Ga-67 scan in patients with intrathoracic esophageal carcinoma planned for surgery.

The authors evaluated Ga-67 scan in 68 patients with intrathoracic esophageal carcinoma initially planned for surgery. Of these, 59 patients were staged pathologically or surgically; their Ga-67 scan results correctly predicted the presence of extraesophageal spread and lymph node metastases. Of 38 clinical Stage II patients, 15 (39%) could be Stage III by the results of Ga-67 scan. However, two of four clinical Stage I patients (50%) and 35 of 38 clinical Stage II patients (92%) were eventually pathologic or surgical Stage III. This high conversion rate and the high incidence of Stage III in esophageal carcinoma patients may not justify routine use of Ga-67 scan only for staging. Ga-67 scan, however, was useful for planning radiotherapy, because missing extraesophageal extension or gross metastases from the radiation fields were detected in six patients of 51 so treated. Ga-67 scan has its value in some patients with intrathoracic esophageal carcinoma.

Amylase released from the parotid gland by pilocarpine elevates the enzyme activity in the submandibular and sublingual glands of rats.

Stimulation by pilocarpine led to a marked increase in amylase activity in the submandibular and sublingual glands and serum, and resulted in a considerable depletion of the enzyme in the parotid gland and pancreas of rats. Parotidectomy reduced these pilocarpine effects. It is concluded that amylase released by pilocarpine from the parotid gland but not from the pancreas elevated the enzyme activity in the submandibular and sublingual glands.

Determination of peptidoaminobenzophenone (2-o-chlorobenzoyl-4-chloro-N-methyl-N'-glycylglycinanilide) and its metabolites in human plasma by glass capillary gas chromatography negative ion chemical ionization mass spectrometry--1.

A highly sensitive and selective glass capillary gas chromatographic negative ion chemical ionization mass spectrometric assay was developed to measure peptidoaminobenzophenone(2-o-chlorobenzoyl-4-chloro-N-methyl-N'-glycylglycinanilide) and its metabolites; chlorodiazepam, chlorodesmethyldiazepam and lorazepam in human plasma. As peptidoaminobenzophenone underwent pyrolysis during gas chromatography, it was converted to a thermally stable aminoquinolone. Internal standards for these compounds were the respective deuterium labelled compounds. Calibration curves were prepared for the range of 1-100 ng ml-1. Interference by endogenous substances was negligible in the isobutane negative ion chemical ionization mode in contrast to the electron impact or positive ion chemical ionization mode. This method was used to determine the plasma levels in humans following oral administration of two 5 mg doses.

A case report of congenital hypomyelination.

A 3 year, 11 month-old Japanese male with congenital hypomyelination is described. Clinical features are delay of motor development, generalized muscle hypotonia and weakness, absent tendon reflexes due to peripheral neuropathy, and normal mental development. Electrophysiologically, nerve conduction velocities could not be measured. Histological examination of the right sural nerve revealed total of absence myelin of most of the myelinated fibers. Electronmicroscopically, there was a concentric network of lamellae formed by double-layered sheets of basement membranes with fragments of Schwann cell cytoplasm around the myelinated fibers, so called "onion-bulbs". These peculiar features were similar to those in the cases reported by Lyon, (1969); Kennedy et al., (1977); Karch et al., (1975); and Anderson et al. (1973).

Hepatic bile acid elution by albumin and bile acid content in isolated rat hepatocytes.

Bile acid contents were determined for isolated rat hepatocytes. During the course of isolating the hepatocytes, perfusion of rat liver with buffer containing 2% albumin eluted a significant amount of bile acids. The elution was proportional to the volume of the buffer and attributable to albumin in the buffer. The isolated hepatocytes prepared by perfusion with 0.1% albumin buffer, which eluted a negligible amount of bile acids, contained 95 +/- 12 micrograms/10(8) cells of bile acids. The major bile acids were cholic acid (22%), beta-muricholic acid (34%) and hyodeoxycholic acid (10%). Levels of the other bile acids were less than 3%. Peak 8, unidentified but presumed to be a trihydroxycholanoic acid, accounted for 19%.

Mechanisms of increase in amylase activity in rat submandibular and sublingual glands after administration of pilocarpine.

The increase in parotid rather than pancreatic-type amylase activity in the submandibular and sublingual glands of rats caused by administration of pilocarpine was abolished or diminished when pilocarpine was injected into rats which had been parotidectomized, sympathectomized by superior cervical ganglionectomy or pretreated with reserpine. These results suggest that the increases in amylase activity in the submandibular and sublingual glands by pilocarpine are not due to increase in enzyme synthesis, but to uptake of enzyme released into the blood in large quantities from the parotid gland and that the release from the parotid gland by pilocarpine is primarily mediated by sympathetic nerves.

Lysozyme dimer formation on lysozyme oxidation with Br2 radical as studied by fluorescence evolution.

Lysozyme dimers produced on oxidation of lysozyme with Br2 radical in aqueous solutions exhibit a fluorescence spectrum (lambda max = 400 nm) closely similar to that of bi-tyrosine. This suggests that the dimer is likely to have a tyrosine-tyrosine bond resulting from the combination of tyrosine phenoxy radicals of two lysozyme molecules. Kinetic studies on dimer formation were made by measuring time-dependent fluorescence after pulsed-electron irradiation over wide pH range. The results lead to the following conclusions. The second-order growth of the dimer fluorescence observed at pH 10.7-12.5 reflects the combination process of the lysozyme radicals, which is rate-determining in the pH range. On the other hand, the first-order growth observed at pH 6.8-10.2 is attributable to the enolization of the keto-form of the dimer. A tentative reaction scheme is proposed for the dimer formation.

Identification of adenovirus 2 early region 4 polypeptides by in vitro translation and tryptic peptide map analysis.

The mRNA species encoded by early region 4 (E4) (map position [mp] 91.5 to 99.3) of adenovirus 2 were isolated from the polysomes of infected KB cells and were purified by hybridization to the cloned HindIII-F fragment (mp 89.5 to 97.3) or to EcoRI-C fragment (mp 89.7 to 100). The mRNA's were translated in vitro using [35S]methionine as a labeled precursor in rabbit reticulocyte lysates treated with micrococcal nuclease as well as in wheat germ lysates. Five major (35,000-molecular-weight [35K], 23K, 22K, 21K, 18K) polypeptides were observed when the reticulocyte lysate was used. The 23K, 22K, 21K, and 18K polypeptides were also observed with the wheat germ lysate, as well as a very prominent 11K polypeptide; the 35K polypeptide was not observed. Assignment of these polypeptides to E4 was further established by hybrid arrested translation. Two-dimensional gel electrophoresis of a wheat germ translate resolved five polypeptides ranging from 18K to 23K, the major 11K polypeptide, and polypeptides of 10K and 9K. The in vitro 23K to 18K and 11K polypeptides migrated to approximately the same positions on two-dimensional gels as did seven 26K to 21K polypeptides and an 11K polypeptide synthesized in vivo (Brackmann et al., J. Biol. Chem, 255:6772--6779, 1980). Two-dimensional tryptic peptide maps demonstrated that the 35K, 23K, 22K, 21K, and 18K polypeptides are related. The peptide map of 11K is different from those of the above polypeptides, although 11K may share one tryptic methionine polypeptide with them. These results indicate that E4 encodes a major 11K polypeptide, as well as major 35K, 23K, 22K, 21K, and 18K polypeptides.

Right ventricular aneurysm due to myocardial fatty infiltration: report of a case.

A case of right ventricular aneurysm due to myocardial fatty infiltration was reported. A 39-year-old man was admitted to our hospital for evaluation of several episodes of syncopal attack presumably induced by malignant arrhythmia. There were frequent observations of ventricular premature beats, which occurred occasionally in couples. Cardiac catheterization disclosed the aneurysm situated at the outflow tract of the right ventricle. Aneurysmectomy was performed and the ventricular premature beats and syncopal attacks were effectively abolished. Etiology of the aneurysm was proved to be a fatty infiltration in the myocardium. To the best of our knowledge, there has been no case report of right ventricular aneurysm due to fatty infiltration in the myocardium.
