A 33-year-old woman with malignant melanoma well responded to a chemotherapy including DTIC, ACNU, and VCR, and intralegional injection of OK-432. Four years prior to admission, a lentigo of 5 mm diameter at her left frontal chest was found, but, histological examination resulted in no distinctly malignant findings. In November 1979, multiple subcutaneous tumors appeared over posterior surface of the chest and left axillar region, which were gradually increasing in number and size, then she was admitted to our hospital in June, 1980. Biopsy of subcutaneous tumors revealed a malignant melanoma and its metastasis to skin. Immunochemotherapy was started immediately based upon this diagnoses. The patient received 100mg DTIC i.v. for 4 days, 100mg ACNU i.v. for one day and 1 mg VCR i.v. for one day. OK-432 was locally injected into some tumors as an immunotherapy. On completion of the third course of chemotherapy, all subcutaneous tumors were decreased in size, especially the tumors injected with OK-432. However, patient didn't respond to a repeated chemotherapy, thereafter malignant melanoma was metasized to uterus and peritoneum in May, 1981, and she died in September, 1981. Major side effects of this chemotherapy were nausea and mild transient leukocytopenia. The combination of chemotherapy including DTIC and local injection of OK-432 appeared to be effective for malignant melanoma.

The stereochemistry of the decarboxylation of meso-alpha,epsilon-diaminopimelate catalyzed by meso-alpha,epsilon-diaminopimelate decarboxylase (EC 4.1.1.20) of Bacillus sphaericus was determined by stereochemical analyses of [6-2H]-L-lysine produced by the reaction in D2O. The product [6-2H]-L-lysine was converted to levorotatory methyl 5-phthalimido[5-2H]valerate by the reactions not affecting the absolute configuration of the asymmetric carbon atom. By contrast, methyl 5-phthalimido[5-2H]valerate derived from [2,6-2H2]-L-lysine, which was produced from [2,6-2H2]diaminopimelate by decarboxylation in H2O, was dextrorotatory. The authentic methyl (R)-5-phthalimido[5-2H]valerate prepared from L-glutamate with glutamate decarboxylase was levorotatory. These results indicate that the meso-alpha,epsilon-diaminopimelate decarboxylase reaction proceeds in an inversion mode. The deuterium label in [6-2H]-L-lysine was fully conserved during the conversion into pelletierine through [1-2H]cadaverine by the stereospecific diamine oxidase reaction. Thus, the enzymatic decarboxylation of meso-alpha,epsilon-diaminopimelate occurs with inversion of configuration in contrast to the other amino acid decarboxylase reported so far.
Effects of catecholamines, enkephalins and related compounds on electrical activity of the bed nucleus of stria terminalis (BST) were studied in vitro on thin BST sections prepared from guinea pig brains. Norepinephrine (NE) and epinephrine (E) suppressed field potentials elicited by a single shock to the stria terminalis (ST). The effects of NE and E were mimicked by phenylephrine and blocked by phenoxybenzamine. Isoproterenol and dichloroisoproterenol were without effect. NE and E suppressed the spontaneous firing of BST neurons and discharges elicited by ST stimulation. Dopamine was a less potent depressant. [D-Ala2]-Met-enkephalinamide (EKA) suppressed the field potentials and spike discharges elicited by ST stimulation. Spikes occurring spontaneously or during administration of glutamate were also suppressed by EKA. The action of EKA was blocked by naloxone. Late inhibition induced by stimulation of the lateral division of the ST was blocked by naloxone in about a third of the neurons examined. These results indicate that norepinephrine suppresses the activity of BST neurons by activating postsynaptic alpha-receptors. It is also suggested that opioid peptides mediate inhibitory control of the amygdala over the BST.

The new Kifa catheter decreased the rte of complications related to the nature of the catheter, enabled the examination to be performed more safely and reduced the examination time compared with the Cook catheter. The Kifa catheter for the right coronary artery could also be used for a subsequent left ventriculography.
Severe myocarditis was induced in inbred DBA/2 mice inoculated with M variant of EMC virus. Yellowish-white patches were seen on the surface of the ventricles and atria, and histologically, myocardial necrosis and calcification was evident on the fourth day after virus inoculation. Myocardial lesions appeared earlier and were more extensive than we observed in previous myocarditis induced by Coxsackie B viruses. Spontaneous perimyocardial lesions were observed in control DBA/2 mice, which were exclusively limited to the right ventricle; however, these lesions were different in localization from lesions observed in infected mice in which extensive myocardial lesions were noted. This animal model is considered to be excellent for studies on the pathogenesis and natural history of viral myocarditis.

Two transplantable granulosa cell tumor lines (designated as T and M lines) induced in mice by intrasplenic ovarian grafting were established. The hormonal milieu which promotes the growth of transplantable granulosa cell tumors was investigated in the present work. The inoculation of the T line tumors into host animals led to tumor formation in intact and gonadectomized animals, but not in hypophysectomized animals.Subcutaneous grafts in intact male hosts were significantly large than those in gonadectomized male hosts, whereas tumors of approximately equal size were formed in androgen-treated gonadectomized male hosts. These results indicate that both androgens and gonadotropins promote the tumor growth in vivo. The behavior of the M line tumors was similar to that of the T line tumors. When the cell growth of the M line tumors was examined in culture, steroid hormone depletion in medium did not affect the growth of the tumor cells, nor did androgen have a growth-promoting effect in serum-free medium or in medium supplemented with charcoal-extracted male mouse serum. It was concluded that the growth-promoting effect of androgens is not direct, and that androgens themselves or their derivative estrogens may produce specific growth factors, which in turn promote the tumor growth in vivo.

Effects of neurotensin on neurons in the bed nucleus of the stria terminalis (BST) were studied in vitro in thin brain sections of the guinea pig. Electrical stimulation to the stria terminalis (ST) elicited a negative field potential in the BST. The field potential was markedly suppressed in the medium containing Ca2+ at a low concentration of Mg2+ at a high concentration. BST neurons discharged in response to ST stimulation. Some of them also fired spontaneously. Neurotensin excited about two-thirds of BST neurons at concentrations of 0.35-35 nM. This excitation was not blocked when synaptic transmission was blocked in a medium containing Ca2+ in a low concentration and Mg2+ in a high concentration. These observations suggest that neurotensin is an excitatory transmitter or modulator in the amygdalo-BST projection.
