Establishment in continuous culture of a T-lymphoid cell line (HD-Mar) from a patient with Hodgkin's lymphoma.

A new cell line, HD-Mar, was established from a pleural effusion of a patient with Hodgkin's lymphoma. Formation of E rosettes, sensitivity to anti-T serum, elevated terminal deoxynucleotidyl transferase activity, presence of T-cell and the common ALL membrane antigens, morphology, and cytochemical staining indicate that the HD-Mar line is of thymic derivation. Absence of any immunoglobulin determinants, the lack of EBNA or any other EBV-associated antigen or function are also characteristics associated with established T-cell-derived lymphoma cell lines. Karyotype analysis indicated a tetraploid origin of the cell line.

Ataxia telangiectasia: chromosomal stability in continuous lymphoblastoid cell lines.

Chromosomal breakage in peripheral lymphocytes, cultured fibroblasts and long-term lymphoblastoid cell lines was investigated in five hitherto undescribed patients with ataxia telangiectasia (AT). Increased chromosomal instability was observed in lymphocytes and fibroblasts, and clones possessing a Dq+ marker were observed. Breakage rates were significantly higher in the fibroblasts than in the lymphocytes of AT patients or in similar tissues from patients with Bloom syndrome or Fanconi anemia. However, chromosome breakage in lymphoblastoid lines established from these five AT patients and six others did not differ from controls. These observations suggests that selection pressures, in vivo or in vitro, or both, act differently on the expression of chromosomal instability in these various cell types.

Carbohydrate of the factor VIII/von Willebrand factor in von Willebrand's disease.

We have examined the plasma Factor VIII/von Willebrand factor (FVIII/vWF) molecule from 16 patients with von Willebrand's disease, and have found no evidence of a significant decrease of carbohydrate content in 15 of these patients. FVIII/vWF was isolated by preparative counter immunoelectrophoresis directly from plasma using antibody to Factor VIII-related antigen, reduced in sodium dodecyl sulfate in the presence of urea, and electrophoresed in 5% polyacrylamide gels to separate the FVIII/vWF subunit from other proteins. Duplicate gels were stained by either the periodic acid-Schiff (PAS) reaction or by Coomassie Brilliant Blue G250. The ratio of Coomassie: PAS was determined by spectrophotometric scanning of the gels. Transferrin was used as an internal reference standard. The ratio for 23 normal individuals was 2.4+/-0.38 and the observed range was 1.8-3.8. 15 patients with von Willebrand's disease fell within this range. One patient independently reported as having decreased FVIII/vWF carbohydrate was also studied by this technique. A ratio of 6.8 was found, indicative of decreased, though not absent, carbohydrate. Cold insoluble globulin did not represent a significant contaminant in these analyses. 11 of the von Willebrand's disease patients with normal FVIII/vWF carbohydrate had abnormal crossed immunoelectrophoretic patterns characterized by absence of the less anodic forms of Factor VIII-related antigen. Four patients had normal patterns. These studies indicate that an absence or decrease of PAS reactive FVIII/vWF carbohydrate is not a consistent abnormality in von Willebrand's disease.

Instrumentation for placement of vitreous carbon endosseous implants.

The instruments required for placement, stabilization, and restoration of vitreous carbon endosseous implants are conventional in character and readily available. With the exception of extra-long burs for socket preparation, all instruments are normally available in dental offices. The surgery instruments should be rigorously sterilized prior to use, including the dental handpiece, burs, suction tip, etc. Gas sterilization techniques provide an effective means of sterilizing the surgical instruments without causing corrosion of burs and the handpiece.

[Statistical considerations on the evaluation of the Krogh-Poulsen clinical functional analysis].

361 patients, who five years previously were supplied with removable partial protheses, were submitted to Krogh-Poulson clinical function analyses. The test showed no suggestion of functional disturbances in only 12% of patients. Investigations showed that the test should be improved as to reliability, objectivity and validity before being used as a screening procedure in practice.

[Orientation studies on the stability of metal-ceramic crowns made from new materials].

Materials for metal-ceramic crowns have been developed further in the last few years and the use of this type of crown increases steadily. Numerous investigations concern themselves with binding and other properties of this material as well as with its favourable moulding capacity. The present investigation is oriented towards establishing the order of magnitude and limits of confidence of fracture resistance which has to be reckoned with when using a metal-ceramic crown in the mouth.

[Comparison of cast and metal-ceramic veneer crowns; results of a follow-up study in wearers of partial prostheses].

The results of follow-up examinations of 635 gold cast and 359 metal ceramic veneer crowns inserted primarily for caries prophylaxis and static reasons in patients with partial dentures on the average 4 1/2 years ago are reported. All crowns fulfilled their caries-preventing functions well. More than 93% were free of secondary caries, this was especially so when the crown margin was still below the gingiva. The conditions at the crown margins were studied. It was found that margin seal, type of crown as well as interdependent variables like age, periodontal condition and personal oral hygiene had a significant influence on whether the crown margin was still in the gingival sulcus. Metal ceramic veneer crowns were vestibularly exposed more frequently than cast crowns. The reason for this was found to be defective construction. The study showed further that lesions at the baked veneer crown occurred more often than had been expected so far. It is estimated that they amount to 3% with widely used ceramic crowns.
