The effect of drug schedule on mouse neuroblastoma treated with cyclophosphamide.

The potential difference in survival due to scheduling of cyclophosphamide treatment was examined in A/J mice inoculated with neuroblastoma C1-1300 cells. Treatment was started on the third day post tumor implantation. There were four groups of animals each consisting of 30 mice: Group A, a control group, was injected with 0.3 cc of normal saline for a week. Group B was injected with cyclophosphamide 50 mg/kg/day every other day for five doses. This cycle was repeated in the same fashion starting on day 17. Group C was injected with cyclophosphamide 250 mg/kg on day 3 and this was repeated on day 17. Group D was injected with cyclophosphamide 50 mg/kg/day for five consecutive days. This cycle was repeated on day 17-21. The median survival for group B was 26.2 days (the same as the control group), for group C 47.7 days (87% increased survival) and for group D 63.2 days (143% increased survival with 30% possible cures). Twenty-three percent of the mice in group B died of pneumonia and another 23% had minor evidence of lung infection. Only three mice in this group had tumor at the time of death. The study suggests that the response of murine neuroblastoma as well as the development of pneumonia in A/J mice treated with cyclophosphamide might be schedule dependent.

Differences in the distribution of urea and creatinine between red cells and plasma in normal and azotemic blood as assessed by autoanalyzer and manual chemical methods.

It has been proposed that urea and creatinine may bind to red cell constituents. In the present studies, whole blood and plasma concentrations of urea and creatinine were compared using autoanalyzer techniques (that require solutes to pass through a dialysis membrane) and manual techniques (with no dialysis membrane). Blood samples from 11 normal and 10 azotemic subjects were studied. Concentration differences between plasma water and red cell water for these solutes were significant in normals with standard manual but not with autoanalyzer methods. Accordingly, red cell water concentrations in normals were greater with manual methods than with autoanalyzer methods. In azotemics, differences between red cell and plasma water concentrations and differences in red cell water concentrations between methods were proportionately less. The findings suggest that accumulation of these solutes in the red cell in azotemia is predominantly in the freely diffusable form. Any chemical interference of red cell proteins and/or solute binding to red cell constituents yields significant discrepancies between manual and autoanalyzer methods only at lower BUN and creatinine concentrations.

Coagulation factors in renal adenocarcinoma.

Coagulation factors, including plasma fibrinogen, serum fibrinogen-fibrin degradation products, platelet counts and prothrombin times, were studied in patients with renal adenocarcinoma. Plasma fibrinogen levels were elevated and correlated with tumor stage, disease activity and therapy. Fibrinogen-fibrin degradation product levels also were elevated, although such elevations did not correlate with other parameters. Platelet count and prothrombin times were normal. Fibrinogen may be a valuable marker of disease activity in patients with renal carcinoma. In addition, since significant intratumoral fibrin deposits have been demonstrated anticoagulants or fibrinolytic agents may enhance cytotoxic therapy and should be considered in adjunctive chemotherapeutic protocols.

Development of a method of evaluating cardiac performance using interactive computer graphics.

The authors discuss an objective method of analyzing local wall motion to detect and measure the pattern of contraction of the left ventricle using computer graphics. The method's advantages include: (a) the use of modular hardward and software, which can be extended and upgraded; (b) a minimal expenditure of time and money; (c) uniformly calculated results; and (d) a graphic and legible report.

The "swinging" tube: a new concept.

A new technique converts a standard biased x-ray tube into a microfocal spot tube with a clinically useful loading capacity. If the tube is pivoted exactly through the focal spot, the film can be scanned with a 5-10 cm wide beam and cover a 17-inch field. Then the target angle can be decreased to a few degrees. The width of the focal spot can be decreased using a grid bias.

A new, simple test for renin sampling.

In 8 of 18 patients with suspected diagnosis of renovascular hypertension, the resting renin values were inconclusive. However, following tourniquet stress, the values became positive. Sixteen of 18 patients showed significant decrease in blood pressure following surgery. The procedure was well tolerated, and its routine use is recommended.

Approximation of the left ventricle by a cone of revolution using biplane measurements.

A new method based only on area measurements was developed for calculating left ventricular volume from biplane (RAO and LAO) left ventriculograms; the ventricle was approximated by a cone of revolution. Twenty-three patients with normal and 23 patients with abnormal left ventricles were studied. The Dodge method provided reference values. There was no significant difference between values for EDV calculated by either method; values for ESV calculated by the cone method were consistently higher, but the error is acceptable. The cone method provides results similar to those obtained by the corrected Dodge method, eliminating the problem of choosing and measuring the long axis.

Comparison of the left ventricular volume using the Ahlkerg and Dodge methods.

Single plane cine left ventriculography was performed in 24 patients, 12 with normal and 12 with abnormal ventricular shapes. End diastolic and systolic volume and ejection fractions were calculated by two methods (Ahlberg and Dodge). The volumes and ejection fraction from either method were not significantly different. If calculated manually, the Ahlberg method took ten times longer than the Dodge method. At present some steps are difficult but a computer can perform the measurements and calculations, and print the results for ventricular areas.

Demonstration of equine infectious anemia virus in primary leukocyte cultures by electron microscopy.

Electron microscopy was used to demonstrate the presence of viral particles in primary cultures of leukocytes taken from a horse after SC inoculation with the Wyoming strain of equine infectious anemia virus. Unlike previous studies, the exposure virus was not passaged through cell culture prior to horse inoculation. Cultures were begun approximately 1 week before and 1 week after the 1st pyrexic period after inoculation. In both samples, viral particles and cytoplasmic alterations were observed resembling those previously reported in equine infectious anemia virus and other retravirus-infected cells.
