A horseradish peroxidase study of rat lingual motoneurons with axons passing through the cervical nerve.

The peripheral course of axons of rat lingual motoneurons was studied by HRP injection into the hypoglossal nerve in combination with transecting of the hypoglossal and/or cervical nerve components of the hypoglossocervical plexus. Furthermore, soma sizes of labeled lingual motoneurons were compared in transverse section with those of labeled geniohyoid and thyrohyoid motoneurons, which are situated adjacent to the lingual motoneurons. We found that axons of the majority of lingual motoneurons lying in the main hypoglossal nucleus passed through the hypoglossal nerve throughout their course to the tongue. In a remaining small number of lingual motoneurons lying in a medial portion of the ventromedial subnucleus in the caudal fourth of the main hypoglossal nucleus, their axons passed through the first cervical nerve to the upper root of the ansa cervicalis to the hypoglossal nerve and then to its medial branch. The labeled lingual motoneurons with axons passing through the cervical nerve were intermingled with those whose axons passed through the hypoglossal nerve. The latter motoneurons, however, diminished in number while being traced caudally, and finally in the most caudal main hypoglossal nucleus the former motoneurons occupied a major part of this nucleus. The lingual motoneurons with axons passing through the cervical nerve were smaller in soma size than those with axons passing through the hypoglossal nerve. These two types of lingual motoneurons were both smaller in soma size than the geniohyoid and thyrohyoid motoneurons, and their soma shape was not as flat as that of the latter types of motoneurons.

Ornithodoros moubata: host immunoglobulin G in tick hemolymph.

Hemolymph proteins of a soft tick, Ornithodoros moubata, were analyzed immunochemically and biochemically. The components of tick hemolymph proteins were shown to be totally different from the host (rabbit) serum proteins by polyacrylamide gel electrophoresis with sodium dodecyl sulfate and Coomassie blue or silver stain. However, in the hemolymph of ticks engorged from rabbits immunoglobulin G was detected by immunoblotting analysis with goat anti-rabbit immunoglobulin G. The concentration of rabbit Immunoglobulin G in tick hemolymph changed with the physiological stages after a blood meal. Immunoglobulin G was isolated from tick hemolymph by affinity chromatography on a Protein A-Sepharose 4B column. Analysis of the isolated immunoglobulin G from tick hemolymph with sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Ouchterlony double diffusion test showed it to be composed of the same subunits as heavy and light chains of host (rabbit) immunoglobulin G. Tracer experiments showed that 125I-labeled heavy and light chains of immunoglobulin G were detected in an intact form in hemolymph from ticks that sucked 125I-labeled rabbit immunoglobulin G through an artificial membrane. These facts suggested that the host rabbit immunoglobulin G ingested in the tick midgut passed through the gut wall without digestion. By solid-phase enzyme immunoassay, immunoglobulin in the hemolymph was shown to retain its antibody activity.

Zoonotic subcutaneous filaria of undetermined classification.

A necrotic adult male filaria was found in sections of a dense, fibrous, subcutaneous nodule excised from below the breast of a 61-year-old woman, resident of Mie Prefecture in Honshu, Japan. Owing to its necrotic condition, and to a lack of essential knowledge of the comparative micro-anatomy of filarial worms, a differentiation between Dirofilaria and Dipetalonema species, or member of a related group, was not possible.

Effect of coenzyme Q10, a quinone derivative, on guinea pig lung and tracheal tissue.

In the present investigation, the effect of coenzyme Q10 (E-0216) on histamine or SRS-A (slow-reacting substance of anaphylaxis) release from passively sensitized guinea pig lung tissue and on the actions of various bronchoactive-agents in isolated perfused guinea pig tracheal strips was studied. The amount of histamine and SRS-A released from passively sensitized guinea pig lung tissue by the challenge of antigen showed a marked decrease by preincubating with coenzyme Q10. The percentage inhibition of histamine and SRS-A was biggest at the concentration of 10 micrograms/ml of coenzyme Q10. Prostaglandin F2 alpha-, acetylcholine-, serotonin-, and bradykinin-induced contractile responses in guinea pig tracheal strips did not show any significant increase by continuous infusion of coenzyme Q10 (0.05-5.0 micrograms/ml). On the other hand, histamine-induced contractile responses showed a significant increase by continuous infusion of 0.5 microgram/ml of coenzyme Q10. Isoprenaline (isoproterenol)- and salbutamol-induced relaxation responses in guinea pig tracheal strips did not show any significant decrease by continuous infusion of coenzyme Q10 (0.05-5.0 micrograms/ml). On the other hand, prostaglandin E2-induced relaxation responses showed a significant decrease by continuous infusion 0.05-5.0 micrograms/ml of coenzyme Q10.

[Cerebral blood flow, cerebral oxygen metabolism, cerebral glucose metabolism, and tissue pH in human acute cerebral infarction using positron emission tomography].

It is necessary for treatment and deciding prognosis to make clear about changes of cerebral blood flow and metabolism in acute cerebral infarction. This preliminary PET study was designed to investigate physiological and biochemical changes in acute cerebral infarction by positron emission tomography (PET). PET studies were performed in six patients with acute cerebral infarction within 48 hours after onset of stroke using continuous inhalation of C15O2 for cerebral blood flow (CBF), 15O2 for cerebral metabolic rate for oxygen (CMRO2), 11CO for cerebral blood volume, the intravenous injection of 11C-dimethyloxazolidinedione for tissue pH and the intravenous injection of 18F-fluorodeoxyglucose for cerebral metabolic rate for glucose (CMRGlu). Metabolic coupling index (MCI) image was made from CBF image and CMRGlu image to investigate relation between CBF and CMRGlu. Also oxygen glucose index (OGI) image was made from CMRO2 image and CMRGlu image to investigate relation between CMRO2 and CMRGlu. Preliminary results demonstrate that reduction of CBF, CMRO2, and CMRGlu in the affected cortex except for reperfusion case. Increase of OER was recognized four of six cases. Patterns of MCI and OGI in the cortex which CMRO2 value is less than 65 mumol/100 g/min were different from those in the cortex which CMRO2 value is more than 65. MCI of the affected cortex (CMRO2 less than 65) decreased relative to that of the cortex (CMRO2 greater than or equal to 65). OGI of the affected cortex (CMRO2 less than 65) significantly decreased in comparison with that of the cortex (CMRO2 greater than or equal to 65).(ABSTRACT TRUNCATED AT 250 WORDS)

Severe Kawasaki heart disease treated with an internal mammary artery graft in pediatric patients. A first successful report.

Two Japanese boys, 6 and 10 years old, required operation for severe Kawasaki heart disease. Both had multiple coronary arterial aneurysms and stenoses, and one had mitral regurgitation as well. The operations consisted of anastomosis between the left internal mammary artery and the left anterior descending artery and insertion of an autologous saphenous vein between the aorta and the posterior descending artery in both patients. The mitral valve was replaced in the one with mitral regurgitation. Angina pectoris has been completely relieved as confirmed by postoperative angiocardiography. Late results of coronary bypass grafting with the saphenous vein in pediatric patients with Kawasaki disease have been less than satisfactory because of the high occlusion rate of the graft. The internal mammary artery may be superior, at least theoretically, to the saphenous vein graft because it is a "living graft" with a high possibility of growing and with less susceptibility to degeneration. This is the first report of successful operation utilizing an internal mammary artery graft in coronary bypass for Kawasaki disease.

[Cerebral blood flow and metabolism in multi-infarct dementia].

Cerebral blood flow and oxygen metabolism were studied in three aged normal volunteers and 10 patients with multi-infarct dementia (MID) by Positron Emission Tomography using O-15. The diagnosis of MID was done according to the Loeb's modified ischemic score and X-ray CT findings. The MID patients, whose X-ray CT showed localized low density areas in the subcortical white matter and basal ganglia and thalamus, were studied. No occlusion was observed at anterior cerebral artery and/or middle cerebral artery on cerebral angiography. All cases of MID were mild dementias. Regional CBF, rOEF and rCMRO2 were measured by the steady state technique described by Terry Jones et al. The values of rCBF in MID patients were significantly low compared with those of aged normal subjects in frontal, temporal, occipital, parietal cortices and thalamus. The values of CMRO2 in MID were significantly low in frontal, temporal, occipital cortices and thalamus compared with normal subjects'. The OEF was 0.46 in aged normal subjects, and 0.52 in MID patients. The MID patients in the early stage of dementia showed the increased oxygen extraction fraction, and this fact suggests that ischemia is a significant pathogenic mechanism in the production and progression of multi-infarct dementia. The decrease of CBF and CMRO2 in MID compared from normal subjects' were most remarkable in frontal cortex. The impairment of mental functions in MID should be caused by the decreased neuronal activities in frontal association cortex.

[Effect of aortocoronary bypass surgery on left ventricular function and coronary sinus blood flow in patients with severe left ventricular dysfunction].

Left ventricular function and coronary sinus blood flow were evaluated in 7 patients with severe left ventricular dysfunction at rest before and after aortocoronary bypass surgery, and during exercise after surgery. Same evaluations were performed in 8 normal subject (G-C). Cardiac index (CI) at rest (2.09 +/- 0.55 l/min/m2) significantly increased after operation (2.94 +/- 0.59 l/min/m2) (p less than 0.02). There was no difference between CI during exercise after operation (5.94 +/- 1.51 l/min/m2) and that in G-C. Left ventricular end-diastolic pressure (LVEDP) at rest before operation (16 +/- 8 mmHg) was significantly higher than that in G-C (p less than 0.05). This difference disappeared after operation. LVEDP during exercise after operation (25 +/- 10 mmHg) was significantly higher than that in G-C (p less than 0.01). Coronary sinus blood flow (CSF) at rest (73 +/- 15 ml/min) significantly increased after operation (123 +/- 44 ml/min) (p less than 0.02). There was no difference between CSF during exercise after operation (282 +/- 99 ml/min) and that in G-C. These data indicated that the aortocoronary bypass surgery was effective on left ventricular function and coronary sinus blood flow in patients with severe left ventricular dysfunction.
