Kinetic properties of the purified 3-hexulosephosphate synthase from Pseudomonas oleovorans.

The kinetic characteristics of the purified 3-hexulosephosphate synthase from the facultative methylotroph Pseudomonas oleovorans were investigated. It could be demonstrated that the dependence of the reaction rate on the rib(ul)ose-5-phosphate as well as the formaldehyde concentration has a complex shape with the appearence of plateau and trough regions. The shape of the curve is changed in dependence on the fixed level of the second substrate. Multiple forms of the 3-hexulosephosphate synthase were found to be responsible for the generation of the complex kinetic characteristics. By means of ion exchange chromatography it was possible to separate four active enzyme forms with different kinetic characteristics. These forms were also found to be interconvertible. This behaviour of the 3-hexulosephosphate synthase is assumed to have the main regulatory function of the enzyme.

Pentoxifylline -- a biomedical profile.

The therapeutical benefit of pentoxifylline (Trental) in vascular disorders and chronic diseases which provoke disturbances in nutritive microcirculation is reviewed. With its hemorheological activity, pentoxifylline offers a new approach to ameliorate impaired blood flow in affected microvasculature. This statement is given evidence by numerous experimental and clinical data which demonstrate efficacy in various indications, wuch as cerebrovascular insufficiency, senile organic brain syndrome, transient ischemic attacks, cerebral infarctus, ocular and otological circulatory disorders, peripheral angiopathies of arteriosclerotic, diabetic, or inflammatory origin with intermittent claudication, leg ulcers and other disturbances. In vitro and in vivo findings disclose that the efficacy is based on hemorheological effects, such as improving local hyperviscosity, hyperaggregability of red cells or platelets, erythrocyte fluidity and hypercoagulability resulting in a better oxygenation of affected tissues.

Cerebrovascular circulatory disorders: new aspects of pathophysiology and therapy.

After reviewing new aspects in the pathophysiology of the poststenotic cerebral ischemia as the cause of the various syndromes of cerebrovascular disorders, therapeutic approaches are discussed. The fatal vicious cycle of the disturbances of the nutritive blood flow within the cerebral microcirculation can be interrupted by means of pentoxifylline. By amelioration of the disturbed flow properties of blood in the microcirculation due to improvement of red cell deformability and inhibition of platelet aggregation, oxygen supply is enhanced and more glucose offered. Membrane permeability and functions of the cells are normalized by the inhibitory action on edematous changes in the brain tissue and by removal of the mechanical obstacles of microcirculation. The disturbed cerebral metabolism is stimulated by enhancement of the energy-rich phosphates in the brain cells. The combined rheological, antiedematous and metabolic effects of pentoxifylline offer a complex therapy of cerebrovascular insufficiency.

[Catamnestic study about the fertility of the man (author's transl)].

Collective investigations of fertility in childless married couples were carried out in 9 andrological health centres of the GDR. Analysis of 1937 spermatograms in comparison to fertility after 1 to 2 years shows: (1) Fertility decreases significantly at sperm density nearly 10 million per ml. (2) Motility below 60 per cent caused reduced fertility. Normal motility is more important than normal density of spermatozoa. (3) Fructose level doesn't correlate to motility or fertility of man.

[Sociocultural requirements for alcohol and tobacco education measures].

If behaviour changes are taken as a criteria for the effectiveness of alcohol and tobacco education, the results of evaluation research are not encouraging. The unefficiency of alcohol and tobacco education is considered to be caused on the one hand by a too limited concept of education applied to teaching programmes. On the other hand a number of antagonisms between educators and pupils result from not taking into consideration sociocultural facts. At the outset, these antagonisms bring about the failure of measures for alcohol and tobacco education. A number of postulates to overcome the contradictions between educational aims and the expectations of pupils are discussed.

[Malignant melanomas of fingers and toes (author's transl)].

From 1968 to 1978 more than 500 patients suffering from histologically verified malignant melanoma were seen. Only in 12 cases (appr. 2%) the tumour was situated on the tip of the fingers and toes. In such cases nowadays amputation and exarticulation of the whole extremity is no longer practiced. It is sufficient only to remove the corresponding finger or toe. Additional chemotherapy should supplement surgery.

[The excretion of hippuric acid and ascorbic acid in the urine of liver-damaged rats (author's transl].

The authors investigated the effects of the administration of thioacetamide, carbon tetrachloride and aminophenazone on the excretion of ascorbic acid and hippuric acid in adult male and female Wistar rats. After a single application of thioacetamide and aminophenazone, the ascorbic acid content in the urine showed a dose-dependent increase, whereas that in the liver had decreased. This increase in the urinary ascorbic acid might be due to a release of stored ascorbic acid from the liver cells. When thioacetamide was given for a prolonged period, the ascorbic acid content in the urine increased at the beginning; later one, at the end of three weeks, it was slightly inferior to the control value. Both single and repeated applications of thioacetamide led to a decrease in the excretion of hippuric acid in the urine, which is attributed to an impairment of the mitochondrial hippuric acid synthesis. Long-term treatment with aminophenazone resulted in an increase of ascorbic acid in the urine, which is indicative of an induction effect, whereas the ascorbic acid content in the liver remained unchanged. There was no effect on the excretion of hippuric acid. In regard to their use in the toxicological evaluation of drugs, these two metabolic effects offer no decisive advantage over current liver function tests.

[Controlled drinking as therapy target in the treatment of alcoholics].

The question of re-acquired controlled drinking as a therapy target in the treatment of alcoholics should be studied under the concrete social conditions of the GDR by specialists in treatment and prevention together with the practitioners, on the basis of an exact programme. Some proposals for the practical proceedings are submitted.

[Seroepidemiology of acute infectious hepatitis (author's transl)].

Hbs-Ag, anti-Hbs, anti-Hbc and anti-HA were determined and the concentration of IgM measured in the sera of cases of acute infectious hepatitis which occurred in the Hannover area in 1975. Although there was a high degree of contamination with hepatitis A virus among the population, acute infectious hepatitis A was rare (n = 56). The hepatitis A virus is principally transmitted by contact with infection or while traveling in southern Europe. The greatest part of infectious hepatitis is due to hepatitis virus B (n = 211). Non-A, non-B hepatitis was less frequently observed (n = 62). A high percentage of patients with serum hepatitis and non-A, non-B hepatitis gave a history of parenteral exposure to possibly infectious material.

Decrease in incidence of hepatitis A infections in Germany.

The hypothesis of a decline in the incidence of hepatitis A infections in Germany in recent decades was confirmed by determining the prevalence of hepatitis A antibody (anti-HAV) in sera collected in 1965 and in 1975 under the same conditions in North Germany. The prevalence of anti-HAV correlated with the year of birth and was independent of the time of serum sampling. The force of infection fell from 0.04 in 1945 to 0.005 in 1965 as judged from a catalytic epidemic model with a sigmoidal decrease.

The seroepidemiological pattern of acute viral hepatitis. An epidemiological study on viral hepatitis in the Hannover region.

The natural incidence of the etiologically distinct types of viral hepatitis was determined by investigating acute phase sera of symptomatic hepatitis cases occuring in the Hannover area in 1975 for the presence of hepatitis B surface antigen, antibodies to hepatitis A, hepatitis B core and surface antigens, and by measuring the IgM serum levels. Fourteen different seroepidemiologic patterns were recognized. Although there was a high prevalence of hepatitis A antibody in the population, the frequency of hepatitis A was low (n = 56) suggesting that the hepatitis A virus does not play a major role in symptomatic hepatitis in the Hannover area at present. Spread of the hepatitis A virus was mostly associated with person-to-person contact or tourist travel in southern Europe. Hepatitis B was the predominant type of hepatitis (n = 211). Hepatitis non-A, non-B was observed infrequently (n = 62). A high percentage of patients with hepatitis B and hepatitis non-A, non-B reported parenteral exposure to potentially contaminated materials. No other findings, however, suggested an infectious etiology of hepatitis non-A, non-B.
