Stabilizing effect of reduced cultivation temperature on human x mouse hybridomas.

Interspecies hybridomas, such as human X mouse hybrid cells, usually show a high degree of unstability for monoclonal antibody (mAb) production. We investigated growth and specific rate of mAb-production during a period of 52 days after recloning human-mouse hybridomas at cultivation temperatures of 37 degrees C and 32 degrees C respectively. The lower cultivation temperature stabilized the overall IgM-mAb-production rate (IgM/ml) over the total cultivation period. At 37 degrees C incubation temperature the overall IgM-mAb-production was reduced to 1/3 due to loss of heavy chain production. This is visible from an increase of the kappa/mu-chain ratio. At 32 degrees C incubation temperature the specific IgM-mAb-production rate (IgM/cell X time) was stabilized, whereas at 37 degrees C incubation temperature a decrease of 80% was observed.

Monoclonal ELISA for the determination of BNYV-virus.

Monoclonal antibodies have been developed against the BNYV-virus (beet-necrotic-yellow-vein) which causes the rhizomania disease in sugar beet (Beta vulgaris). The B103 hybridoma produced predominantly IgG2a at greater than 100 micrograms/ml. IgM-production could not, however, be ruled out. In a polyclonal-monoclonal indirect sandwich ELISA, the B103-antibody bound to BNYVV of Austrian, German, French and Bulgarian origin at a sensitivity of 1 ng/ml or less. Comparison of sugar beet samples in ELISA and immuno-electronmicroscopy showed good correlation, demonstrating the good performance of B103 antibody for broad application in BNYVV diagnosis.

[In vitro and in vivo studies using 111 indium oxine, 111 indium oxine sulfate and 99m Tc oxine in erythrocyte labeling].

The optimal conditions for red blood cell labelling using 111indium oxine, 111indium oxine sulphate and 99mTc oxine were established both in vitro as well as in vivo. The coagulant had no effect on labelling efficiency. Other variables such as the incubation time, temperature, duration, cell number and concentration of the complex exert a significant influence on labelling efficiency. Labelling efficiency of red blood cells is very high also under non-optimum conditions as compared with other cells (leucocytes, platelets).

The intrainfusion platelet rebound during and following PGE1-infusion is faster and more intensive than that with PGI2.

During continuous long-term prostacyclin infusion an intra- and post-infusion rebound of platelets has been reported. During the present study 8 patients suffering from peripheral vascular disease in stage IIb according to Fontaine have been infused intravenously at a constant rate of 5 ng/kg/minute for seven days. Throughout this time the platelet function parameters were followed daily in the morning. An intrainfusion platelet rebound occurs using PGE1 as well. In comparison to the data for PGI2 published earlier it is noteworthy that the rebound for PGE1 occurs faster and more intensive. We conclude that for both the antiaggregatory prostaglandins PGE1 and PGI2, an intermittent treatment scheme seems to be optimal at the moment.

On the metabolism of allopurinol. Formation of allopurinol-1-riboside in purine nucleoside phosphorylase deficiency.

Allopurinol-1-riboside, a major metabolite of allopurinol, is commonly thought to be directly synthesized by purine nucleoside phosphorylase (PNP) in vivo. As this enzyme is otherwise believed to function in vivo primarily in the direction of nucleoside breakdown, we have determined by high performance liquid chromatography and a conventional chromatographic method the urinary metabolites of allopurinol in a child deficient of PNP. In this patient approximately 40% of urinary allopurinol metabolites consisted of allopurinol-1-riboside, thus proving the possibility of indirect formation of allopurinol-1-riboside via allopurinol-1-ribotide in vivo, catalysed by hypoxanthine guanine phosphoribosyltransferase (HGPRT) and a phosphatase.

Purification of HBsAg produced by the human hepatoma cell line PLC/PRE/5 by affinity chromatography using monoclonal antibodies and application for ELISA diagnostic.

The human cell line PLC/PRF/5 (5) was used for the production of hepatitis B surface antigen subtype ad (HBsAg ad) and purified by affinity chromatography (AC) with monoclonal antibodies (mAb). mAb to HBsAg from mouse ascites have been purified by Protein A - AC prior coupling to AH-Sepharose 4B (Pharmacia). The combined procedure of ammonium-sulphate-precipitation of HBsAg from culture supernatants and immunosorbent-AC leads to approx. 700-fold purification. ELISA results using the mAb and the HBsAg for diagnostics of human serum, positive for anti-HBsAg-antibodies correlate with the RIA (AUSAB, Abbott).

Tolerance for deviance: subjective evaluation of the social validity of the focus of treatment in mental retardation.

Staff members from an American institution, Israeli institutions, and a number of American group homes were asked to rate the social tolerability of 84 types of behavior that reflected either skill deficits or social maladaptation. There was considerable agreement among the three sample groups about the relative rank ordering of the 84 items. Physical violence and socially disgusting behavior were perceived as most intolerable whereas deficits in academic skills were least intolerable. The Israeli sample was uniformly less tolerant than either American sample.

Formation and degradation of deoxyadenosine nucleotides in inherited adenosine deaminase deficiency.

dATP, dADP, and dAMP equalled or exceeded the depleted levels of ATP, ADP, and AMP in erythrocytes from two children with adenosine deaminase (ADA; EC 3.5.4.4) deficiency. dATP and dADP were identified in the mononuclear cells of only one child. The levels of deoxyadenosine compounds fell dramatically after enzyme replacement therapy and were no longer detectable in the urine or in mononuclear cells. Erythrocyte adenosine nucleotide levels showed a corresponding increase. Intact erythrocytes prior to treatment contained adenine, presumed to be from deoxyadenosine degraded during extraction. Adenosine at high concentrations in vitro increased both dATP and ATP levels and decreased intracellular deoxyadenosine levels. There was no significant deamination of either [8-14C]adenosine or deoxyadenosine by intact ADA-deficient erythrocytes. About 90% of adenosine was metabolized to ATP at substrate concentrations from 10-100 microM, compared to 40-60% of deoxyadenosine metabolized to dATP. These studies suggest that (i) high intracellular deoxyadenosine levels may be necessary in vivo to sustain the raised dATP levels in ADA deficiency. (ii) When ADA is inhibited or absent, deoxyadenosine is removed rapidly from the circulation by the human erythrocyte utilizing an adenosine transport system linked to both ADA and adenosine kinase (EC 2.7.1.20).

Mentally handicapped children in special schools in Israel: a focus on social behavior.

As a part of a needs-assessment study, done in Israeli special schools for the mentally handicapped, social skills and personality characteristics were assessed on the Adaptive Behavior Scale (Nihira, Foster, Shellhaas, & Lelans, 1974). Structured observations were done in the classrooms of the behavior of each child and classroom climate questionnaire was administered to the children. Social behavior was found to be multidimensional consisting of: independence skills, personal maladaptive behaviors and social maladaptation. Correlations done between the ABS dimensions and the observations and classroom's climate, indicated that the more active and agitated child was less successful in terms of "vocational activity" and "self direction" (ABS domains), which were the behaviors most valued by the teachers. The more active child was also characterized by exhibiting positive and negative social behaviors, as indicated by responses given to the classroom climate questionnaire. The general implications of the study to social learning programs are that any curriculum in this area should take into consideration the multidimensionality of social behavior while at the same time take into account the socio-cultural pattern of the society and the personality makeup of mentally handicapped children.

Tomography of the internal acoustic meatus. A critical evaluation of the radiological appearance in normals and in patients with acoustic neuromas.

The radiological appearance of the internal acoustic meatus, obtained by tomography of the temporal bone, is presented for 115 patients with normal inner-ear function. These patients served as normal controls, and their tomograms are compared with those obtained in 48 patients who were initially suspected of having acoustic neuromas but in whom Pantopaque cisternography was negative. These are also compared with the findings obtained in 59 patients with surgically-verified acoustic neuromas. In normal persons the size of the vertical diameter of the meatus on the two sides, as measured by tomography, differed in only a few cases by more than 1 mm. The results obtained in patients who were suspected of having a neuroma, but in whom Pantopaque cisternography had excluded such a diagnosis, coincided completely with the results found in the normal group, and a difference of more than 1 mm. between the right and left sides is therefore indicative of a pathological process in the internal and acoustic meatus. A normal meatus does not exclude the presence of a tumour, since 10 per cent of patients with neuromas have identical meatus. No evidence of radiological destruction was found, either in the normal material or in the group of patients suspected of having a neuroma. Destruction of the bone surrounding the meatus must therefore be regarded as highly suggestive of the presence of a tumour, but lack of destruction does not exclude a tumor. Tomography of the temporal bone should be applied routinely in the search for acoustic neuromas, in patients with unilateral acoustic or vestibular complaints. The evaluation of the tomograms, however, should be performed by a few, specially interested radiologists.

Leisure activities of mentally retarded adults.

Interviews were conducted with 44 moderately and mildly retarded adults about their leisure activities. Comparisons were made between these adults and nonretarded adults from the general population (Katz & Gurevitz, 1976) who were matched for age and amount of schooling completed. Results showed that the retarded adults went out in the evenings and engaged in social visiting significantly less frequently than did the general population. The two groups did not differ significantly with regard to participation in a variety of daytime activities, such as sports and excursions, or in a variety of home activities.

Computed tomography in soft tissue disorders of the lower extremities.

Computed Tomography (CT) constitutes an important new imaging modality in the evaluation of soft tissue tumors. They are detectable by CT because their density is different from that of muscles and because they cause displacement of normal structures. CT was found to be useful in demonstrating the true extent of lesions in the soft tissue of the lower extremities of 24 patients as confirmed by examination of the surgically removed tumors. CT was also able to define the lesions' relation to and involvement with the surrounding tissue. We were not able to distinguish between benign and malignant lesions by CT. Generally, the specific attenuation values were not helpful in differentiation between various types of tumors. However, in cases of tumors such as lipomas, cysts and neurofibromas it may be of some use.

Computed tomography of musculoskeletal disorders of the truncus.

Computed Tomography (CT) is a new diagnostic tool in the evaluation of the musculoskeletal system. In the present study 29 patients with benign or malignant lesions in the body have been examined by this method. The purpose has been to evaluate the value of CT scanning in disorders of the musculoskeletal tissues. CT is an effective modality for demonstrating the three-dimensional extent of lesions, but in about one-third of the cases the examinations were not able to distinguish between benign and malignant lesions.
