microM levels averaged between 94 and 101%. The proposed method was applied to the determination of bound sulfur in human and various animal sera. The mean concentration in normal human serum was 1.16 +/- 0.09 microM for males (n = 5) and 1.07 +/- 0.18 microM for females (n = 5). The mean concentrations ranged from 1.55 to 6.18 microM in animal sera. The form of the sulfur bound in serum was also discussed.

Isomer-specific analysis and toxic evaluation of polychlorinated biphenyls in striped dolphins affected by an epizootic in the western Mediterranean sea.

Isomer-specific concentrations of polychlorinated biphenyls (PCBs) including planar, mono- and di-ortho congeners and concentrations of DDT were determined in striped dolphins affected by a morbillivirus epizootic in the western Mediterranean in 1990. Extremely high concentrations of PCBs ranging from 94 to 670 micrograms/g (wet wt) were detected in the blubber. Similarly, DDT concentrations were high, between 22 and 230 micrograms/g (wet wt). The concentrations of three non-ortho coplanar PCBs were 43 (3,3',4,4'-T4CB), 6.8 (3,3',4,4',5-P5CB), and 7.8 (3,3',4,4',5,5'-H6CB) ng/g (wet wt), respectively, the highest residue levels reported to date. The estimated 2,3,7,8-TCDD toxic equivalents of non-, mono- and di-ortho PCB congeners in striped dolphins were several times higher than those observed for other marine mammals and humans. Mono-ortho congeners contributed greater 2,3,7,8-TCDD toxic equivalents than non-ortho members. The higher ratio of 3,3',4,4',5,5'-H6CB/3,3',4,4',5-P5CB (IUPAC 169/126) suggested a strong induction of mixed function oxidase enzymes and highlighted the possibility of using this ratio as an index for risk assessment of PCB contamination in marine mammals. Elevated concentrations of PCBs may have played a role in the immune depression in striped dolphins, ultimately leading to the development of morbillivirus disease.

Changes in cardiac rhythm in man during underwater submersion and swimming studied by ECG telemetry.

A previously reported method for electrocardiographic (ECG) telemetry in water using frequency-modulated current was improved to obtain more stable ECGs. The ECGs of seven healthy men were monitored using the improved method during and after whole-body submersion or underwater swimming. Bradycardia and arrhythmias were observed during the submersion, and transient tachycardia was detected after the start of underwater swimming, followed by bradycardia with arrhythmias. Three different types of arrhythmias were observed: sinus arrhythmia (SA), supraventricular extrasystole (SE) and ventricular extrasystole (VE). SA and SE tended to develop during the latter half of the period of submersion or underwater swimming, and especially after the restart of breathing. VEs were detected in only one subject during submersion, whereas they occurred in most subjects during and after underwater swimming. Individual variations were found in development of arrhythmias, one subject showing no arrhythmia. Bradycardia, SA and SE could depend on vagal suppression in underwater conditions, and VE may be related to the effect of muscular movement on cardiac function in addition to vagal inhibition.

Temporal trends of persistent organochlorine residues in human adipose tissue from Japan, 1928-1985.

Trend monitoring of organochlorine contaminants, viz. PCBs, DDTs, HCHs and CHLs, was carried out by using formalin-preserved adipose tissue of Japanese males from 1928 through 1985 for understanding the long-term trends in their contamination levels. The highest residual concentrations were observed during the periods of maximum production (or import) and usage of these compounds in Japan. Time trends of the contaminants varied with the usage pattern and their physico-chemical properties. Effectiveness of government regulations on the production and use of organochlorines was seen faster in the case of DDTs and HCHs, which showed a prominent declining trend in their residues, whereas PCB levels exhibited a continuing increase and maintained a steady state even after two decades of a ban on their production, indicating that the exposure to PCBs is still prevailing. Generally, in terms of the spatial view, unlike aquatic fauna from the point-source environment (aquatic ecosystem) humans in the non-point-source environment (terrestrial habitat) revealed a slower rate of reduction in organochlorine residue burdens.

Embryonic abnormalities and organochlorine contamination in double-crested cormorants (Phalacrocorax auritus) and Caspian terns (Hydroprogne caspia) from the upper Great Lakes in 1988.

Persistent organochlorine contaminants including polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs) and polychlorinated biphenyls (PCBs) were determined in eggs with normal and deformed embryos collected in 1988 from different colonies during an epizootiological survey of double-crested cormorants (Phalacrocorax auritus) and Caspian terns (Hydroprogne caspia) from the upper Great Lakes. PCBs and p,p'-DDE were found in the highest concentrations in eggs of both species. The residue pattern of PCB isomers and chlordane compounds suggested that double-crested cormorants have greater metabolic capacity to degrade contaminants than Caspian terns. According to the toxicity evaluation using the 2,3,7,8-TCDD equivalents (TEQs) approach, non-ortho coplanar PCBs contributed much more toxicity than PCDDs and PCDFs. Total TEQ of dioxin-like compounds was likely associated with occurrence of live-deformed embryos in double-crested cormorants eggs. The toxic effects of these contaminants were also estimated in Caspian tern eggs, where elevated levels of coplanar PCBs, PCDDs and PCDFs were observed in concordance with increased rate of anomalies in eggs during a breeding season in the Great Lakes.
