A family of hitherto undescribed sialoglycoproteins has been isolated from the eggs of Pacific herring in a yield about 10 times as much as phosvitin known as a major protein component of the eggs. The sialoglycoproteins are composed of 60-50% of carbohydrates and 40-50% of protein. The carbohydrate composition is similar to that of glycoproteins known to have complex types of heterosaccharide units whereas the amino acid composition is unique. The molecular weights ranging over 8800-13000 were determined for the sialoglycoprotein fractions by sedimentation equilibrium. The low mobility of the sialoglycoproteins during sodium dodecyl sulfate gel electrophoresis is discussed in terms of low values of hydrophobicity.
Extensive necrosis of the skin, muscle and bone was produced after prolonged cooling in a case of myeloma with cryoglobulinemia. Skin grafting was beneficial.
To investigate the mechanism of the development of insulin resistance in hypothalamic obesity, we studied insulin binding, glucose transport, and glucose oxidation in adipocytes from ventromedial hypothalamus (VMH)-lesioned rats 1 and 2 weeks after injury. One week after injury, insulin binding and insulin-stimulated glucose oxidation were increased, but insulin-stimulated glucose transport was similar to control. Two weeks after injury, insulin binding and insulin-stimulated glucose transport were similar to control, but insulin-stimulated glucose oxidation was decreased. Thus, cellular insulin responsiveness was increased in adipocytes from VMH-lesioned rats 1 week after injury; this increase in insulin responsiveness was due mainly to the increased intracellular glucose metabolism. On the other hand, cellular insulin resistance existed in adipocytes from VMH-lesioned rats 2 weeks after injury; this insulin resistance was also due mainly to a defect in intracellular glucose metabolism.
Twenty-four-hour variations of plasma hPP levels together with plasma glucose, IRI and GI levels were studied in four normal human subjects. Before examination, their food intakes were strictly controlled during two days' hospitalization. Plasma hPP values were assayed by a radioimmunoassay. They showed sharp peaks after each meal. Mean levels of hPP just prior to breakfast were 88 +/- 26 pg/ml (SEM), and the peak values of the plasma hPP one hour after each meal were 307 +/- 124 pg/ml, 398 +/- 77 pg/ml, and 544 +/- 118 pg/ml, respectively. During the nocturnal sleeping period the hPP levels were stable and returned to the fasting level of the previous morning. IRI increased significantly after each meal, but no significant change was found in plasma GI levels.
The influence of inflammation on the motility of the human gallbladder was investigated through an in-vitro experiment. Motility of longitudinally cut strips from 50 patients were examined by the same method as described in our previous articles. We pathohistologically classified the severity of the inflammation of the specimens into four grades: grade 0 is normal, grade I, slightly inflamed; grade II, moderately inflamed; grade III, highly inflamed. Relationships between the grade of inflammation and the motility of the strips were summarized as follows. 1) In grade 0 cases, the strongest tonic contractions were observed in the strips from the fundus. This tendency was also found in cases of grade I and II but III inflammation. 2) The tonic contractions of the strips from the fundus or body decreased along with the grade of the inflammation. 3) In any strips, the frequency of the rhythmic contractions decreased progressively with the grade of the inflammation. 4) The amplitudes of rhythmic contractions of the strips with grade I and II inflammation were larger than those of grade 0 inflammation.
The serum gastrin responses after ingestion of hard boiled eggs and Nutrient Broth solution were examined in peptic ulcer patients and normal subjects. The values observed of the serum gastrin response to these test meals were compared and contrasted. A significant enhancement of gastrin release in response to protein was obtained. There were no significant differences between the releases of gastrin after the two test meals.
Hemodialysis leukopenia was studied using various dialyzers and membranes. We found that dialyzers with cellulosic membranes caused marked leukopenia, but in recently developed non cellulosic membranes, its occurrence was significantly less. Additionally, our results showed a newly developed cellulose acetate membrane to correlate well with the non cellulose membranes regarding leukopenia, in spite of it being a derivative of cellulose. The extent of white blood cell decrease seemed to correlate inversely with an increase in the ultrafiltration rate per membrane area of dialyzer. The relationship between leukopenia and hypoxemia was examined also. The results of this investigation are included, however, we feel that more research is necessary before any conclusions can be made.
The study was performed in 70 patients with maxillary cancer to evaluate how far the long-term survival rate of the maxillary cancer can be improved with maximum combination of radiation therapy using 6 mV Linac X-ray, intra-arterial chemotherapy using 5-fluorouracil, sufficiently extensive maxillectomy and primary reconstruction. In 52 cases of squamous cell carcinoma, the 3-year and 5-year actual survival rates were 85.2% and 71.1% respectively. In the total 70 cases including those of undifferentiated carcinoma and adenocarcinoma, the 3-year and 5-year actual survival rates were 75.2% and 64.1% respectively. Primary reconstruction, which consisted chiefly of covering the entire wound surface with the deltopectoral flap, enabled us to perform as much resection of the subcutaneous tissue of the facial skin, tissue around the mandible and cranial base as necessary, without consideration of the postoperative facial disfigurement and functional impairments.

The effect of 5-S-cysteinyl-L-3,4-dihydroxyphenylalanine (cys-dopa), an intermediate in the pathway from L-3,4-dihydroxyphenylalanine (L-dopa) to pheomelanin, on the growth of eight human tumor cell lines in culture was compared to that of L-dopa. The tumor cell lines tested comprise two neuroblastomas (NB-1 and YT-nu), two amelanotic melanomas (HMV and SEKI), a gastric carcinoma (MKN-28), and three squamous cell carcinomas (HeLa-S3, KB, and a salivary gland carcinoma). Cys-dopa at a concentration of 1 mM inhibited growth of NB-1 (66%), YT-nu (67%), HMV (44%), SEKI (60%), MKN-28 (47%), HeLa-S3 (24%), KB (64%), and salivary gland carcinoma (33%), while L-dopa exhibited similar or even lower degree of inhibition at a concentration of 6 mM. On the other hand, both catechols had little effect on the growth of two fibroblasts derived originally from normal tissues (mouse fibroblast L929 and Chinese hamster fibroblast Don-6). Cys-dopa and L-dopa inhibited DNA and protein synthesis in YT-nu cells, but RNA synthesis was less affected. Treatment with cys-dopa at a dose of 1000 mg/kg i.p. for 7 days prolonged by 50% the life span of mice inoculated with L1210 leukemia. Normal mice given cys-dopa at a dose of 1000 mg/kg for 12 days showed no signs of toxicity. These results suggest the potential of cys-dopa as an antitumor agent.
