Circulating immune complexes in Takayasu disease.

Circulating immune complexes in Takayasu disease were investigated to assess the involvement of autoimmune mechanisms. Antibody dependent cell-mediated cytotoxicity (ADCC) and Raji cell assay revealed: The 10 percent inhibition of ADCC in 37 patients was 35.6 +/- 5.2, this being a non-statistically significant value as compared with the values of 25.6 +/- 3.0 in 33 apparently healthy controls. Raji cell assay values were 30.4 +/- 11.5 and 3.5 +/- 2.3 for patients and controls, respectively. As 54% of all these patients had a negative reaction and there was no correlation between immune complex levels and BSR, CRP, or ASO, immune complexes, while accelerating or modifying the pathophysiological state, probably are not primary causative factors.

Experimental coxsackievirus B3 perimyocarditis in the right ventricle in BALB/c mice: a one-year follow-up study.

We described severe perimyocarditis in weanling BALB/c mice inoculated with coxsackievirus B3 (Nancy strain) which was exclusively limited to the right ventricle. Yellowish-white patches were seen on the surface of the right ventricle of the heart on the seventh day through 360th day after inoculation with virus. Following myocardial degeneration and necrosis, marked perimyocardial fibrosis with calcification was observed 90 days after inoculation with virus and persisted through 360th day. Spontaneous pericardial lesions were observed in the right ventricle in a few control mice, but these changes were different in severity and localization from perimyocarditis observed in mice infected with coxsackievirus B3. This animal model may provide an opportunity to study the natural course of perimyocarditis of viral etiology and possible sequel chronic or constrictive pericarditis in humans.

Antigen-specific factors produced by carrier-primed New Zealand Black mice.

We studied antigen-specific suppression in NZB and B/W mice to investigate immunoregulatory circuits associated with autoimmune disease. Young NZB and B/W mice produced ample suppressor factor, and acceptor cells were present even in older mice. A low m.w. antigen-specific enhancing factor capable of augmenting the secondary in vitro IgG antibody response to DNP was found. These results indicate the complexity of immunoregulation in these autoimmune-susceptible mice.

A multivariate approach to feeling of fatigue in relation to individual workers' working and living conditions.

In an attempt to understand fatigue of industrial workers in an integrated framework of their daily life behavior, the occurrence of fatigue feeling during a one-month period was related to working and living conditions of individual workers. The data were collected using a questionnaire from every day for one month from 32 workers engaged in four different types of jobs in a chemical factory in Tokyo. Six kinds of daily behavior reported on via the questionnaire were taken as explanatory variables for occurrence of fatigue feeling, together with age, job type, and number of days from the last off-day. The statistical analyses including multi-discriminant analysis by Hayashi's Quantification II method disclosed that the workers' fatigue feeling was influenced by various factors, and that such behavioral variables as liquor intake and physical exercise on the previous day played decisive roles to the extent similar to age or job type variables.

Characteristics of in vitro production of antibodies to DNA in normal and autoimmune mice.

The in vitro production of antibodies to dsDNA was studied with spleen cells from normal and autoimmune mice. After culture for 4 days, the binding of dsDNA in the culture supernatant was measured by a radioimmunoprecipitation assay. The production of antibodies to dsDNA by spleen cells appeared at 15 hr after culture and reached a plateau at 24 hr. No antibodies were produced by thymus cells or splenic T cells. The specificity for dsDNA was shown by competitive inhibition with nonradioactive nucleic acids. Autoimmune strains of mice (NZB/NZW, BXSB, MRL/1) produced more antibodies to dsDNA than did several control strains. Young B/W mice and control strain mice produced mainly IgM antibodies, whereas older B/W mice produced predominantly IgG antibodies to dsDNA. The in vitro production of antibodies to dsDNA by aged B/W spleen cells was macrophage and T cell dependent.

Lymphocyte-potentiating factor from human peripheral blood adherent cells.

Low-molecular-weight factor, which can restore the diminished concanavalin A (Con A) response of adherent-cell-depleted human peripheral blood lymphocytes, was partially purified from the culture supernatant of human peripheral blood adherent cells by ion-exchange column chromatography, gel filtration and preparative paper chromatography. This factor apparently has a mechanism of action distinct from that of 2-mercaptoethanol. It affects the cells just before the onset of DNA synthesis possibly as one of the second signals in the Con-A-induced activation of human peripheral blood lymphocytes. However, it was demonstrated that the effective activation of lymphoyctes also required the presentation of Con A by adherent cells to lymphocytes at the triggering stage of the activation, indicating that adherent cells have two distinct functions in the Con-A-induced activation of lymphocytes, i.e. the the presentation of Con A to lymphoyctes and the elaboration of soluble factors.

Effect of salt loading and calcium antagonist on the pressor response to angiotensin infusion in the rat.

Elucidation of mechanisms of angiotensin tachyphylaxis will be necessary for explaning the continuous vasocontricting action of angiotensin in the vascular smooth muscle, which might be closely correlated to the high blood pressure in renin-dependent or vasoconstricting hypertension. From the results obtained sodium loading rats showed not only augmented pressor response to angiotensin but also inhibiting the appearance of its tachphylaxis under the infusion of Ca-antagonist dose enough to suppress the response to angiotensin in the rats of water drinking. Speculated mechanisms about this phenomenon has been discussed.
