Cross flow diafiltration of serum with basal medium suitable for growth of hybridomas, sterilization and protein reduction.

Fetal Calf Serum (FCS) was extensively extracted by cross flow diafiltration (Pellicon, Millipore) and sterilized using the basal growth medium (DMEM) for the extraction. Ultrafiltration membranes of 10(5) and 3 X 10(5) Dalton cut off were used respectively. The diafiltrates were used for hybridoma cultivation and the results of growth and mAb-production were compared with standard medium (DMEM + 5% FCS). Slightly reduced mAb-titers were achieved. These were, however, compensated by decreased concentration of contaminating protein and higher specific mAb/protein ratio as examined by SDS-PAGE and enzyme linked immuno electro transfer blot (EITB).

Myoadenylate deaminase deficiency: successful symptomatic therapy by high dose oral administration of ribose.

A 55 years old patient suffering from exercise-induced muscle pain and stiffness due to primary myoadenylate deaminase deficiency has been successfully treated with D-ribose since 1984: single doses of 4 grams administered at the beginning of exercise prevented the symptoms completely; on continuation of exercise this dose had to be repeated all 10-30 min. Total doses of 50-60 g per day were tolerated without side-effects.

Parental attitudes toward the developmentally disabled among Arab communities in Israel: a cross-cultural study.

The present study investigated the attitudes of 184 Arab families toward their developmentally disabled children, using a questionnaire on Attitudes Towards Disabled Persons, (Yuker, Block and Campbel, 1960). The major findings were that generally attitudes were positive, with the Druze showing the most positive ones, while the Muslims and Christians expressed less positive attitudes. A major factor that was found to correlate with less positive attitudes was education. In our sample the more educated (many Christians) showed less favorable attitudes. The main conclusion of this study was that since Arab society in Israel is dynamic, and old traditions are forespoken for new ones, it is important to preserve positive attitudes--which have their roots in past traditions--towards developmentally disabled by enhancing modern concepts relating to their education and rehabilitation.

Feeding resistance after parenteral hyperalimentation.

Infants requiring prolonged hyperalimentation for a variety of conditions may experience difficulty in establishing oral feedings. Indeed, active resistance to oral feeding is often observed. We describe an infant who was deprived of normal oral feedings for the first ten months of life. Because subsequent feeding resistance was apparently due to behavioral and developmental factors, we suggest that the approach to such cases should involve particular attention to these areas. The child development literature and our surgical experience with esophageal atresia give supporting evidence.

Dyssegmental dysplasia (report of two cases with a review of the literature).

Two cases of dyssegmental dysplasia, a rare lethal skeletal dysplasia in the newborn, are reported. Two different forms of dyssegmental dysplasia can be distinguished--the lethal Silverman type and the less severe Rolland-Desbuquois type. In this report, both cases are of the Silverman type. Histopathology of chondro-osseous tissue confirms the similarity in pathologic findings to Kniest dysplasia.

Motivation, vocational interests and job satisfaction of mentally retarded adults.

The relationship between vocational interests of 83 mildly to moderately retarded adults in a residential facility in Israel, their actual work and the factors which they perceived as the most important motivators for them at work and job satisfaction were investigated. Two questionnaires were used: the Illustrated Vocational Inventory (Whelan & Reiter, 1980) and a specially designed questionnaire on motivation to work based on Herzberg, Mausner, and Snyderman's (1959). The results demonstrate the importance of taking into consideration mentally retarded persons' vocational interests when assignig them to different jobs. It further demonstrates the importance of the environment in influencing mentally retarded individuals to seek instrinsic or extrinsic rewards and satisfaction from work.

Attitudes of superintendents of institutions for the mentally retarded towards deinstitutionalization: a cross-cultural perspective.

A cross-cultural study was conducted on the attitudes of superintendents of institutions regarding deinstitutionalization in the United States and in Israel. The attitudes of 29 superintendents from Israel were compared with the attitudes of 181 American superintendents. The most general finding was that in spite of important legislative and demographic differences, in both countries the attitudes were neither highly in favor nor strongly against deinstitutionalization. Most respondents in both countries believed that the parents of the mentally retarded do not generally support deinstitutionalization.

Production kinetics of monoclonal antibodies.

Two typical patterns of kinetics of monoclonal antibody (mAb) production are observed. Hybridomas which produce high mAb-titers do not show any feed back inhibition upon secretion of mAb into the culture supernatant over long periods of cultivation. "Low producing hybridomas" generally display high initial rates of mAb production, however, after a few hours the mAb secretion is stopped at a certain low concentration level in the order of some 100 mg mAb per ml. To illustrate the case that the high initial production rates of mAb of low producing hybridomas could be maintained for a period of at least 24 hours, mAb titers comparable to those produced by high-mAb-titer producing hybridomas were achieved.
