[Isolation of cisplatin-resistant subline from human ovarian cancer cell line and analysis of its cell-biological characteristics].

The cisplatin (CDDP)-resistant subline TYK-nu (R) was developed by culturing TYK-nu (human ovarian cancer cell line in vitro) with exposure to CDDP in stepwise increasing concentrations. The characteristics of both cell lines were compared and the results were as follows; 1. Both cell lines formed a monolayer in a pavement-like arrangement and large cells were occasionally present in TYK-nu (R) rather than TYK-nu. The population doubling time of TYK-nu and TYK-nu (R) was 43 hr and 48 hr, respectively. 2. IC50 (microgram/ml) in 96 hr treatment with CDDP and carboplatin (CBDCA) was 0.035 and 0.5 in TYK-nu and 0.62 and 2.0 in TYK-nu (R), respectively. 3. In the intracellular CDDP concentration, there was no marked difference between TYK-nu and TYK-nu (R) after CDDP (2.0 micrograms/ml) treatment in 2hr. 4. After treatment with CDDP (0.2 microgram/ml) or CBDCA (2.0 micrograms/ml), a decrease in S and G2 + M compartments was observed in the pattern of the DNA histogram of TYK-nu, but not in that of TYK-nu (R). 5. The majority of chromosomes in both cell lines were in hyperdiploid areas and the mode of TYK-nu and TYK-nu (R) was 56 and 51, respectively. The karyotype of TYK-nu (R) showed deletion of chromosome 7q. Thus, compared to TYK-nu, TYK-nu (R) was 17.7 times more resistant to CDDP and 4 times more resistant to CBDCA. The results suggest that the mechanism of resistance to CDDP may be due to genetic changes at the cellular level.

[A comparative study of CT and plain X-ray film in the diagnosis of interstitial pulmonary disease in patients with rheumatoid arthritis].

In order to detect the interstitial pulmonary disease of rheumatoid arthritis (RA), we studied 91 patients with RA and compared the appearances of high resolution CT with those of plain chest X-ray film. We found 43 cases of interstitial pneumonitis on CT. We classified the CT and X-ray appearances as 4 groups (0-3) based on the degrees of pulmonary lesions. The both classifications accorded only in 46 cases (51%). In 46 cases who were not detected any changes by plain X-ray film, 17 were detected interstitial changes by CT. As to the early interstitial changes, the detectability of CT was superior to that of plain X-ray film.

[Pharmacokinetics of antimicrobial agents in the elderly].

The elderly can be easily infected by certain organisms due to underlying diseases and complications. The pharmacokinetics of cefotiam, ceftriaxone, ceftizoxime, cefpodoxime proxetil, carumonam, clarithromycin, amikacin, ofloxacin and lomefloxacin in the elderly should be considered when choosing antibiotics for elderly patient with regard to infected site, causative organisms, pharmacokinetics and side effects. Pharmacokinetics in the elderly reflect potential renal dysfunction, so that t1/2 was elongated and AUC increased with age. Intravenous administration in the elderly, especially in the subjects with low body weight, the administered dose and intervals must be considered. With oral administration there was no constant tendency of the intestinal absorption.

[Fate of the false lumen after surgery and assessment of operative procedure for DeBakey IIIb dissecting aortic aneurysm].

Eighteen patients with DAA IIIb were divided into two groups. Seven patients with major abdominal arteries originated from true lumen (group I) and 11 ones with renal artery or arteries from false lumen (group II). group I: In early postoperative phase (25-55 days), the descending aortic false lumen had been thrombo-occluded in all patients and the upper abdominal aortic false lumen had been still enhanced in three. In late postoperative phase (9-35 mos.), false lumen had been disappeared in three patients, so those aortae appeared normal morphologically and in other three patients thrombo-occluded false lumen reduced in size. group II: There were re-entries and the false originated renal arteries were well perfused in all patients. In nine patients with no leakage at anastomotic site, these descending aortic false lumens were thrombo-occluded. But as in the upper abdominal aortic false lumens, there was still enough blood flow to perfuse the false originated renal arteries. These suggest that the complete entry closure is the most important, so we recommend to graft the descending aorta containing entry with prosthesis, and this operation leads false lumen to 1) thrombo-occlusion, 2) absorption of thrombus and finally 3) normalization of injured aorta morphologically.

[Surgical treatment of atherosclerotic aneurysms of the thoracic aorta].

To determine the problem of the surgical treatment of atherosclerotic aneurysm of thoracic aorta, we reviewed our experiences with 113 consequtive patients who underwent thoracic aortic aneurysmectomy from January 1983 to December 1985. Fifty-two patients had atherosclerotic aneurysms and 61 patients had aneurysm with other etiologies. Hospital mortality was 19% in former group and was higher than in the latter group (6.5%). Especially, the mortality was the highest (50%) among the patients who required the intraoperative clamping of the arch vessels. Many complications related to atherosclerosis, such as atheroemboli, bleeding due to wall weakness and etc, was found among the patients died soon after the surgery. Also, age over 70 and renal or respiratory dysfunction were major risk factors to determine the operative death. On the other hand, asymptomatic ischemic heart disease and cerebrovascular disorders of three months after the onset were not significant determinant. It was concluded that the surgical indication should be contemplated with extreme care specifically among the patients with such risk factors.

Effect of the content of divinylbenzene in ion-exchange resins on the chromatographic separation of alpha-cyclodextrin and glucose.

In the chromatographic separation of alpha-cyclodextrin and glucose, the parameters relating to the adsorption equilibria and rate processes of the solutes were evaluated for the sodium forms of cation-exchange resins with contents of divinylbenzene (DVB) of 2, 4, 6 and 8%. By using these parameters, the chromatograms of the solutes were then calculated. It is concluded that the resin with 6% DVB is most suitable for the chromatographic separation of the solutes on a preparative scale.

Chemical structure of neutral sugar chains isolated from human mature milk kappa-casein.

The carbohydrate chains linked to human kappa-casein from mature milk were released by alkaline borohydride treatment as reduced oligosaccharides. The neutral oligosaccharides of lower molecular weight were fractionated and purified by gel filtration and preparative thin layer chromatographies. Seven neutral oligosaccharides (a di- (0.5%), two tetra- (30.5%), two penta- (5.4%) and two hexasaccharide alditols (10.9%] were obtained in homogeneity, and followed by methylation analysis with gas-liquid chromatography-mass spectrometry and by anomer analysis with 13C nuclear magnetic resonance. Their chemical structures were identified to be Gal beta 1----3GalNAc-ol (I), Gal beta 1----3[Gal beta 1----4GlcNAc beta 1----6]GalNAc-ol (II), Gal beta 1----3[Fuc alpha 1----4GlcNAc beta 1----6]GalNAc-ol (III), GlcNAc beta 1----3/6Gal beta 1----3[Gal beta 1----4GlcNAc beta 1----6]GalNAc-ol (IV), GlcNAc beta 1----3/6Gal beta 1----3[Fuc alpha 1----4GlcNAc beta 1----6]GalNAc-ol (V), Fuc alpha 1----4GlcNAc beta 1----3/6Gal beta 1----3[Gal beta 1----4GlcNAc beta 1----6]GalNAc-ol (VI) and Fuc alpha 1----4GlcNAc beta 1----3/6Gal beta 1----3[Fuc alpha 1----4GlcNAc beta 1----6]GalNAc-ol (VII). Five oligosaccharide alditols (III-VII) were the novel carbohydrate chains of kappa-casein from mammalian milk.

Effects of denaturation with HCl on the immunological staining of bromodeoxyuridine incorporated into DNA.

Immunochemical procedures for detection of BrdUrd incorporated into DNA require a denaturation step of DNA. Denaturation with HCl is widely used for flow cytometric analysis of the cell cycle and for histological preparations. This brief communication describes an attempt to standardize a denaturation procedure with HCl. Various denaturation conditions at 20 degrees C were examined for human promyelocytic leukemia cells (HL-60 cells) fixed in ethanol. After denaturation of DNA, the cells were stained by an indirect immunofluorescence method using a commercially available monoclonal anti-BrdUrd antibody or by propidium iodide. The relative fluorescence intensities of stained BrdUrd and double-stranded DNA were altered reciprocally by changing HCl concentration and/or denaturation time. Treatment with 4N HCl for 10-20 min at 20 degrees C allowed denaturation of more than 80% of DNA and the maximum BrdUrd-linked immunofluorescence. Under this condition, the coefficient of variation of the DNA histograms remained relatively small.

Developmental changes in steroidogenic responses of ovarian follicles of amago salmon (Oncorhynchus rhodurus) to chum salmon gonadotropin during oogenesis.

Changes in amago salmon (Oncorhynchus rhodurus) ovarian thecal and granulosa layer function in association with the production of two biologically important ovarian mediators of oocyte growth and maturation in salmonids, estradiol-17 beta and 17 alpha,20 beta-dihydroxy-4-pregnen-3-one (17 alpha,20 beta-diOHprog), were investigated using isolated follicular preparations in vitro. A distinct shift of steroidogenic responses of intact follicles from estradiol-17 beta to 17 alpha,20 beta-diOHprog in response to partially purified chum salmon gonadotropin (SGA) occurred immediately prior to oocyte maturation. Aromatase activity in granulosa layers increased during vitellogenesis and decreased rapidly prior to oocyte maturation. This decrease in aromatase activity was coincident with the decreased ability of intact follicles to produce estradiol-17 beta in response to SGA. Since testosterone production in thecal layers did not decline during this time, the reduced production of estradiol-17 beta by postvitellogenic follicles is due, in part, to decreased aromatase activity in granulosa layers. Immediately prior to oocyte maturation, intact follicles acquire an increased ability to produce 17 alpha,20 beta-diOHprog in response to SGA. Although granulosa layers first acquired the ability to convert exogenous 17 alpha-hydroxyprogesterone (17 alpha-OHprog) to 17 alpha,20 beta-diOHprog (20 beta-hydroxysteroid dehydrogenase, 20 beta-HSD, activity) in response to SGA about 2 months prior to oocyte maturation, thecal layers did not develop the ability to produce 17 alpha-OHprog in response to SGA until immediately prior to oocyte maturation. Thus, changes in thecal cell function are critical for intact follicles to acquire the ability to produce 17 alpha,20 beta-diOHprog in response to gonadotropin.

Detection of T-factor in lung cancer using magnetic resonance imaging and computed tomography.

The ability of magnetic resonance imaging (MRI) to detect T-factor was compared with that of computed tomography (CT) in 52 patients with primary lung cancer proven by surgery or autopsy, and the results were analyzed in relation to the operative and pathologic findings. In the diagnosis of tumor invasion of the heart and great vessels, MRI provided information as accurate as CT. The T1-weighted images in particular were of considerable value in separating the tumor from the mediastinal and hilar fat. Tumor extent in accompanying peripheral obstructive pneumonia or collapse was demarcated in 21% of the cases studied by CT and in 33% to 53% by MRI. On the other hand, the T2-weighted images obtained with longer echo time (TE) were useful in distinguishing the tumor from secondary changes.

Current surgical treatment and results for aneurysm of the thoracic aorta in elderly patients.

During the period from January, 1983 to December, 1986, a total of 199 patients with thoracic aortic aneurysm underwent surgical treatment in our Cardiovascular Surgical Service. During this period, criteria for surgical indications were established and general surgical principles and techniques were standardized. As a consequence, surgical results appeared to stabilize during this period. It was clearly established that the surgical result for patients over 65 years was poor with high mortality compared to the younger age group (38.6% vs 7.2%). This tendency was in sharp contrast to that of abdominal aortic aneurysm surgery where low mortality (3%) was obtained regardless of age. Other factors influencing high mortality were as follows: 1) atherosclerosis as an etiological background, 2) aneurysm situated at the aortic arch, 3) Urgency for surgery, 4) pre- and postoperative status of respiratory and renal function. Late follow-up results showed that 25% of patients died, while 66% are in fair condition.
