Characteristics of C3H/He mouse embryo cell lines established by culture with or without benzaldehyde.

The characteristics of RME-5-3-1 cell line, which had been established from C3H/He mouse embryos by culture in benzaldehyde-containing medium, were compared with those of a benzaldehyde-untreated cell line, RME-5-1, derived from the same embryos as the former and with those of RME-5-1/TMT cell line, reestablished from the tumors induced by implantation of RME-5-1 cells into syngeneic mice. The characterization of these cell lines covered cell morphology, chromosome distribution, population doubling time, saturation density, fibronectin, epidermal growth factor receptor, plasminogen activator, ornithine decarboxylase, anchorage-independent growth and transplantability into mice. The results indicated that RME-5-3-1 cells had well-preserved normal phenotypes, while both RME-5-1 and RME-5-1/TMT cells showed malignant phenotypes to varying degrees.

[On the follow up period of postmolar patient (author's transl)].

We discussed how long should postmole patients be followed up by the analysis of 44 cases of postmolar invasive mole and 14 cases of postmolar choriocarcinoma treated in our hospital between 1971 and 1979. All of the invasive mole have been detected before their urinary hCG titer fell below normal pituitary LH level and by the 5 th month after the termination of molar pregnancy. While only 2 cases out of 14 cases of postmolar choriocarcinoma have been detected before their urinary hCG titer fell below LH level and by the 5 th postmole month. The other 12 cases of choriocarcinoma were diagnosed later than the 6 th postmole month. The longest interval between termination of mole and diagnosis of postmolar choriocarcinoma was 7 years and 2 months, the interval of which was also the longest among all of the postmolar choriocarcinoma registered to the Committee of Registration for Trophoblastic Disease in Japan Society of Obstetrics and Gynecology in 1974 through 1978. Therefore from the empirical point of view, the postmole patient should be followed up to 7 years and 2 months at the shortest. Besides almost all of the secondary sequelae after mole which were detected before urinary hCG titer fell below LH level were actually invasive mole (44 cases). Choriocarcinoma (2 cases) in this stage was exceptional (2 cases/46 cases = 4.4%). On the other hand, once the molar patient showed normal LH level in their urinary hCG determination, the secondary sequela was invariably choriocarcinoma. This way of discriminating invasive mole from choriocarcinoma is the key in the management of postmolar patient.

[Studies on neurobehavioral response (Scanlon test) in newborns after epidural anesthesia with various anesthetic agents for cesarean section].

In order to examine the influence of larger doses of anesthetic drugs on newborns, 146 infants delivered by cesarean section with epidural anesthesia employing various anesthetic agents were assessed by Apgar score, blood gas analysis, drug concentration as well as Scanlon's examination. The following results were obtained: 1. The fetal/maternal concentration ratio for bupivacaine was lowest compared with those of lidocaine and mepivacaine. 2. There was no significant difference among the epidural anesthesia with four kinds of anesthetic agents and spinal anesthesia groups in pH, Pco2, Po2 values or Apgar scores obtained from blood from the maternal artery or the umbilical vein. 3. The groups whose mothers had received epidural block with either lidocaine or mepivacaine for cesarean section showed significantly lower scores in Scanlon test compared with the groups with bupivacaine or chloroprocaine and spinal anesthesia group. From these findings, it may be concluded that bupivacaine or chloroprocaine does offer some advantages over (three local anesthetic agents) lidocaine, mepivacaine or tetracaine to the obstetric patients who need cesarean section for delivery.

Metastatic ameloblastoma of the skull.

A rare case of a metastatic ameloblastoma of the skull in a 29-year-old woman is reported. An ameloblastoma of the left mandible underwent histological malignant transformation following repeated operations and eventually metastasized to the left temporoparietal region of the skull. The skull tumor showed osteolytic change and marked vascularity in roentgenological examinations. It was removed totally. The histological findings were similar to those of the tumor of the left mandible. The route of the metastasis was considered to be via the left external carotid artery.

Neural regulation of interdigestive motor activity in canine jejunum.

The hypothesis that extrinsic innervation of the small bowel provides pathways for initiation and coordinated propagation of the interdigestive migrating contractions (IMC) was reinvestigated in dogs. Motor activity was measured by chronically implanted force transducers. After a control study, 40-cm segments of the jejunum were extrinsically denervated. All IMC migrated distally through the extrinsically denervated segments. Thiry loops were then constructed from the extrinsically denervated segments, and continuity of the intestine was restored by end-to-end anastomosis. IMC proximal to the anastomosis did not migrate through the extrinsically denervated loop but migrated to sites across the anastomosis. In the extrinsically denervated loop, bands of strong contractions, quite similar to the IMC, occurred at the orad end of the loop independent of the IMC and propagated distally to the caudad end of the loop. The duration, frequency, and migrating velocity of these bands of contraction were different from those of IMC. These results suggest that extrinsic innervation is not essential for the initiation and orad sequential propagation of periodic motor activity like IMC, even when intrinsic innervation is discontinued.

The clinical significances of carcinoembryonic proteins in patients with ovarian carcinoma.

Carcinoembryonic antigen (CEA), alpha-fetoprotein (AFP), placental alkaline phosphatase (ALP4), human chorionic gonadotropin (HCG), human chorionic gonadotropin-beta-subunit (beta-HCG) and human placental lactogen (HPL) were simultaneously and serially determined in the sera of the patients with ovarian carcinoma. The incidence of patients whose tests for carcinoembryonic proteins were positive was as follows: CEA, 29/55 (56.4%); AFP, 7/58 (13.5%); ALP4, 3/47 (6.4%); HCG, 20/50 (40%); beta-HCG, 13/50 (26%); HPL, 0. Determination of CEA in serum was considered to be a useful test for screening ovarian carcinoma and for follow-up studies, especially for so-called mucin-producing tumors (mucinous cystadenocarcinoma, Krukenberg's tumor, and pseudomyxoma peritonei). The specificity of AFP to ovarian carcinoma, especially in tumors of germ cell origin, seemed to be very high, thus AFP can be an extremely effective means for the diagnosis and the prognosis of patients with ovarian tumors arising from germ cells. But pure dysgerminoma, which was not positive in AFP, showed relatively high levels beta-HCG preoperatively, and its clinical course was associated with serial beta-HCG assay. There was no tumor specificity in positive HCG. ALP4 seemed to have little clinical significance because the detection was infrequent.

Immunofluorescence studies on complement components in the hair follicles of normal scalp and of scalp affected by alopecia areata.

The deposition of complement components (Clq, C4 C3, C5 and C9) and properdin in scalp hair follicles was examined by the immunofluorescence method in patients with alopecia areata and in normal subjects. C3, C5 and C9 were found deposited there. The complement deposition was most frequent in the case of C3 and less frequent in C9 and C5. There was no difference regarding deposition between normal subjects and patients with alopecia areata. No deposition of Clq, C4 or properdin was observed in the hair follicles of the scalp. These findings suggest a relationship between the hair cycle and the activity of C3 and its late complement components in the scalp.

Lack of effective messenger RNA for beta 2-microglobulin in a gestational human choriocarcinoma cell line (GCH-1).

When two lines of gestational human choriocarcinoma cells (GCH-1 and GCH-2) were incubated with [35S]methionine, the labeled beta 2-microglobulin was not detected on sodium dodecyl sulfate:polyacrylamide gel electrophoresis after purification of the labeled protein with affinity chromatography, using rabbit antibody against beta 2-microglobulin as the ligand. In contrast, in the case of nongestational choriocarcinoma cells originating from male stomach cells, the incorporation of [35S]methionine into beta 2-microglobulin was observed. When polyadenylate-containing messenger RNA prepared from GCH-1 cells was incubated with [35S]methionine in a rabbit reticulocyte lysate system, the labeling of beta 2-microglobulin was not shown. On the other hand, when polyadenylate-containing messenger RNA from nongestational choriocarcinoma cells was incubated in the same cell-free system, labeled beta 2-microglobulin was detected. These results indicate that beta 2-microglobulin is not synthesized in gestational human choriocarcinoma cells and at least in the case of GCH-1, beta 2-microglobulin is not synthesized owing to the lack of effective messenger RNA for beta 2-microglobulin.
